    Differential expression of microRNAs in mouse pain models

    <p>Abstract</p> <p>Background</p> <p>MicroRNAs (miRNAs) are short non-coding RNAs that inhibit translation of target genes by binding to their mRNAs. The expression of numerous brain-specific miRNAs with a high degree of temporal and spatial specificity suggests that miRNAs play an important role in gene regulation in health and disease. Here we investigate the time course gene expression profile of miR-1, -16, and -206 in mouse dorsal root ganglion (DRG), and spinal cord dorsal horn under inflammatory and neuropathic pain conditions as well as following acute noxious stimulation.</p> <p>Results</p> <p>Quantitative real-time polymerase chain reaction analyses showed that the mature form of miR-1, -16 and -206, is expressed in DRG and the dorsal horn of the spinal cord. Moreover, CFA-induced inflammation significantly reduced miRs-1 and -16 expression in DRG whereas miR-206 was downregulated in a time dependent manner. Conversely, in the spinal dorsal horn all three miRNAs monitored were upregulated. After sciatic nerve partial ligation, miR-1 and -206 were downregulated in DRG with no change in the spinal dorsal horn. On the other hand, axotomy increases the relative expression of miR-1, -16, and 206 in a time-dependent fashion while in the dorsal horn there was a significant downregulation of miR-1. Acute noxious stimulation with capsaicin also increased the expression of miR-1 and -16 in DRG cells but, on the other hand, in the spinal dorsal horn only a high dose of capsaicin was able to downregulate miR-206 expression.</p> <p>Conclusions</p> <p>Our results indicate that miRNAs may participate in the regulatory mechanisms of genes associated with the pathophysiology of chronic pain as well as the nociceptive processing following acute noxious stimulation. We found substantial evidence that miRNAs are differentially regulated in DRG and the dorsal horn of the spinal cord under different pain states. Therefore, miRNA expression in the nociceptive system shows not only temporal and spatial specificity but is also stimulus-dependent.</p

    SOLIS PRAIA HOTEL ITAPEMA: os desafios na contratação de mão de obra

    O Solis Praia Hotel Itapema, localizado na região central da cidade de Itapema, conta com aproximadamente 6.000 m2 de área construída, 85 apartamentos e total estrutura física e administrativa, para um estabelecimento de categoria superior. Situado no litoral Norte Catarinense, sofre com o problema da falta de mão de obra em determinadas épocas do ano, por estar numa região turística de alta sazonalidade. Junto com esta temática, outros fatores como a alta rotatividade no setor, a concorrência com outros estabelecimentos de serviços e a falta de capacitação também contribuem para agravar esta situação. A contratação temporária de um grande número de funcionários torna difícil a manutenção de um padrão de qualidade no atendimento ao hóspede, tendo em vista esta falta capacitação da mão de obra e o pouco tempo para ambientação e treinamento destas pessoas. Contudo o objetivo principal deste estudo é analisar estes fatores, verificando como cada um deles interfere no produto Solis, permitindo ao hotel atrair pessoas comprometidas, incentivar aqueles que lá estão e, assim, garantir a sua permanência neste mercado competitivo

    Water deficit tolerance of bean cultivars

    O objetivo deste trabalho foi avaliar a resposta das cultivares de feijão (Phaseolus vulgaris) Triunfo, Garapiá e BRS-FC104 ao déficit hídrico representado pela fração de água transpirável no solo (FATS). O delineamento experimental foi inteiramente casualizado, em arranjo bifatorial 3×2 (cultivares × irrigação e sem irrigação). Avaliaram-se a transpiração, o crescimento e a produtividade na safra e na safrinha. Na safra, com baixa demanda atmosférica do ar (inferior a 15 hPa em 55% dos dias), 'Garapiá' apresentou fechamento precoce dos estômatos com FATS crítica (FATSc) de 0,36, mas teve a mesma produtividade que 'Triunfo', cujos estômatos fecharam tardiamente (FATSc = 0,23). Na safrinha, com alta demanda atmosférica do ar (superior a 15 hPa em 83,3% dos dias), 'Garapiá' apresentou fechamento estomático precoce (FATSc = 0,17) e maior produtividade (392,2 kg ha-1 a mais que 'Triunfo'). Na safra e na safrinha, 'Garapiá' apresentou tolerância ao déficit hídrico no período reprodutivo. O déficit hídrico resultou em menores valores de estatura, massa seca da parte aérea, massa seca da raiz e área foliar. Tanto 'Garapiá' como 'Triunfo' apresentaram alta produtividade na safra, mas apenas Garapiá na safrinha. 'Garapiá' apresenta tolerância ao déficit hídrico, com controle estomático eficiente e com altos crescimento e produtividade.The objective of this work was to evaluate the response of the Garapiá, Triunfo, and BRS-FC104 bean (Phaseolus vulgaris) cultivars to the water deficit represented by the fraction of transpirable soil water (FTSW). The experimental design was completely randomized in a 3×2 bifactorial arrangement (cultivars × irrigation and no irrigation). Transpiration, growth, and yield were evaluated in the crop season and off-season. In the crop season, with a low atmospheric air demand (below 15 hPa in 55% of the days), 'Garapiá' presented early stomatal closure with the critical FTSW (FTSWc) of 0.36, but had the same yield as 'Triunfo', whose stomata closed late (FTSWc = 0.23). In the off-season, with a high atmospheric air demand (greater than 15 hPa in 83.3% of the days), 'Garapiá' presented an early stomatal closure (FTSWc = 0.17) and a higher yield (392.2 kg kg ha-1 more than 'Triunfo'). In the crop season and off-season, 'Garapiá' showed tolerance  to water deficit in the reproductive period. Water deficit resulted in  lower values of height, shoot dry mass, root dry mass, and leaf area. Both 'Garapiá' and 'Triunfo' showed a high yield in the crop season, but only 'Garapiá' in the off-season.     Centrosome amplification in chondrosarcomas: A primary cell culture and cryopreserved tumor sample study

