Enzymatic engineering of the porcine genome with transposons and recombinases
<p>Abstract</p> <p>Background</p> <p>Swine is an important agricultural commodity and biomedical model. Manipulation of the pig genome provides opportunity to improve production efficiency, enhance disease resistance, and add value to swine products. Genetic engineering can also expand the utility of pigs for modeling human disease, developing clinical treatment methodologies, or donating tissues for xenotransplantation. Realizing the full potential of pig genetic engineering requires translation of the complete repertoire of genetic tools currently employed in smaller model organisms to practical use in pigs.</p> <p>Results</p> <p>Application of transposon and recombinase technologies for manipulation of the swine genome requires characterization of their activity in pig cells. We tested four transposon systems- <it>Sleeping Beauty</it>, <it>Tol2</it>, <it>piggyBac</it>, and <it>Passport </it>in cultured porcine cells. Transposons increased the efficiency of DNA integration up to 28-fold above background and provided for precise delivery of 1 to 15 transgenes per cell. Both Cre and Flp recombinase were functional in pig cells as measured by their ability to remove a positive-negative selection cassette from 16 independent clones and over 20 independent genomic locations. We also demonstrated a Cre-dependent genetic switch capable of eliminating an intervening positive-negative selection cassette and activating GFP expression from episomal and genome-resident transposons.</p> <p>Conclusion</p> <p>We have demonstrated for the first time that transposons and recombinases are capable of mobilizing DNA into and out of the porcine genome in a precise and efficient manner. This study provides the basis for developing transposon and recombinase based tools for genetic engineering of the swine genome.</p
Market Games In Finance Education
An electronic share market trading game was introduced to a large first year undergraduate finance course to allow students to experience share market trading. The response from students was positive.  We surveyed a sample of 51 of the students in this class who undertook a further one-hour trading session as part of a separate research experiment.  These students rate the game as a valuable learning experience.  They suggest that their use of the game increased their understanding of share market and the way that prices are set.  While the study results cannot be generalised to all students in the course, the results suggest that there are benefits to be gained from including an electronic share market trading game as part of the course
The outer halos of elliptical galaxies
Recent progress is summarized on the determination of the density
distributions of stars and dark matter, stellar kinematics, and stellar
population properties, in the extended, low surface brightness halo regions of
elliptical galaxies. With integral field absorption spectroscopy and with
planetary nebulae as tracers, velocity dispersion and rotation profiles have
been followed to ~4 and ~5-8 effective radii, respectively, and in M87 to the
outer edge at ~150 kpc. The results are generally consistent with the known
dichotomy of elliptical galaxy types, but some galaxies show more complex
rotation profiles in their halos and there is a higher incidence of
misalignments, indicating triaxiality. Dynamical models have shown a range of
slopes for the total mass profiles, and that the inner dark matter densities in
ellipticals are higher than in spiral galaxies, indicating earlier assembly
redshifts. Analysis of the hot X-ray emitting gas in X-ray bright ellipticals
and comparison with dynamical mass determinations indicates that non-thermal
components to the pressure may be important in the inner ~10 kpc, and that the
properties of these systems are closely related to their group environments.
First results on the outer halo stellar population properties do not yet give a
clear picture. In the halo of one bright galaxy, lower [alpha/Fe] abundances
indicate longer star formation histories pointing towards late accretion of the
halo. This is consistent with independent evidence for on-going accretion, and
suggests a connection to the observed size evolution of elliptical galaxies

GASKAP -- The Galactic ASKAP Survey
A survey of the Milky Way disk and the Magellanic System at the wavelengths
of the 21-cm atomic hydrogen (HI) line and three 18-cm lines of the OH molecule
will be carried out with the Australian Square Kilometre Array Pathfinder
telescope. The survey will study the distribution of HI emission and absorption
with unprecedented angular and velocity resolution, as well as molecular line
thermal emission, absorption, and maser lines. The area to be covered includes
the Galactic plane (|b|< 10deg) at all declinations south of delta = +40deg,
spanning longitudes 167deg through 360deg to 79deg at b=0deg, plus the entire
area of the Magellanic Stream and Clouds, a total of 13,020 square degrees. The
brightness temperature sensitivity will be very good, typically sigma_T ~ 1 K
at resolution 30arcsec and 1 km/s. The survey has a wide spectrum of scientific
goals, from studies of galaxy evolution to star formation, with particular
contributions to understanding stellar wind kinematics, the thermal phases of
the interstellar medium, the interaction between gas in the disk and halo, and
the dynamical and thermal states of gas at various positions along the

