    Single low-dose cyclophosphamide combined with interleukin-12 gene therapy is superior to a metronomic schedule in inducing immunity against colorectal carcinoma in mice

    The use of conventional cytotoxic agents at metronomic schedules, alone or in combination with targeted agents or immunotherapy, is being explored as a promising anticancer strategy. We previously reported a potent antitumor effect of a single low-dose cyclophosphamide and interleukin-12 (IL-12) gene therapy against advanced gastrointestinal carcinoma, in mice. Here, we assessed whether the delivery of IL-12 by gene therapy together with metronomic cyclophosphamide exerts antitumor effects in a murine model of colorectal carcinoma. This combination therapy was able, at least in part, to reverse immunosuppression, by decreasing the number of regulatory T cells (Tregs) as well as of splenic myeloid-derived suppressor cells (MDSC s). However, metronomic cyclophosphamide plus IL-12 gene therapy failed to increase the number of tumor-infiltrating T lymphocytes and, more importantly, to induce a specific antitumor immune response. With respect to this, cyclophosphamide at a single low dose displayed a superior anticancer profile than the same drug given at a metronomic schedule.     Adipose tissue from metabolic syndrome mice induces an aberrant miRNA signature highly relevant in prostate cancer development

    Prostate cancer (PCa) remains an important public health concern in Western countries. Metabolic syndrome (MeS) is a cluster of pathophysiological disorders with increasing prevalence in the general population that is a risk factor for PCa. Several studies have determined that a crosstalk between white adipose tissue (WAT) and solid tumors favors cancer aggressiveness. In this work, our main goal was to investigate the interaction between WAT and PCa cells through microRNAs (miRNAs), in MeS mice. We developed a MeS-like disease model using C57BL/6J mice chronically fed with high-fat diet (HFD) that were inoculated with TRAMP-C1 PCa cells. A group of five miRNAs (mmu-miR-221-3p, 27a-3p, 34a-5p, 138-5p, and 146a-5p) were increased in gonadal WAT (gWAT), tumors, and plasma of MeS mice compared to control animals. Three of these five miRNAs were detected in the media from gWAT and TRAMP-C1 cell cocultures, and significantly increased in MeS context. More importantly, hsa-miR-221-3p, 146a-5p, and 27a-3p were increased in bloodstream of PCa patients compared to healthy donors. Using miRNA microarrays, we found that 121 miRNAs were differentially released to the coculture media between HFD-gWAT and tumor cells compared to control diet-gWAT and tumor cells. Target genes for the 66 most deregulated miRNAs were involved in common pathways, mainly related to fatty acid metabolism, ER protein processing, amino acid degradation, PI3K AKT signaling, and PCa. Our findings show for the first time a signature of five miRNAs as important players involved in the interaction between WAT and PCa in MeS mice. Further research will be necessary to track these miRNAs in the interaction between these tissues as well as their role in PCa patients with MeS.Fil: Massillo, Cintia Lorena.     A population-based study on myelodysplastic syndromes in the Lazio Region (Italy), medical miscoding and 11-year mortality follow-up. The Gruppo Romano-Laziale Mielodisplasie experience of retrospective multicentric registry

    Data on Myelodysplastic Syndromes (MDS) are difficult to collect by cancer registries because of the lack of reporting and the use of different classifications of the disease. In the Lazio Region, data from patients with a confirmed diagnosis of MDS, treated by a hematology center, have been collected since 2002 by the Gruppo Romano-Laziale Mielodisplasie (GROM-L) registry, the second MDS registry existing in Italy. This study aimed at evaluating MDS medical miscoding during hospitalizations, and patients' survival. For these purposes, we selected 644 MDS patients enrolled in the GROM-L registry. This cohort was linked with two regional health information systems: the Hospital Information System (HIS) and the Mortality Information System (MIS) in the 2002-2012 period. Of the 442 patients who were hospitalized at least once during the study period, 92% had up to 12 hospitalizations. 28.5% of patients had no hospitalization episodes scored like MDS, code 238.7 of the International Classification of Disease, Ninth Revision, Clinical Modification (ICD-9-CM). The rate of death during a median follow-up of 46 months (range 0.9-130) was 45.5%. Acute myeloid leukemia (AML) was the first cause of mortality, interestingly a relevant portion of deaths is due to cerebro-cardiovascular events and second tumors. This study highlights that MDS diagnosis and treatment, which require considerable healthcare resources, tend to be under-documented in the HIS archive. Thus we need to improve the HIS to better identify information on MDS hospitalizations and outcome. Moreover, we underline the importance of comorbidity in MDS patients' survival

    Acceleration of TAA-Induced liver fibrosis by stress exposure is associated with upregulation of nerve growth factor and glycopattern deviations

