    In recent years, the use of whole-cell biocatalysts and biocatalytic enzymes in biotechnological applications originating from the genus Pseudomonas has greatly increased. In 2014, two new species within the Pseudomonas putida group were isolated from Swiss forest soil. In this study, the high quality draft genome sequences of Pseudomonas wadenswilerensis CCOS 864T and Pseudomonas reidholzensis CCOS 865T were used in a comparative genomics approach to identify genomic features that either di ered between these two new species or to selected members of the P. putida group. The genomes of P. wadenswilerensis CCOS 864T and P. reidholzensis CCOS 865T were found to share genomic features for the degradation of aromatic compounds or the synthesis of secondary metabolites. In particular, genes encoding for biocatalytic relevant enzymes belonging to the class of oxidoreductases, proteases and isomerases were found, that could yield potential applications in biotechnology. Ecologically relevant features revealed that both species are probably playing an important role in the degradation of soil organic material, the accumulation of phosphate and biocontrol against plant pathogens

    Isolation and identification of actinomycetes strains from Switzerland and their biotechnological potential

    Actinomycetes strains isolated from different habitats in Switzerland were investigated for production of antibacterial and antitumoral compounds. Based on partial 16S rRNA gene sequences, the isolated strains were identified to genus level. Streptomyces as the largest genus of Actinobacteriawas isolated the most frequently. A screening assay using the OmniLog instrument was established to facilitate the detection of active compounds from actinomycetes. Extracts prepared from the cultivated strains able to inhibit Staphylococcus aureusand Escherichia coliwere further analysed by HPLC and MALDI-TOF MS to identify the produced antibiotics. In this study, the bioactive compound echinomycin was identified from two isolated Streptomycesstrains. Natural compounds similar to TPU-0037-C, azalomycin F4a 2-ethylpentyl ester, a derivative of bafilomycin A1, milbemycin-α8 and dihydropicromycin were detected from different isolated Streptomyces strains. Milbemycin-α8 showed cytotoxic activity against HT-29 colon cancer cells. The rare actinomycete,Micromonospora sp. Stup16_C148 produced a compound that matches with the antibiotic bottromycin A2. The draft genome sequence from Actinokineospora strain B136.1 was determined using Illumina and nanopore-based technologies. The isolated strain was not able to produce antibacterial compounds under standard cultivation conditions. The antiSMASH bioinformatics analyses of the genome from strain B136.1 identified biosynthetic gene clusters with identity values between 4% to 90% to known gene clusters encoding antibiotics. The combinations of cultivation conditions, screening assays, analytical methods and genome mining are important tools to characterize strains of actinomycetes for the identification of their potential to produce natural compounds with antimicrobial activity

    O ensino de ciências e a Educação Ambiental emancipatória com abordagem CTSA na formação de professores

     Environmental Education is essential in the formation of the human being, so it is essential that it permeates the training of teachers and becomes effective in practice with students from the beginning of primary education. This research sought to answer the questions: How are Emancipatory Environmental Education and the CTSA perspective addressed and worked on in continuing education, and do they contribute to raising awareness about environmental awareness, linked to Science Teaching? In what aspects can the methodology of the Three Pedagogical Moments (3MP), with activities from the CTSA perspective, help in the construction of an interdisciplinary proposal aimed at SEA? To do this, a sample group of 13 teachers from the initial years of EF was interviewed. Based on these data, the continuing education course for teachers was developed, consisting of 5 modules with synchronous and asynchronous activities, totaling 40 hours. With the aim of demystifying the difficulties of working on socio-environmental issues related to curricular content and Science Teaching, the training proposal was implemented with 23 participants, promoting discussions on CTSA relations, with an emphasis on EAE, and raising awareness among these professionals about his role in the emancipation of the student. Results were achieved such as raising awareness about EA, the ability to capture socio-environmental problems, restructuring classes and planning with the support of 3MP, in addition, the identification of the contributions of CTSA in the teaching process.La Educación Ambiental es fundamental en la formación del ser humano, por lo que es fundamental que permee la formación de los docentes y se haga efectiva en la práctica con los estudiantes desde el inicio de la formación primaria. Esta investigación pretendió responder a las preguntas: ¿De qué manera la Educación Ambiental Emancipadora y la perspectiva CTSA, son abordadas y trabajadas en la educación permanente, y contribuyen estas a sensibilizar sobre la conciencia ambiental, vinculada a la Enseñanza de las Ciencias? ¿En qué aspectos la metodología de los Tres Momentos Pedagógicos (3MP), con actividades de la perspectiva CTSA, puede ayudar en la construcción de una propuesta interdisciplinar dirigida a la EAE? Para ello, se entrevistó a un grupo muestra de 13 docentes de los años iniciales de EF. A partir de estos datos se elaboró ​​el curso de formación continua para docentes, compuesto por 5 módulos con actividades sincrónicas y asincrónicas, totalizando 40 horas. Con el objetivo de desmitificar las dificultades de trabajar cuestiones socioambientales relacionadas con los contenidos curriculares y la Enseñanza de las Ciencias, la propuesta de formación fue implementada con 23 participantes, promoviendo discusiones sobre las relaciones CTSA, con énfasis en EAE, y concientizando a estos profesionales sobre su rol en la emancipación del estudiante. Se lograron resultados como la sensibilización sobre EA, la capacidad de captar problemas socioambientales, la reestructuración de las clases y la planificación con el apoyo de 3MP, además, la identificación de las contribuciones de CTSA en el proceso de enseñanza.A Educação Ambiental é fundamental na formação do ser humano, assim, é essencial que perpasse a formação de professores e se efetive na práxis junto aos alunos desde o início da escolarização.     Estudo sociocultural alimentício dos alunos da UNILA

