    Animal infection studies of two recently discovered African bat paramyxoviruses, Achimota 1 and Achimota 2.

    Bats are implicated as the natural reservoirs for several highly pathogenic viruses that can infect other animal species, including man. Here, we investigate the potential for two recently discovered bat rubulaviruses, Achimota virus 1 (AchPV1) and Achimota virus 2 (AchPV2), isolated from urine collected under urban bat (Eidolon helvum) roosts in Ghana, West Africa, to infect small laboratory animals. AchPV1 and AchPV2 are classified in the family Paramyxoviridae and cluster with other bat derived zoonotic rubulaviruses (i.e. Sosuga, Menangle and Tioman viruses). To assess the susceptibility of AchPV1 and AchPV2 in animals, infection studies were conducted in ferrets, guinea pigs and mice. Seroconversion, immunohistological evidence of infection, and viral shedding were identified in ferrets and guinea pigs, but not in mice. Infection was associated with respiratory disease in ferrets. Viral genome was detected in a range of tissues from ferrets and guinea pigs, however virus isolation was only achieved from ferret tissues. The results from this study indicate Achimota viruses (AchPVs) are able to cross the species barrier. Consequently, vigilance for infection with and disease caused by these viruses in people and domesticated animals is warranted in sub-Saharan Africa and the Arabian Peninsula where the reservoir hosts are present.Royal Society Wolfson research merit award.     Terra nullius : Lacanian ethics and Australian fictions of origin

    The fiction of terra nullius, that Australia was 'no-one's land' at the time of British colonisation, was confirmed in law in 1971. At precisely this moment it had begun to fail as the ballast of white Australian identity and the fulcrum of race relations between Indigenous and non-Indigenous. Where white Australia had historically produced a gap, an empty centre from which the white Australian subject could emerge, fully formed, there was now a presence. The emergence of the Aboriginal subject into this empty space inaugurated the anxiety of white Australia that has characterised the period from the 1970s to the present. During these decades of anxiety, the story of this nation's origin-the story of 'settlement'-has retained its pivotal part in the inscription and reinscription of national meanings. Each of the three novels analysed in the thesis is a fictional account of the story of 'settlement published during the closing decades of the twentieth century. Of all the contemporary Australian fiction written about 'settlement' and the race relations conducted in its midst, these texts have been chosen because each is emblematic of a particular national fantasy, and, as is argued in this thesis, a particular orientation, to the tale it tells. The structure of each fantasy-of the frontier, of captivity, of the explorer and of the Great Australian Emptiness- offers particular opportunities for the refantasisation of that national story. The thesis asks how each novel is oriented towards the national aim of not failing to reproduce a satisfactory repetition of the story of national origin and the inevitable failure of that project. All of these questions are framed by an overarching one: what is an ethics of interpretation? The thesis offers a Lacanian response. Interpretation, for Lacan, is apophantic; it points to something, or lets it be seen. It points beyond meaning to structure; it alms to show an orientation not to a 'topic' but to a place. Lacanian psychoanalytic theory offers an ethics of interpretation that includes and accounts for that which exceeds or escapes meaning, and it does this without rendering that excess irrelevant. That something remains constitutive yet enigmatic, making interpretation, in turn, not merely the recovery and rendering of meaning but also a process which seeks to understand the function of this enigmatic structural term. Through its theory of repetition and the pleasures that repetition holds, Lacanian theory offers an approach to analysing the pleasures for the non-Indigenous Australian reader in hearing again the fictions of the nation's founding. It now seems possible for a white Australian encountering any such retelling to ask how our pleasure is taken, and to see the intransigence of our national story, its incapacity to respond to its many challengers, as a particular mode of enjoyment that is too pleasurable to renounce. A Lacanian ethics of interpretation opens up the question: what are the possibilities of re-orientating ourselves in our relation to our founding story such that we did not simply repeat what gives us pleasure

    The conformational analysis of small, flexible molcules using NMR of liquid crystalline solutions