    Liver fibrosis results from many chronic injuries and may often progress to cirrhosis andhepatocellular carcinoma (HCC). In fact, up to 90% of HCC arise in a cirrhotic liver. Conversely,stress is implicated in liver damage, worsening disease outcome. Hence, stress could play a role indisrupting liver homeostasis, a concept that has not been fully explored. Here, in a murine modelof TAA-induced liver fibrosis we identified nerve growth factor (NGF) to be a crucial regulatorof the stress-induced fibrogenesis signaling pathway as it activates its receptor p75 neurotrophinreceptor (p75NTR), increasing liver damage. Additionally, blocking the NGF decreased liver fibrosiswhereas treatment with recombinant NGF accelerated the fibrotic process to a similar extent thanstress challenge. We further show that the fibrogenesis induced by stress is characterized by specificchanges in the hepatoglycocode (increased β1,6GlcNAc-branched complex N-glycans and decreasedcore 1 O-glycans expression) which are also observed in patients with advanced fibrosis compared topatients with a low level of fibrosis.     CTBP1/CYP19A1/Estradiol axis together with adipose tissue impacts over prostate cancer growth associated to metabolic syndrome

    Metabolic syndrome (MeS) increases prostate cancer (PCa) risk and aggressiveness. Cterminal binding protein 1 (CTBP1) is a transcriptional co-repressor of tumor suppressor genes that is activated by low NAD+/NADH ratio. Previously, our group established a MeS and PCa mice model that identified CTBP1 as a novel link associating both diseases. We found that CTBP1 controls the transcription of aromatase (CYP19A1), a key enzyme that converts androgens to estrogens. The aim of this work was to investigate the mechanism that explains CTBP1 as a link between MeS and PCa based on CYP19A1 and estrogen synthesis regulation using PCa cell lines, MeS/PCa mice and adipose co-culture systems. We found that CTBP1 and E1A binding protein p300 (EP300) bind to CYP19A1 promoter and downregulate its expression in PC3 cells. Estradiol, through the estrogen receptor beta, released CTBP1 from CYP19A1 promoter triggering its transcription and modulating PCa cell proliferation. We generated NSG and C57BL/6J MeS mice by chronically feeding animals with high fat diet. In the NSG model, CTBP1 depleted PCa xenografts showed an increase in the CYP19A1 expression with the subsequent increment in intratumor estradiol concentrations. Additionally, in C57BL/6J mice, MeS induces hypertrophy, hyperplasia and inflammation of the white adipose tissue, which leads to a proinflammatory phenotype and increases serum estradiol concentration. Thus, MeS increased PCa growth and Ctbp1, Fabp4 and IL-6 expression levels. These results describe, for the first time, a novel CTBP1/CYP19A1/Estradiol axis that explains, in part, the mechanism for prostate tumor growth increase by MeS.Fil: Massillo, Cintia Lorena.     Reversal of gastrointestinal carcinoma-induced immunosuppression and induction of antitumoural immunity by a combination of cyclophosphamide and gene transfer of IL-12

    Immunotherapy-based strategies for gastrointestinal carcinomas (GIC) have been exploited so far, but these approaches have to face strong mechanisms of immune escape induced by tumours. We previously demonstrated that sub-therapeutic doses of an adenovirus expressing IL-12 genes (AdIL-12) mediated a potent antitumour effect against subcutaneous (s.c.) colorectal carcinomas (CRC) in mice pre-treated with low doses of cyclophosphamide (Cy). In our study we used this combination to assess its impact on the immunosuppressive microenvironment. In s.c. CRC model we demonstrated that non-responder mice failed to decrease Tregs in tumour, spleen and peripheral blood. Reconstitution of Tregs into tumour-bearing mice treated with combined therapy abolished the antitumoural effect. In addition, Cy + AdIL-12 modified Tregs functionality by inhibiting the in vitro secretion of IL-10 and TGF-β and their ability to inhibit dendritic cells activation. Combined treatment decreased the number of myeloid-derived suppressor cells (MDSCs) in comparison to non-treated mice and, interestingly, administration of Tregs restored splenic MDSCs population. Furthermore, combined therapy potently generated specific cytotoxic IFN-γ-secreting CD4+ T cells able to eradicate established CRC tumours after adoptive transfer. Finally, we evaluated the combination on disseminated CRC and pancreatic carcinoma (PC). Cy + AdIL-12 were able to eradicate liver metastatic CRC (47%) and PC tumour nodules (40%) and to prolong animal survival.     La modulación de la expresión de ácido hialurónico reduce la resistencia a la quimioterapia de células iniciadoras de tumor en cáncer de pulmón experimental