    The genetics background underlying the aggressiveness of chondrosarcoma (CS) is poorly understood. One possible cause of malignant transformation is chromosomal instability, which involves an error in mitotic segregation due to numerical and/or functional abnormalities of centrosomes. The present study aimed to evaluate centrosome amplification in cryopreserved samples of tumor tissue from patients with CS. An analysis was performed on 3 primary cultures of tumors from patients who underwent surgery between January 2012 and December 2012 at the Department of Orthopedics at the Barretos Cancer Hospital (Barretos, Brazil). Additionally, cryopreserved tumor specimens were analyzed from 10 patients. The data were assessed using immunocytochemistry and immunohistochemistry staining techniques with monoclonal antibody anti-gamma-tubulin. A total of 4 samples of CS cultured cells were obtained from 3 patients. A recurrence of a histological grade III tumor was detected in a female patient with Ollier's syndrome. The other 2 cases were grade I and III. The incidence of centrosome amplification in the primary cultures ranged from 15-64% of the cells. Whereas control cultured fibroblasts showed baseline levels of 4% amplified cells. For the cryopreserved specimens, two independent observers analyzed each sample and counted the cells stained with.-tubulin, verifying the percentage of affected cells to be a mean of 14%, with the number of clusters ranging between 0-6 per slide. In conclusion, centrosome amplification was found to be a consistent biological feature of CS and may underlie chromosomal instability in this tumor.info:eu-repo/semantics/publishedVersio

    Contribuições da extensão universitária na formação de acadêmicos de enfermagem

    Aim: to analyze the contributions of extension actions in the teaching of egressed students from the extension projects of AIDS, education and citizenship Program. Method: quantitative cross-sectional study. The participating population of the study was 43 egressed students from the extension project, linked to the nursing course of a Federal University. Data colletion occured through questionnaires that were sent by email from April to May, 2015 and were analyzed by Prictive Analytics software program. Results: the autonomies obtained after the participation in extension activities wesr emphasized, such as the autonomy on personal development, related to their own learning process, the teamwork, the mutual respect in the work enviroment and the academic improvement, since they had the opportunity to perform activities that are not found in the curriculum. Conclusion: The participation of academics in extension projects increases the responsibility to contribute decisively to social, individual and collective changesObjetivo: analizar las contribuciones de la extensión en la formación de egresos del proyecto de extensión del programa SIDA, la educación y la ciudadanía. Método: estudio transversal cuantitativo. Los participantes fueron de 43 egresos de proyectos de extensión vinculados al Curso de Enfermería de una Universidad Federal. La recolección de datos fue desarrollada en abril-mayo de 2015, el cuestionario fue enviado por correo electrónico. Los datos fueron analizados por el programa Predictive Analytics Software. Resultados: se destacó la autonomía adquirida después de la participación en las actividades extensionistas, tanto en el desarrollo personal, relacionado con el aprendizaje, trabajo en equipo y respeto al otro, cuanto en el desarrollo académico, en que tuvieron la oportunidad de realizar actividades que no son encontradas en el plan de estudios.     Multinucleon transfer reactions and proton transfer channels

    Transfer reactions have always been of great importance for nuclear structure and reaction mechanism studies. So far, in multinucleon transfer studies, proton pickup channels have been completely identified in atomic and mass numbers at energies close to the Coulomb barrier only in few cases. We measured the multinucleon transfer reactions in the 40Ar+208Pb system near the Coulomb barrier, by employing the PRISMA magnetic spectrometer. By using the most neutron-rich stable 40Ar beam we could populate, besidesneutron pickup and proton stripping channels, also neutron stripping and proton pickup channels. Comparison ofcross sections between different systems with the 208Pb target and with projectiles going from neutron-poor to neutron-rich nuclei, as well as between the data and GRAZING calculations, was carried out.Finally, recent results concerning the measurement of the excitation function from the Coulomb barrier to far below for the 92Mo+54Fe system, where both proton stripping and pickup channels were populated with similar strength, will be discussed