    In this thesis a procedure for the conformational analysis of small flexible molecules by modelling experimental data is developed. The dipolar couplings between the nuclear spins in the small molecules, obtained using NMR of the molecules dissolved in liquid crystalline solution, provide the experimental data which is subsequently modelled using the Additive Potential (AP) Model to give the probability distribution over the molecular flexibility. The way in which the AP model is applied has been investigated along with the way in which the molecular flexibility is described. The use of the Rotational Isomeric State (RIS) Model has been compared with the use of a continuous potential model for the description of the rotations around certain bonds in each molecule analysed.The molecules which have been used in the development are phenyl acetate, a single rotor example, benzene-1,2-diacetate and ortho-dimethoxybenzene, which are double rotor examples, and 1,2,3-trimethoxybenzene, which has three rotors on the benzene ring.</p

    Cetacean biodiversity, spatial and temporal trends based on stranding records (1920-2016), Victoria, Australia.

    Cetacean stranding records can provide vital information on species richness and diversity through space and time. Here we collate stranding records from Victoria, Australia and assess them for temporal, spatial and demographic trends. Between 1920 and 2016, 424 stranding events involving 907 individuals were recorded across 31 Cetacea species from seven families, including five new species records for the state. Seven of these events were mass strandings, and six mother and calf strandings were recorded. Importantly, 48% of the species recorded are recognised as data deficient on the IUCN Red List. The most commonly recorded taxa were Tursiops spp. (n = 146) and Delphinus delphis (common dolphins, n = 81), with the greatest taxonomic richness (n = 24) and highest incidence of stranding events documented within the Otways mesoscale bioregion. We found no seasonal stranding patterns anywhere in the state. While our findings improve understanding of the spatial and temporal patterns of cetacean diversity within Victoria, we suggest greater effort to collect demographic data at stranding events in order to better study state-wide patterns through time. We conclude with guidelines for minimum data collection standards for future strandings to maximise information capture from each event

    Performance characteristics of five immunoassays for SARS-CoV-2: a head-to-head benchmark comparison

    Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has caused a global pandemic in 2020. Testing is crucial for mitigating public health and economic effects. Serology is considered key to population-level surveillance and potentially individual-level risk assessment. However, immunoassay performance has not been compared on large, identical sample sets. We aimed to investigate the performance of four high-throughput commercial SARS-CoV-2 antibody immunoassays and a novel 384-well ELISA.We did a head-to-head assessment of SARS-CoV-2 IgG assay (Abbott, Chicago, IL, USA), LIAISON SARS-CoV-2 S1/S2 IgG assay (DiaSorin, Saluggia, Italy), Elecsys Anti-SARS-CoV-2 assay (Roche, Basel, Switzerland), SARS-CoV-2 Total assay (Siemens, Munich, Germany), and a novel 384-well ELISA (the Oxford immunoassay). We derived sensitivity and specificity from 976 pre-pandemic blood samples (collected between Sept 4, 2014, and Oct 4, 2016) and 536 blood samples from patients with laboratory-confirmed SARS-CoV-2 infection, collected at least 20 days post symptom onset (collected between Feb 1, 2020, and May 31, 2020). Receiver operating characteristic (ROC) curves were used to assess assay thresholds.At the manufacturers' thresholds, for the Abbott assay sensitivity was 92·7% (95% CI 90·2–94·8) and specificity was 99·9% (99·4–100%); for the DiaSorin assay sensitivity was 96·2% (94·2–97·7) and specificity was 98·9% (98·0–99·4); for the Oxford immunoassay sensitivity was 99·1% (97·8–99·7) and specificity was 99·0% (98·1–99·5); for the Roche assay sensitivity was 97·2% (95·4–98·4) and specificity was 99·8% (99·3–100); and for the Siemens assay sensitivity was 98·1% (96·6–99·1) and specificity was 99·9% (99·4–100%). All assays achieved a sensitivity of at least 98% with thresholds optimised to achieve a specificity of at least 98% on samples taken 30 days or more post symptom onset.Four commercial, widely available assays and a scalable 384-well ELISA can be used for SARS-CoV-2 serological testing to achieve sensitivity and specificity of at least 98%. The Siemens assay and Oxford immunoassay achieved these metrics without further optimisation. This benchmark study in immunoassay assessment should enable refinements of testing strategies and the best use of serological testing resource to benefit individuals and population health.Public Health England and UK National Institute for Health Research

