    Reconstructing seasonality through stable-isotope and trace-element analyses of the Proserpine stalagmite, Han-sur-Lesse cave, Belgium : indications for climate-driven changes during the last 400 years

    Fast-growing speleothems allow for the reconstruction of palaeoclimate down to a seasonal scale. Additionally, annual lamination in some of these speleothems yields highly accurate age models for these palaeoclimate records, making these speleothems valuable archives for terrestrial climate. In this study, an annually laminated stalagmite from the Han-sur-Lesse cave (Belgium) is used to study the expression of the seasonal cycle in northwestern Europe during the Little Ice Age. More specifically, two historical 12-year-long growth periods (ca. 1593-1605 CE and 1635-1646 CE) and one modern growth period (1960-2010 CE) are analysed on a sub-annual scale for their stable-isotope ratios (delta C-13 and delta O-18) and trace-element (Mg, Sr, Ba, Zn, Y, Pb, U) contents. Seasonal variability in these proxies is confirmed with frequency analysis. Zn, Y and Pb show distinct annual peaks in all three investigated periods related to annual flushing of the soil during winter. A strong seasonal in-phase relationship between Mg, Sr and Ba in the modern growth period reflects a substantial influence of enhanced prior calcite precipitation (PCP). In particular, PCP occurs during summers when recharge of the epikarst is low. This is also evidenced by earlier observations of increased delta C-13 values during summer. In the 17th century intervals, there is a distinct antiphase relationship between Mg, Sr and Ba, suggesting that processes other than PCP, i.e. varying degrees of incongruent dissolution of dolomite, eventually related to changes in soil activity and/or land-use change are more dominant. The processes controlling seasonal variations in Mg, Sr and Ba in the speleothem appear to change between the 17th century and 1960-2010 CE. The Zn, Y, Pb, and U concentration profiles; stable-isotope ratios; and morphology of the speleothem laminae all point towards increased seasonal amplitude in cave hydrology. Higher seasonal peaks in soil-derived elements (e.g. Zn and Y) and lower concentrations of host-rock-derived elements (e.g. Mg, Sr, Ba) point towards lower residence times in the epikarst and higher flushing rates during the 17th century. These observations reflect an increase in water excess above the cave and recharge of the epikarst, due to a combination of lower summer temperatures and increased winter precipitation during the 17th century. This study indicates that the transfer function controlling Mg, Sr and Ba seasonal variability varies over time. Which process is dominant - either PCP, soil activity or dolomite dissolution - is clearly climate driven and can itself be used as a palaeoenvironment proxy

    The Schwinger-Dyson formalism is used to check the consistency of instanton model solutions for the quark propagator with recent models of confining gluon propagators. We find that the models are not consistent. A major discrepancy is the absence of a vector condensate in the instanton model that is present in the solutions with nonperturbative confining gluons.Comment: Latex file, no figure

    The invisible paradox of inflammatory bowel disease: An analysis of men's blogs.

    Inflammatory bowel disease is associated with stigmatising symptoms. Online support platforms eschew stigma, thus may appeal more to men who avoid traditional forms of health support. Using a critical realist netnographic approach and inductive thematic analysis, this study examined six blogs written by UK-based men to explore how inflammatory bowel disease was narrated. Three subthemes and one overarching theme - The (in)visible paradox of IBD - were developed. Findings suggest private aspects of inflammatory bowel disease risk experiential erasure, whereas public aspects lack control. Blogging facilitates the regaining of control, leading to important support connections and a re-imagining of the male inflammatory bowel disease body

    A multi-subunit Chlamydia vaccine inducing neutralizing antibodies and strong IFN-Îł(+) CMI responses protects against a genital infection in minipigs

    Chlamydia is the most widespread sexually transmitted bacterial disease and a prophylactic vaccine is highly needed. Ideally, this vaccine is required to induce a combined response of Th1 cell-mediated immune (CMI) response in concert with neutralizing antibodies. Using a novel Göttingen minipig animal model, we evaluated the immunogenicity and efficacy of a multi-subunit vaccine formulated in the strong Th1-inducing adjuvant CAF01. We evaluated a mixture of two fusion proteins (Hirep1 and CTH93) designed to promote either neutralizing antibodies or cell-mediated immunity, respectively. Hirep1 is a novel immunogen based on the variant domain (VD) 4 region from major outer membrane protein (MOMP) serovar (Sv) D, SvE and SvF, and CTH93 is a fusion molecule of three antigens (CT043, CT414 and MOMP). Pigs were immunized twice intramuscularly with either Hirep1+CTH93/CAF01, UV-inactivated Chlamydia trachomatis SvD bacteria (UV-SvD/CAF01) or CAF01. The Hirep1+CTH93/CAF01 vaccine induced a strong CMI response against the vaccine antigens and high titers of antibodies, particularly against the VD4 region of MOMP. Sera from Hirep1+CTH93/CAF01 immunized pigs neutralized C. trachomatis SvD and SvF infectivity in vitro. Both Hirep1+CTH93/CAF01 and UV-SvD/CAF01 vaccination protected pigs against a vaginal C. trachomatis SvD infection. In conclusion, the Hirep1+CTH93/CAF01 vaccine proved highly immunogenic and equally protective as UV-SvD/CAF01 showing promise for the development of a subunit vaccine against Chlamydia

