Risks and Assets: A Qualitative Study of a Software Ecosystem in the Mining Industry
Digitalization and servitization are impacting many domains, including the
mining industry. As the equipment becomes connected and technical
infrastructure evolves, business models and risk management need to adapt. In
this paper, we present a study on how changes in asset and risk distribution
are evolving for the actors in a software ecosystem (SECO) and
system-of-systems (SoS) around a mining operation. We have performed a survey
to understand how Service Level Agreements (SLAs) -- a common mechanism for
managing risk -- are used in other domains. Furthermore, we have performed a
focus group study with companies. There is an overall trend in the mining
industry to move the investment cost (CAPEX) from the mining operator to the
vendors. Hence, the mining operator instead leases the equipment (as
operational expense, OPEX) or even acquires a service. This change in business
model impacts operation, as knowledge is moved from the mining operator to the
suppliers. Furthermore, as the infrastructure becomes more complex, this
implies that the mining operator is more and more reliant on the suppliers for
the operation and maintenance. As this change is still in an early stage, there
is no formalized risk management, e.g. through SLAs, in place. Rather, at
present, the companies in the ecosystem rely more on trust and the incentives
created by the promise of mutual future benefits of innovation activities. We
believe there is a need to better understand how to manage risk in SECO as it
is established and evolves. At the same time, in a SECO, the focus is on
cooperation and innovation, the companies do not have incentives to address
this unless there is an incident. Therefore, industry need, we believe, help in
systematically understanding risk and defining quality aspects such as
reliability and performance in the new business environment
Unusual infections: Femoral abscess due to Nocardia abscessus in a patient suffering from metastatic peripheral bronchial carcinoma and hygienic consequences
During the course of a peripheral bronchial carcinoma with pulmonary and cerebral metastases a femoral abscess developed in a 49 year-old patient after radio- and chemotherapy. A bacterial strain, which was isolated four times from a deep wound of the left thigh was tentatively identified as a member of the genus Nocardia on the basis of selected phenotypic and chemotaxonomic characteristics. The isolate was confirmed to belong to Nocardia abscessus by 16S rRNA gene sequencing. This species had previously been described as the causative agent of soft tissue infections. Although rare, nocardia soft tissue infections as complication of systemic nocardiosis are typical air born infections of immuncompromised patients and need specific attention. Infection prevention can be realized by restriction of soil and aerosol (builder's dust) together with antibiotic intervention (oral administration of trimethoprim/sulfamethoxazole)

В процессе работы был составлен технологический проект на строительство вертикальной разведочной скважины глубиной 2500 м на месторождении Томской области. Были спроектированы решения во всех основных сферах: технологической, экономической, безопасности труда и охраны окружающей среды. Спроектированные технологические решения отвечают требованиям производственной и экологической безопасности.The object of research is a Deposit of the Tomsk region. The purpose of this work is to design optimal technological solutions for well construction. In the course of work, a technological project was drawn up for the construction of a vertical exploration well with a depth of 2500 m in the field of the Tomsk region. Solutions were designed in all the main areas: technological, economic, labor safety and environmental protection. Impaired Abcb1a function and red meat in a translational colitis mouse model induces inflammation and alters microbiota composition
Inflammatory Bowel Disease (IBD) affects approximately 0.3% of the global population, with incidence rates rising dramatically worldwide. Emerging evidence points to an interplay between exposome factors such as diet and gut microbiota, host genetics, and the immune system as crucial elements in IBD development. ATP-binding cassette (ABC) transporters, including human p-glycoprotein encoded by the Abcb1 gene, influence intestinal inflammation, and their expression may interact with environmental factors such as diet and gut microbes. Our study aimed to examine the impact of protein sources on a genetic colitis mouse model. Methods Abcb1a-deficient colitis mice were fed either casein or red meat-supplemented diets to investigate potential colitis-aggravating components in red meat and their effects on host-microbiota interactions. We conducted deep label free quantitative proteomic inflammation profiling of gastrointestinal tissue (colon, ileum) and urine, and determined the overall microbiome in feces using 16S rRNA gene sequencing. Results A quantitative discovery based proteomic analysis of intestinal tissue and urine revealed associations between ileum and urine proteomes in relation to Abcb1a deficiency. The absence of Abcb1a efflux pump function and diet-induced intestinal inflammation impacted multiple systemic immune processes, including extensive neutrophil extracellular trap (NET) components observed in relation to neutrophil degranulation throughout the gastrointestinal tract. Conclusion Insights into dysregulated biological pathways in this disease model might offer translational biomarkers based on NETs and improved understanding of IBD pathogenesis in human patients. Our findings demonstrate that drug transporter deficiency induces substantial changes in the microbiota, leading to increased levels of IBD-associated strains and resulting in intestinal inflammation. Bulk cell density and Wnt/TGFbeta signalling regulate mesendodermal patterning of human pluripotent stem cells
In vitro differentiation of human pluripotent stem cells (hPSCs) recapitulates early aspects of human embryogenesis, but the underlying processes are poorly understood and controlled. Here we show that modulating the bulk cell density (BCD: cell number per culture volume) deterministically alters anteroposterior patterning of primitive streak (PS)-like priming. The BCD in conjunction with the chemical WNT pathway activator CHIR99021 results in distinct paracrine microenvironments codifying hPSCs towards definitive endoderm, precardiac or presomitic mesoderm within the first 24 h of differentiation, respectively. Global gene expression and secretome analysis reveals that TGFß superfamily members, antagonist of Nodal signalling LEFTY1 and CER1, are paracrine determinants restricting PS progression. These data result in a tangible model disclosing how hPSC-released factors deflect CHIR99021-induced lineage commitment over time. By demonstrating a decisive, functional role of the BCD, we show its utility as a method to control lineage-specific differentiation.