    La mayoría de los pacientes con cáncer de pulmón de células no pequeñas (NSCLC) progresan o recaen luego del tratamiento con taxanos-platinos. En el microambiente tumoral (TME) las cancer stem cells (CSCs), que expresan CD133, están involucradas en la recurrencia. El ácido hialurónico (HA) del TME regula, en parte, la función de las CSCs. Analizamos HA en el TME y si su modulación con la cumarina 4-Methylumbelliferona (4Mu) modifica las propiedades de las CSCs. Utilizamos líneas murinas (Lewis Lung Carcinoma; LLC) y humanas (A549) para evaluar viabilidad luego de la exposición al paclitaxel, pemetrexed o cisplatino, o combinados con 4Mu. Analizamos sintasas HAS y hialuronidasas HYAL y factores de transcripción de CSCs a partir de datos de pacientes (TCGA) y por qPCR. Aislamos células CD133+ y estudiamos la expresión de HA y la capacidad clonogénica y tumorigénica. Observamos que los tumores de LLC producen HA, generado en parte por estas células. El 8.53 ± 0.35% de LLC son CD133+, y expresan más HA y CD44 en comparación a las CD133- (p<0,05). El análisis de TCGA mostró que HAS3 se correlaciona positivamente con los niveles de KLF4 y SOX2, pero HYAL2 se correlaciona inversamente con la expresión de SOX2. La viabilidad de las CD133+ tratadas con 4Mu+quimioterapia (Qx) y la expresión génica de HAS, CD44, CD47 y SOX2 disminuyeron significativamente (p<0.05). A partir de estos hallazgos, sugerimos que la inhibición de HA podría aumentar la susceptibilidad de las CSCs a la Qx, mejorando su eficacia y/o previniendo la recurrencia del tumor.Many patients with non-small cell lung cancer (NSCLC) will progress or relapse after treatment with platinum-taxanes. Tumors are composed by a bulk of cells including cancer stem cells (CSCs), which express CD133, and are involved in recurrence. Hyaluronic acid (HA) a glycosaminoglycan of tumor microenvironment (TME) regulates, in part, the function of the CSCs. The aim of this work was to analyze HA in TME and if its modulation using coumarin 4-Methylumbelliferone (4Mu) modifies the properties of the CSCs. We used murine (Lewis Lung Carcinoma; LLC) and human (A549) lines to evaluate cell viability after exposure to paclitaxel, pemetrexed or cisplatin, alone or combined with 4Mu. Expression of HA synthases (HAS), hyaluronidase HYAL and CSC transcription factors were analyzed from patient data (TCGA) and by qPCR. We isolated CD133+ cells and studied the expression of HA before and after the treatments. We evaluated clonogenic and tumorigenic capacity. We observed that LLC tumors produce HA, generated in part by these cells. The 8.53 ± 0.35% of LLC are CD133+, and express more HA and CD44 compared to CD133- (p<0.05). The TCGA analysis showed that HAS3 is positively correlated with the levels of KLF4 and SOX2, while HYAL2 is inversely correlated with the expression of SOX2. The viability of CD133+ treated with 4Mu+chemotherapy (Qx) and the gene expression of HAS, CD44, CD47 and SOX2 decreased significantly (p<0.05).     Recently identified mutations in the genes encoding the neuronal nicotinic ACh receptor (nAChR) subunits in patients affected by sporadic amyotrophic lateral sclerosis (sALS) may represent a factor which enhances disease susceptibility, in particular in association with ambient causes such as cigarette smoking. In this work, we characterize the functional properties of nAChRs containing the β4R349C subunit, the mutation most frequently encountered in sALS patients. The mutation was coexpressed with wild-type α3 or α4 subunits or with mutant α4R487Q subunit, which has been detected in one patient together with β4R349C mutation. None of the functional parameters examined showed differences between α4β4 and α4R487Qβ4 nAChRs. By contrast, β4R349C mutation, independent of the companion α subunit, caused the reduction in potency of both ACh and nicotine, decreased the density of whole-cell current evoked by maximal transmitter concentrations, and altered the kinetics of ACh-evoked whole-cell currents. These data confirm that sALS-associated mutations in nicotinic subunits may markedly perturb cholinergic transmission in individuals bearing the mutations

    Cancer-Associated Fibroblasts in the Breast Tumor Microenvironment

    Years of investigation have shed light on a theory in which breast tumor epithelial cells are under the effect of the stromal microenvironment. This review aims to discuss recent findings concerning the phenotypic and functional characteristics of cancer associated fibroblasts (CAFs) and their involvement in tumor evolution, as well as their potential implications for anti-cancer therapy. In this manuscript, we reviewed that CAFs play a fundamental role in initiation, growth, invasion, and metastasis of breast cancer, and also serve as biomarkers in the clinical diagnosis, therapy, and prognosis of this disease.Fil: Giorello, Maria Belen.
