    Restoring Female Agency: Wicked as a Feminist Fairy-Tale Revision

    One method of promoting gender equality that has gained popularity in recent years involves revising the fairy tales, primarily the ones compiled and revised by Charles Perrault, Hans Christian Anderson, and the Grimm brothers, to create versions in which the female characters have agency and purpose outside of furthering patriarchal gender ideals. The primary goal of the feminist fairy-tale revision is to give the female characters agency, not because they are women, but because they are functioning characters within the story. While there are many authors who attempt to create fairy-tale revisions that embody a feminist perspective, not all are successful, and some do even more harm than good in their attempts. This thesis argues that Gregory Maguire’s Wicked: The Life and Times of the Wicked Witch of the West stands as an effective feminist fairy-tale revision because its female protagonist possesses the agency to explore her gender identity, to pursue education, and to influence the world around her with violent actions. By presenting Elphaba’s life and choices without judgment or apology, Maguire creates a fairy-tale revision that encapsulates the message that so many feminist revisionists have failed to convey: the female protagonist deserves agency not merely because she is female, but because she is a fully-developed person

    Home-based counseling is increasingly an alternative mode of providing counseling services for children and families, reduces barriers to accessing traditional counseling services, and has also been shown to be effective. As such, the purpose of this qualitative study was to explore and describe the competencies needed to provide such counseling services. This study yielded five categories of competencies—necessary knowledge sets, case conceptualization, counseling behaviors, flexibility in session, and professional dispositions and behaviors. We also outline implications for counseling practice, counselor education, and public policy

    Loss of Num1-mediated cortical dynein anchoring negatively impacts respiratory growth

    Num1 is a multifunctional protein that both tethers mitochondria to the plasma membrane and anchors dynein to the cell cortex during nuclear inheritance. Previous work has examined the impact loss of Num1-based mitochondrial tethering has on dynein function in Saccharomyces cerevisiae; here, we elucidate its impact on mitochondrial function. We find that like mitochondria, Num1 is regulated by changes in metabolic state, with the protein levels and cortical distribution of Num1 differing between fermentative and respiratory growth conditions. In cells lacking Num1, we observe a reproducible respiratory growth defect, suggesting a role for Num1 in not only maintaining mitochondrial morphology, but also function. A structure–function approach revealed that, unexpectedly, Num1- mediated cortical dynein anchoring is important for normal growth under respiratory conditions. The severe respiratory growth defect in Δnum1 cells is not specifically due to the canonical functions of dynein in nuclear migration but is dependent on the presence of dynein, as deletion of DYN1 in Δnum1 cells partially rescues respiratory growth. We hypothesize that misregulated dynein present in cells that lack Num1 negatively impacts mitochondrial function resulting in defects in respiratory growth

    “Nerves” (choreographed by Kendra Fox) is an original dance composition created for five dancers that explores musicality, partnering and the artistry of each dancer. Created for an open space, the work can be performed on a stage or in a site-specific location. The piece formed as an investigation of space, shape and time, and was propelled forward to performance with the support of the Baroni Family Dance Entrepreneurship Grant. “Nerves” was first showcased at the American College Dance Association’s regional festival at Boston University in February 2018 as a culmination and synthesis of movement principles and concepts in choreographic form. Through continued investigation and exploration of physicality and relationships between the dancers, the piece has taken on its’ current form. The newest version of the work will be performed under a new title at the Spring 2018 Contemporary Dance Ensemble with PSU Choirs Collaborative Showcase

    Phase II Trial of IL-12 Plasmid Transfection and PD-1 Blockade in Immunologically Quiescent Melanoma.

    PurposeTumors with low frequencies of checkpoint positive tumor-infiltrating lymphocytes (cpTIL) have a low likelihood of response to PD-1 blockade. We conducted a prospective multicenter phase II trial of intratumoral plasmid IL-12 (tavokinogene telseplasmid; "tavo") electroporation combined with pembrolizumab in patients with advanced melanoma with low frequencies of checkpoint positive cytotoxic lymphocytes (cpCTL).Patients and methodsTavo was administered intratumorally days 1, 5, and 8 every 6 weeks while pembrolizumab (200 mg, i.v.) was administered every 3 weeks. The primary endpoint was objective response rate (ORR) by RECIST, secondary endpoints included duration of response, overall survival and progression-free survival. Toxicity was evaluated by the CTCAE v4. Extensive correlative analysis was done.ResultsThe combination of tavo and pembrolizumab was well tolerated with adverse events similar to those previously reported with pembrolizumab alone. Patients had a 41% ORR (n = 22, RECIST 1.1) with 36% complete responses. Correlative analysis showed that the combination enhanced immune infiltration and sustained the IL-12/IFNÎł feed-forward cycle, driving intratumoral cross-presenting dendritic cell subsets with increased TILs, emerging T cell receptor clones and, ultimately, systemic cellular immune responses.ConclusionsThe combination of tavo and pembrolizumab was associated with a higher than expected response rate in this poorly immunogenic population. No new or unexpected toxicities were observed. Correlative analysis showed T cell infiltration with enhanced immunity paralleling the clinical activity in low cpCTL tumors

    Genome data from a sixteenth century pig illuminate modern breed relationships

    Ancient DNA (aDNA) provides direct evidence of historical events that have modeled the genome of modern individuals. In livestock, resolving the differences between the effects of initial domestication and of subsequent modern breeding is not straight forward without aDNA data. Here, we have obtained shotgun genome sequence data from a sixteenth century pig from Northeastern Spain (Montsoriu castle), the ancient pig was obtained from an extremely well-preserved and diverse assemblage. In addition, we provide the sequence of three new modern genomes from an Iberian pig, Spanish wild boar and a Guatemalan Creole pig. Comparison with both mitochondrial and autosomal genome data shows that the ancient pig is closely related to extant Iberian pigs and to European wild boar. Although the ancient sample was clearly domestic, admixture with wild boar also occurred, according to the D-statistics. The close relationship between Iberian, European wild boar and the ancient pig confirms that Asian introgression in modern Iberian pigs has not existed or has been negligible. In contrast, the Guatemalan Creole pig clusters apart from the Iberian pig genome, likely due to introgression from international breeds
