    Ultrastructure and localization of Neorickettsia in adult digenean trematodes provides novel insights into helminth-endobacteria interaction

    Abstract Background Neorickettsia are a group of intracellular α proteobacteria transmitted by digeneans (Platyhelminthes, Trematoda). These endobacteria can also infect vertebrate hosts of the helminths and cause serious diseases in animals and humans. Neorickettsia have been isolated from infected animals and maintained in cell cultures, and their morphology in mammalian cells has been described. However, limited information is available on the morphology and localization of Neorickettsia in the trematode host. Methods We used a Neorickettsia-infected strain of the model trematode Plagiorchis elegans to infect Syrian Golden hamsters to produce adult worms. Ultrastructure of Neorickettsia was assessed by transmission electron microscopy of high pressure freezing/freeze substitution fixed specimens. A Neorickettsia surface protein from P. elegans (PeNsp-3) was cloned and antibodies against the recombinant protein were used to localize Neorickettsia by immunohistochemistry. Results Ultrastructural analysis revealed moderate numbers of pleomorphic endobacteria with a median size of 600 × 400 nm and characteristic double membranes in various tissue types. Endobacteria showed tubular membrane invaginations and secretion of polymorphic vesicles. Endobacteria were unevenly localized as single cells, or less frequently as small morula-like clusters in the ovary, Mehlis’ gland, vitelline follicles, uterus, intrauterine eggs, testis, cirrus-sac, tegument, intestine and the oral and ventral sucker. Examination of hamster small intestine infected with P. elegans showed many endobacteria at the host-parasite interface such as the oral and ventral sucker, the tegument and the excretory pore. Conclusions We conclude that adult P. elegans trematodes carry Neorickettsia endobacteria in varying numbers in many tissue types that support vertical transmission, trematode to trematode transmission via seminal fluid, and possibly horizontal transmission from trematodes to vertebrate hosts. These means appear to be novel mechanisms of pathogen transmission by endoparasitic worms

    Effect of random interactions in spin baths on decoherence

    We study the decoherence of a central spin 1/2 induced by a spin bath with intrabath interactions. Since we are interested in the cumulative effect of interaction and disorder, we study baths comprising Ising spins with random ferro- and antiferromagnetic interactions between the spins. Using the resolvent operator method which goes beyond the standard Born-Markov master equation approach, we show that, in the weak coupling regime, the decoherence of the central spin at all times is entirely determined by the local-field distribution or equivalently, the dynamical structure factor of the Ising bath. We present analytic results for the Ising spin chain bath at arbitrary temperature for different distributions of the intrabath interaction strengths. We find clear evidence of non-Markovian behavior in the low temperature regime. We also consider baths described by Ising models on higher-dimensional lattices. We find that interactions lead to a significant reduction of the decoherence. An important feature of interacting spinbaths is the saturation of the asymptotic Markovian decay rate at high temperatures, as opposed to the conventional Ohmic boson bath.Comment: 13 page

    Condensate fragmentation as a sensitive measure of the quantum many-body behavior of bosons with long-range interactions

    The occupation of more than one single-particle state and hence the emergence of fragmentation is a many-body phenomenon universal to systems of spatially confined interacting bosons. In the present study, we investigate the effect of the range of the interparticle interactions on the fragmentation degree of one- and two-dimensional systems. We solve the full many-body Schr\"odinger equation of the system using the recursive implementation of the multiconfigurational time-dependent Hartree for bosons method, R-MCTDHB. The dependence of the degree of fragmentation on dimensionality, particle number, areal or line density and interaction strength is assessed. It is found that for contact interactions, the fragmentation is essentially density independent in two dimensions. However, fragmentation increasingly depends on density the more long-ranged the interactions become. The degree of fragmentation is increasing, keeping the particle number NN fixed, when the density is decreasing as expected in one spatial dimension. We demonstrate that this remains, nontrivially, true also for long-range interactions in two spatial dimensions. We, finally, find that within our fully self-consistent approach, the fragmentation degree, to a good approximation, decreases universally as N1/2N^{-1/2} when only NN is varied.Comment: 8 pages of RevTex4-1, 5 figure

    Coccolithophore fluxes in the open tropical North Atlantic: influence of thermocline depth, Amazon water, and Saharan dust

    Coccolithophores are calcifying phytoplankton and major contributors to both the organic and inorganic oceanic carbon pumps. Their export fluxes, species composition, and seasonal patterns were determined in two sediment trap moorings (M4 at 12 degrees N, 49 degrees W and M2 at 14 degrees N, 37 degrees W) collecting settling particles synchronously from October 2012 to November 2013 at 1200 m of water depth in the open equatorial North Atlantic. The two trap locations showed a similar seasonal pattern in total coccolith export fluxes and a predominantly tropical coccolithophore settling assemblage. Species fluxes were dominated throughout the year by lower photic zone (LPZ) taxa (Florisphaera profunda, Gladiolithus flabellatus) but also included upper photic zone (UPZ) taxa (Umbellosphaera spp., Rhabdosphaera spp., Umbilicosphaera spp., Helicosphaera spp.). The LPZ flora was most abundant during fall 2012, whereas the UPZ flora was more important during summer. In spite of these similarities, the western part of the study area produced persistently higher fluxes, averaging 241 x 10(7) +/- 76 x 10(7) coccoliths m(-2) d(-1) at station M4 compared to only 66 x 10(7) +/- 31 x 10(7) coccoliths m(-2) d(-1) at station M2. Higher fluxes at M4 were mainly produced by the LPZ species, favoured by the westward deepening of the thermocline and nutricline. Still, most UPZ species also contributed to higher fluxes, reflecting enhanced productivity in the western equatorial North Atlantic. Such was the case of two marked flux peaks of the more opportunistic species Gephyrocapsa muellerae and Emiliania huxleyi in January and April 2013 at M4, indicating a fast response to the nutrient enrichment of the UPZ, probably by wind-forced mixing. Later, increased fluxes of G. oceanica and E. huxleyi in October-November 2013 coincided with the occurrence of Amazon-River-affected surface waters. Since the spring and fall events of 2013 were also accompanied by two dust flux peaks, we propose a scenario in which atmospheric dust also provided fertilizing nutrients to this area. Enhanced surface buoyancy associated with the river plume indicates that the Amazon acted not only as a nutrient source, but also as a surface density retainer for nutrients supplied from the atmosphere. Nevertheless, lower total coccolith fluxes during these events compared to the maxima recorded in November 2012 and July 2013 indicate that transient productivity by opportunistic species was less important than "background" tropical productivity in the equatorial North Atlantic. This study illustrates how two apparently similar sites in the tropical open ocean actually differ greatly in ecological and oceanographic terms. Therefore, for the investigation and optimisation of integrated circuits it is essential to develop suitable models and simulation surroundings which allow for memory and timeefficient calculation of the behaviour. By means of the dynamic reconstruction theory and the radial basis functions nets the so-called black box models are provided. The description of black box models is derived from the input and output behaviour or so-called time series of a dynamic system. Concerning the time series, the black box model adapts its parameters via the extended Kalman filter.     Improved Conformal Mapping of the Borel Plane

    The conformal mapping of the Borel plane can be utilized for the analytic continuation of the Borel transform to the entire positive real semi-axis and is thus helpful in the resummation of divergent perturbation series in quantum field theory. We observe that the rate of convergence can be improved by the application of Pad\'{e} approximants to the Borel transform expressed as a function of the conformal variable, i.e. by a combination of the analytic continuation via conformal mapping and a subsequent numerical approximation by rational approximants.