    Anais do IV Encontro de Iniciação Científica da Unila - “UNILA 5 anos: Integração em Ciência, Tecnologia e Cultura na Tríplice Fronteira” - 05 e 06 de novembro de 2015 – Sessão Ciências Biológicas e Saúde Coletiva: Alterações ambientais e comportamentais podem causar interferências no peso através de alterações da massa adiposa, ocasionadas por modificações dos hábitos alimentares. As modificações do ambiente geram um estresse no indivíduo sendo que, segundo estudos, há diferentes adaptações ao estresse. Portanto, mudanças de países ou localidades podem gerar estresse, tanto emocionais, como físicos. Assim, o objetivo deste estudo é conhecer o perfil alimentar dos alunos da UNILA, correlacionando os fatores culturais e dietéticos utilizando dados obtidos através de questionário semiestruturado. Inicialmente, os alunos foram sensibilizados por meio de cartazes, stand convidativo, convites e divulgações em redes sociais. Passou-se à etapa de coleta dos dados antropométricos, participando 23 alunos, oriundos de diferentes nacionalidades e ambos sexos, aonde foi entregue o termo de consentimento livre e esclarecido. Os dados antropométricos, peso e altura, foram obtidos através da Balança Digital G-Life e Estadiômetro Alturexata, respectivamente. A estes alunos participantes foi enviado um questionário sociocultural alimentício semiestruturado online, criado a partir do Google Drive. Os dados recolhidos foram expressos em gráficos, representando uma análise descritiva buscando relações entre a qualidade de vida e fatores culturais e dietéticos. Ao analisarmos os dados, a maioria cozinha desde que chegaram a Foz do Iguaçu e muitos encontraram diferenças quanto a comida. Para 60% dos alunos, essas diferenças interferem na qualidade de vida. Quase a mesma quantidade de alunos concordaram ou negaram possuir um hábito alimentar saudável. A maioria tem horários irregulares para alimentação. A maioria se consideram indivíduos normais, seguido por magros, sobrepeso e obesos. A mesma quantidade de indivíduos com peso normal se sente bem com seu corpo. Grande parte dos alunos recebem recurso de pais ou tutores, assim como benefício de alimentação, transporte e moradia. Contudo, 65% não recebe dinheiro de bolsas e 70% não recebem de seus países. A maioria relata apresentar cansaço, sendo que 30% se sentem muito incomodados, 35% intermediário, 22% pouco e 4% não sentem incomodados com o cansaço. Há aumento na insatisfação relacionado ao sistema de saúde. A maioria dos alunos se sente normal ou muito satisfeito com a universidade e muitos apresentam insatisfação com o restaurante. A maioria utiliza transporte público para ir à universidade. Exatamente 4 estudantes consideram ter uma qualidade de vida e um estado de saúde ruim, se comparado a um ano atrás. Porém, outros alunos acreditam ter boa qualidade de vida ou mediana, sendo que estão em bom estado de saúde e alguns estão melhores agora (maioria não relata ter modificações significativas). O estudo foi importante para compreender a situação dos discentes e colaborar na criação de programas em prol da melhoria de qualidade de vida discente da UNILA.     Global Reconstruction of Naturalized River Flows at 2.94 Million Reaches

    Spatiotemporally continuous global river discharge estimates across the full spectrum of stream orders are vital to a range of hydrologic applications, yet they remain poorly constrained. Here we present a carefully designed modeling effort (Variable Infiltration Capacity land surface model and Routing Application for Parallel computatIon of Discharge river routing model) to estimate global river discharge at very high resolutions. The precipitation forcing is from a recently published 0.1° global product that optimally merged gauge-, reanalysis-, and satellite-based data. To constrain runoff simulations, we use a set of machine learning-derived, global runoff characteristics maps (i.e., runoff at various exceedance probability percentiles) for grid-by-grid model calibration and bias correction. To support spaceborne discharge studies, the river flowlines are defined at their true geometry and location as much as possible—approximately 2.94 million vector flowlines (median length 6.8 km) and unit catchments are derived from a high-accuracy global digital elevation model at 3-arcsec resolution (~90 m), which serves as the underlying hydrography for river routing. Our 35-year daily and monthly model simulations are evaluated against over 14,000 gauges globally. Among them, 35% (64%) have a percentage bias within ±20% (±50%), and 29% (62%) have a monthly Kling-Gupta Efficiency ≥0.6 (0.2), showing data robustness at the scale the model is assessed. This reconstructed discharge record can be used as a priori information for the Surface Water and Ocean Topography satellite mission's discharge product, thus named “Global Reach-level A priori Discharge Estimates for Surface Water and Ocean Topography”. It can also be used in other hydrologic applications requiring spatially explicit estimates of global river flows