    Molecular Gas in the Host Galaxy of a Quasar at Redshift z=6.42

    Observations of the molecular gas phase in quasar host galaxies provide fundamental constraints on galaxy evolution at the highest redshifts. Molecular gas is the material out of which stars form; it can be traced by spectral line emission of carbon--monoxide (CO). To date, CO emission has been detected in more than a dozen quasar host galaxies with redshifts (z) larger 2, the record holder being at z=4.69. At these distances the CO lines are shifted to longer wavelengths, enabling their observation with sensitive radio and millimetre interferometers. Here we present the discovery of CO emission toward the quasar SDSS J114816.64+525150.3 (hereafter J1148+5251) at a redshift of z=6.42, when the universe was only 1/16 of its present age. This is the first detection of molecular gas at the end of cosmic reionization. The presence of large amounts of molecular gas (M(H_2)=2.2e10 M_sun) in an object at this time demonstrates that heavy element enriched molecular gas can be generated rapidly in the earliest galaxies.Comment: 12 pages, 2 figures.     Gene expression profiling of early intervertebral disc degeneration reveals a down-regulation of canonical Wnt signaling and caveolin-1 expression: implications for development of regenerative strategies

    INTRODUCTION: Early degeneration of the intervertebral disc (IVD) involves a change in cellular differentiation from notochordal cells (NCs) in the nucleus pulposus (NP) to chondrocyte-like cells (CLCs). The purpose of this study was to investigate the gene expression profiles involved in this process using NP tissue from non-chondrodystrophic and chondrodystrophic dogs, a species with naturally occurring IVD degeneration. METHODS: Dual channel DNA microarrays were used to compare 1) healthy NP tissue containing only NCs (NC-rich), 2) NP tissue with a mixed population of NCs and CLCs (Mixed), and 3) NP tissue containing solely CLCs (CLC-rich) in both non-chondrodystrophic and chondrodystrophic dogs. Based on previous reports and the findings of the microarray analyses, canonical Wnt signaling was further evaluated using qPCR of relevant Wnt target genes. We hypothesized that caveolin-1, a regulator of Wnt signaling that showed significant changes in gene expression in the microarray analyses, played a significant role in early IVD degeneration. Caveolin-1 expression was investigated in IVD tissue sections and in cultured NCs. To investigate the significance of Caveolin-1 in IVD health and degeneration, the NP of 3-month-old Caveolin-1 knock-out mice was histopathologically evaluated and compared with the NP of wild-type mice of the same age. RESULTS: Early IVD degeneration involved significant changes in numerous pathways, including Wnt/β-catenin signaling. With regard to Wnt/β-catenin signaling, axin2 gene expression was significantly higher in chondrodystrophic dogs compared with non-chondrodystrophic dogs. IVD degeneration involved significant down-regulation of axin2 gene expression. IVD degeneration involved significant down-regulation in Caveolin-1 gene and protein expression. NCs showed abundant caveolin-1 expression in vivo and in vitro, whereas CLCs did not. The NP of wild-type mice was rich in viable NCs, whereas the NP of Caveolin-1 knock-out mice contained chondroid-like matrix with mainly apoptotic, small, rounded cells. CONCLUSIONS: Early IVD degeneration involves down-regulation of canonical Wnt signaling and Caveolin-1 expression, which appears to be essential to the physiology and preservation of NCs. Therefore, Caveolin-1 may be regarded an exciting target for developing strategies for IVD regeneration

    S-COL: A Copernican turn for the development of flexibly reusable collaboration scripts

    Collaboration scripts are usually implemented as parts of a particular collaborative-learning platform. Therefore, scripts of demonstrated effectiveness are hardly used with learning platforms at other sites, and replication studies are rare. The approach of a platform-independent description language for scripts that allows for easy implementation of the same script on different platforms has not succeeded yet in making the transfer of scripts feasible. We present an alternative solution that treats the problem as a special case of providing support on top of diverse Web pages: In this case, the challenge is to trigger support based on the recognition of a Web page as belonging to a specific type of functionally equivalent pages such as the search query form or the results page of a search engine. The solution suggested has been implemented by means of a tool called S-COL (Scripting for Collaborative Online Learning) and allows for the sustainable development of scripts and scaffolds that can be used with a broad variety of content and platforms. The tool’s functions are described. In order to demonstrate the feasibility and ease of script reuse with S-COL, we describe the flexible re-implementation of a collaboration script for argumentation in S-COL and its adaptation to different learning platforms. To demonstrate that a collaboration script implemented in S-COL can actually foster learning, an empirical study about the effects of a specific script for collaborative online search on learning activities is presented. The further potentials and the limitations of the S-COL approach are discussed