In search of the authentic nation: landscape and national identity in Canada and Switzerland
While the study of nationalism and national identity has flourished in the last decade, little attention has been devoted to the conditions under which natural environments acquire significance in definitions of nationhood. This article examines the identity-forming role of landscape depictions in two polyethnic nation-states: Canada and Switzerland. Two types of geographical national identity are identified. The first – what we call the ‘nationalisation of nature’– portrays zarticular landscapes as expressions of national authenticity. The second pattern – what we refer to as the ‘naturalisation of the nation’– rests upon a notion of geographical determinism that depicts specific landscapes as forces capable of determining national identity. The authors offer two reasons why the second pattern came to prevail in the cases under consideration: (1) the affinity between wild landscape and the Romantic ideal of pure, rugged nature, and (2) a divergence between the nationalist ideal of ethnic homogeneity and the polyethnic composition of the two societies under consideration
Matrilineal behavioral and physiological changes following the death of a non-alpha matriarch in rhesus macaque
In many species, the loss of alpha matriarchs is associated with a number of negative outcomes such as troop fission, eviction, wounding, and reduced vitality. However, whether
the dramatic consequences of their loss are due to their role as an old experienced figure or
to their alpha status remains unclear. In a retrospective study, we tested that in a semi-free
ranging colony of rhesus macaques (Macaca mulatta), the removal of a non-alpha matriarch, who had a large set of kin, led to changes in behavior and physiological stress within
her matriline. Following her removal, her matriline increased in aggression, vigilance, and
social grooming. Additionally, hierarchical stability, measured by levels of rank changes,
decreased within her matriline, and levels of intense aggression by high-ranking animals
were more frequent, as well as matrilineal wounding. Although ordinal rank was positively
associated with higher chronic hair cortisol concentrations (HCCs) in the months before the
matriarch’s removal, following her removal, only those who experienced large increases in
rank within her matriline displayed higher HCCs. Changes in matrilineal stability, aggression, behavior, and HCCs within the other two matrilines in the troop were not evident,
although caution is needed due to the small sample sizes. We conclude that the removal of
the non-alpha matriarch led to matrilineal instability, characterized by higher levels of
aggression and subsequent vigilance, rank changes, physiological stress, and grooming.
We suggest that non-alpha matriarchs with a large number of kin and social support can be

Communicating employability: the role of communicative competence for Zimbabwean highly skilled migrants in the UK
Skilled migration is an increasingly important topic for both policy and research internationally. OECD governments in particular are wrestling with tensions between their desire to use skilled migration to be on the winning side in the ‘global war for talent’ and their pandering to and/or attempts to outflank rising xenophobia. One aspect that has received relatively little attention is skilled migration from the African Commonwealth to the UK, a situation in which skilled migrants have relatively high levels of linguistic capital in the language of the host country. We focus here on the case of Zimbabwe. In spite of its popular image as a failed state, Zimbabwe has an exceptionally strong educational tradition and high levels of literacy and fluency in English. Drawing on 20 in-depth interviews of Zimbabwean highly skilled migrants, we explore the specific ways in which the communicative competences of these migrants with high formal levels of English operate in complex ways to shape their employability strategies and outcomes. We offer two main findings: first, that a dichotomy exists between their high level formal linguistic competence and their ability to communicate in less formal interactions, which challenges their employability, at least when they first move to the UK; and second, that they also lack, at least initially, the competence to narrativise their employability in ways that are culturally appropriate in England. Thus, to realise the full potential of their high levels of human capital, they need to learn how to communicate competently in a very different social and occupational milieu.