    Prospective observational cohort study on grading the severity of postoperative complications in global surgery research

    Background The Clavien–Dindo classification is perhaps the most widely used approach for reporting postoperative complications in clinical trials. This system classifies complication severity by the treatment provided. However, it is unclear whether the Clavien–Dindo system can be used internationally in studies across differing healthcare systems in high- (HICs) and low- and middle-income countries (LMICs). Methods This was a secondary analysis of the International Surgical Outcomes Study (ISOS), a prospective observational cohort study of elective surgery in adults. Data collection occurred over a 7-day period. Severity of complications was graded using Clavien–Dindo and the simpler ISOS grading (mild, moderate or severe, based on guided investigator judgement). Severity grading was compared using the intraclass correlation coefficient (ICC). Data are presented as frequencies and ICC values (with 95 per cent c.i.). The analysis was stratified by income status of the country, comparing HICs with LMICs. Results A total of 44 814 patients were recruited from 474 hospitals in 27 countries (19 HICs and 8 LMICs). Some 7508 patients (16·8 per cent) experienced at least one postoperative complication, equivalent to 11 664 complications in total. Using the ISOS classification, 5504 of 11 664 complications (47·2 per cent) were graded as mild, 4244 (36·4 per cent) as moderate and 1916 (16·4 per cent) as severe. Using Clavien–Dindo, 6781 of 11 664 complications (58·1 per cent) were graded as I or II, 1740 (14·9 per cent) as III, 2408 (20·6 per cent) as IV and 735 (6·3 per cent) as V. Agreement between classification systems was poor overall (ICC 0·41, 95 per cent c.i. 0·20 to 0·55), and in LMICs (ICC 0·23, 0·05 to 0·38) and HICs (ICC 0·46, 0·25 to 0·59). Conclusion Caution is recommended when using a treatment approach to grade complications in global surgery studies, as this may introduce bias unintentionally

    The surgical safety checklist and patient outcomes after surgery: a prospective observational cohort study, systematic review and meta-analysis

    © 2017 British Journal of Anaesthesia Background: The surgical safety checklist is widely used to improve the quality of perioperative care. However, clinicians continue to debate the clinical effectiveness of this tool. Methods: Prospective analysis of data from the International Surgical Outcomes Study (ISOS), an international observational study of elective in-patient surgery, accompanied by a systematic review and meta-analysis of published literature. The exposure was surgical safety checklist use. The primary outcome was in-hospital mortality and the secondary outcome was postoperative complications. In the ISOS cohort, a multivariable multi-level generalized linear model was used to test associations. To further contextualise these findings, we included the results from the ISOS cohort in a meta-analysis. Results are reported as odds ratios (OR) with 95% confidence intervals. Results: We included 44 814 patients from 497 hospitals in 27 countries in the ISOS analysis. There were 40 245 (89.8%) patients exposed to the checklist, whilst 7508 (16.8%) sustained ≥1 postoperative complications and 207 (0.5%) died before hospital discharge. Checklist exposure was associated with reduced mortality [odds ratio (OR) 0.49 (0.32–0.77); P\u3c0.01], but no difference in complication rates [OR 1.02 (0.88–1.19); P=0.75]. In a systematic review, we screened 3732 records and identified 11 eligible studies of 453 292 patients including the ISOS cohort. Checklist exposure was associated with both reduced postoperative mortality [OR 0.75 (0.62–0.92); P\u3c0.01; I2=87%] and reduced complication rates [OR 0.73 (0.61–0.88); P\u3c0.01; I2=89%). Conclusions: Patients exposed to a surgical safety checklist experience better postoperative outcomes, but this could simply reflect wider quality of care in hospitals where checklist use is routine
