Glucose regulates steady-state levels of PDX1 via the reciprocal actions of GSK3 and AKT kinases

The pancreatic beta cell is sensitive to even small changes in PDX1 protein levels; consequently, Pdx1 haploinsufficiency can inhibit beta cell growth and decrease insulin biosynthesis and gene expression, leading to compromised glucose-stimulated insulin secretion. Using metabolic labeling of primary islets and a cultured β cell line, we show that glucose levels modulate PDX1 protein phosphorylation at a novel C-terminal GSK3 consensus that maps to serines 268 and 272. A decrease in glucose levels triggers increased turnover of the PDX1 protein in a GSK3-dependent manner, such that PDX1 phosphomutants are refractory to the destabilizing effect of low glucose. Glucose-stimulated activation of AKT and inhibition of GSK3 decrease PDX1 phosphorylation and delay degradation. Furthermore, direct pharmacologic inhibition of AKT destabilizes, and inhibition of GSK3 increases PDX1 protein stability. These studies define a novel functional role for the PDX1 C terminus in mediating the effects of glucose and demonstrate that glucose modulates PDX1 stability via the AKT-GSK3 axis.Centro de Endocrinología Experimental y Aplicad

VALORACION ECONOMICA DE LA AFECTACION POR DESCORTEZADORES EN CUATRO COMUNIDADES DE JALAPA, NUEVA SEGOVIA, 1999-2001

    Get PDF

Human umbilical artery smooth muscle exhibits a 2-APB-sensitive capacitative contractile response evoked by vasoactive substances and expresses mRNAs for STIM, Orai and TRPC channels

After depletion of intracellular Ca2+ stores the capacitative response triggers an extracellular Ca2+ influx through store-operated channels (SOCs) which refills these stores. Our objective was to explore if human umbilical artery smooth muscle presented this response and if it was involved in the mechanism of serotonin- and histamine-induced contractions. Intracellular Ca2+ depletion by a Ca2+-free extracellular solution followed by Ca2+ readdition produced a contraction in artery rings which was inhibited by the blocker of Orai and TRPC channels 2-aminoethoxydiphenyl borate (2-APB), suggesting a capacitative response. In presence of 2-APB the magnitude of a second paired contraction by serotonin or histamine was significantly less than a first one, likely because 2-APB inhibited store refilling by capacitative Ca2+ entry. 2-APB inhibition of sarcoplasmic reticulum Ca2+ release was excluded because this blocker did not affect serotonin force development in a Ca2+-free solution. The PCR technique showed the presence of mRNAs for STIM proteins (1 and 2), for Orai proteins (1, 2 and 3) and for TRPC channels (subtypes 1, 3, 4 and 6) in the smooth muscle of the human umbilical artery. Hence, this artery presents a capacitative contractile response triggered by stimulation with physiological vasoconstrictors and expresses mRNAs for proteins and channels previously identified as SOCs.Facultad de Ciencias ExactasFacultad de Ciencias MédicasCentro de Endocrinología Experimental y AplicadaGrupo de Investigación en Fisiología Vascula

Pancreatic duodenal homeobox-1 and islet neogenesis-associated protein: A possible combined marker of activateable pancreatic cell precursors

The aim of this work was to study the possible relationship between pancreatic duodenal homeobox-1 (Pdx-1) and islet neogenesis-associated protein (INGAP) during induced islet neogenesis. Pregnant hamsters were fed with (S) and without (C) sucrose, and glycemia, insulin secretion in vitro, and pancreas immunomorphometric parameters were measured in their 7-day-old offspring. S offspring had significantly lower glycemic levels than C animals. Insulin release in response to increasing glucose contrations in the incubation medium (2-16 mM glucose) did not increase in pancreata from either C or S offspring. However, pancreata from S offspring released more insulin than those from C animals. In S offspring, β-cell mass, β-cell replication rate and islet neogenesis increased significantly, with a simultaneous decrease in β-cell apoptotic rate. INGAP- and Pdx-1-positive cell mass also increased in the islets and among acinar and duct cells. We found two subpopulations of Pdx-1 cells: INGAP-positive and INGAP-negative. Pdx-1/INGAP-positive cells did not stain with insulin, glucagon, somatostatin, pancreatic polypeptide, or neurogenin 3 antibodies. The increment of Pdx-1/INGAP-positive cells represented the major contribution to the Pdx-1 cell mass increase. Such increments varied among pancreas subsectors: ductal>insular>extrainsular. Our results suggested that INGAP participates in the regulation of islet neogenesis, and Pdx-1/INGAP-positive cells represent a new stem cell subpopulation at an early stage of development, highly activateable in neogenesis.Centro de Endocrinología Experimental y AplicadaFacultad de Ciencias Médica

Palabras clave: Gastropoda, riqueza, distribución geográfica, tallas, Guerrero Abstract.- The aim of this research was the description of the taxocoenosis of Prosobranchia and Pulmonata subclasses (Gastropoda) associated with the rocky intertidal zone of the Marine Priority Region 32, Guerrero, Mexico. Based on known species richness, we examine the composition of the community based on the representation of families, assess abundance, the geographic distribution of species, estimate descriptive statistics of size, and estimate diversity. Sampling was conducted at 7 sites, the sampling unit was 1 m-2 and the sampling area was 10 m-2. One hundred and four species of subclass Prosobranchia and 4 species of subclass Pulmonata were identified. Columbellidae and Muricidae families showed higher species richness, meanwhile family Muricidae was the most abundant. Acanthais triangularis was the most representative species of the study area, and Macrocypraea cervinetta showed the greatest size. Twenty three species were found with broad distribution, 27 taxa with frequent distribution and 54 taxa with restricted distribution. Twenty eight species were represented by a single organism and 6 species with 2 organisms, these species were considered rare and were found in sites whose characteristic wave exposure is low to medium. Species richness was high and corresponds to that expected in the tropics. Key words: Gastropoda, species richness, geographic distribution, size, Guerrer
