Poly(2-propylacrylic acid)/poly(lactic-co-glycolic acid) blend microparticles as a targeted antigen delivery system to direct either CD4+ or CD8+ T cell activation.
Poly(lactic-co-glycolic acid) (PLGA) based microparticles (MPs) are widely investigated for their ability to load a range of molecules with high efficiency, including antigenic proteins, and release them in a controlled manner. Micron-sized PLGA MPs are readily phagocytosed by antigen presenting cells, and localized to endosomes. Due to low pH and digestive enzymes, encapsulated protein cargo is largely degraded and processed in endosomes for MHC-II loading and presentation to CD4+ T cells, with very little antigen delivered into the cytosol, limiting MHC-I antigenic loading and presentation to CD8+ T cells. In this work, PLGA was blended with poly(2-propylacrylic acid) (PPAA), a membrane destabilizing polymer, in order to incorporate an endosomal escape strategy into PLGA MPs as an easily fabricated platform with diverse loading capabilities, as a means to enable antigen presentation to CD8+ T cells. Ovalbumin (OVA)-loaded MPs were fabricated using a water-in-oil double emulsion with a 0% (PLGA only), 3 and 10% PPAA composition. MPs were subsequently determined to have an average diameter of 1 µm, with high loading and a release profile characteristic of PLGA. Bone marrow derived dendritic cells (DCs) were then incubated with MPs in order to evaluate localization, processing, and presentation of ovalbumin. Endosomal escape of OVA was observed only in DC groups treated with PPAA/PLGA blends, which promoted high levels of activation of CD8+ OVA-specific OT-I T cells, compared to DCs treated with OVA-loaded PLGA MPs which were unable activate CD8+ T cells. In contrast, DCs treated with OVA-loaded PLGA MPs promoted OVA-specific OT-II CD4+ T cell activation, whereas PPAA incorporation into the MP blend did not permit CD4+ T cell activation. These studies demonstrate PLGA MP blends containing PPAA are able to provide an endosomal escape strategy for encapsulated protein antigen, enabling the targeted delivery of antigen for tunable presentation and activation of either CD4+ or CD8+ T cells
Eye drop Self-medication: Comparative Questionnaire-based study of two Latin American cities.
A broad spectrum of ocular symptoms are treated by self-medication with commercial eye-drops. This behavior threatens individuals' visual health. In Latin America, evidence is poor.
Objective: To detect, characterize and compare patterns of ophthalmic self-medication between Córdoba (Argentina) and Barranquilla (Colombia).Design: Analytic, cross-sectional and comparative population-based study. Setting: Two private tertiary care ophthalmology centers from Córdoba, Argentina, and Barranquilla, Colombia.Participants: Patients 18 years of age or older who consulted for the first time in this two institutions duringAugust-November 2009, were included. A number of 570 patients were enrrolled.Methods: Data collected through a semi-structured questionnaire. Main outcome measure: To determine thefrequency of self-medication with eyedrops on a specific population of two cities in Latin America.Results: Comparable rates of ocular self-medication were found (25.6% and 25.7% for Cordoba and Barranquilla, respectively). The percentage of men and women who self-medicated was not significantly different between both samples. The major source of eye drops recommendation in the Argentineans patients was the pharmacist (31%); while the social source was predominant in Colombian individuals (53%). In Cordoba, the most frequently used product was a non-steroidal anti-inflammatory drop in combination with a vasoconstrictive agent (32%); while in Barranquilla, antibiotic eye drops were preferred (33%). Self-medication was higher between the ages of 31 and 50 years old in Argentinean citizens (28%) and between 18 to 31 years old in the Colombiancommunity (39%). This habit was found mostly in patients who completed university studies in Cordoba (33%); in Barranquilla, individuals with lower educational level practice more this behavior (36%).Conclusion: In both populations, patients commonly treat ocular conditions by self-medicating. Currently, anincreasing number of eye drops are obtainable without prescription and a high percentage of self-medicated patients in both samples ignore the possible side effects of the used medication.Fil: Marquez, Gabriel. Network process of children : influence of gender and type of game during cooperative-opposition games
The children, when playing, are communicating, since this is one of the forms of disinhibition and interaction with the group in which they are inserted (Lima, 2014). Ten children, five boys (n = 5; 5.8 ± 0.4 years old of age) and five girls (n = 5; 5.6 ± 0.5 years old of age) from a elementar school were observed during classes of motor expression. These classes took place once a week for a month, lasting one hour for each session. The interactions were observed with two cameras. It was counted as an interaction when a child passed one ball to another. The results suggest that it was in the “Free Game” that there were significant diferences between the type of game, since it was found in this one that it was easier for a player to be connected with his colleagues, being later the most requested, at the moment they had to pass the ball. Between the type of game and the gender, no significant interactions were found, as children who were the ones most requested by their classmates when they had to pass the ball were not verified. However, children who were crucial to maintaining cross-pass connections were also not checked. With regard to gender, no significant differences were found because no child, both male and female, has become important in sustaining the connections between the passes and the fact that no boys and / or girls were the most requested to make the passes with the ball. Finally, among the game types, no significant differences were found, since the children did not become essential to maintain the connection of passes between colleagues, in any of the matches played. Contribution of non-genetic factors to the reproductive performance of mirandesa cows
Characterization of reproductive traits in Mirandesa beef cattle is important for breed improvement and conservation, mainly due to its little genetic diversity. Reduced individual and maternal performance is often associated with inbreeding depression, which could be further aggravate the environmental effects. In this study, 7386 herd records for Mirandesa were used to characterize the main reproductive traits, like age at first calving (AFC), pregnancy length (PL), calving interval (CI), yearly calving distribution (CD) and productive lifespan (PLf). The non-genetic effects were tested using non-parametric methods, as the target variables were not normally distributed. The median for AFC in Mirandesa was close to 32 months; AFC was affected by the production system, farm and by the year and season of birth. The mean for PL was 287±8.9 days, being affected by parity and calf gender. The median CI, of 378 days, was only affected by the breeding program, parity, season and year. Calving season was unevenly distributed over the year, showing different patterns after the production system. The mean productive lifespan of Mirandesa was 6.45 years, though 20% of the cows presented a PLf longer than ten years. The main non-genetic effects suggested that farmer´s decision and nutrition may constrain the expression of the reproductive traits in Mirandesa breed. This aspect needs to be addressed when designing any breeding programs which should prioritise for the increase in the number of calves per year along with a careful selection of reproducers to decrease reported inbreeding.The authors wish to thank ACBRM (Associação de

Relationship between Prenatal or Postnatal Exposure to Pesticides and Obesity: A Systematic Review
In recent years, the worldwide prevalence of overweight and obesity among adults and children has dramatically increased. The conventional model regarding the onset of obesity is based on an imbalance between energy intake and expenditure. However, other possible environmental factors involved, such as the exposure to chemicals like pesticides, cannot be discarded. These compounds could act as endocrine-disrupting chemicals (EDC) that may interfere with hormone activity related to several mechanisms involved in body weight control. The main objective of this study was to systematically review the data provided in the scientific literature for a possible association between prenatal and postnatal exposure to pesticides and obesity in offspring. A total of 25 human and 9 animal studies were analyzed. The prenatal, perinatal, and postnatal exposure to organophosphate, organochlorine, pyrethroid, neonicotinoid, and carbamate, as well as a combined pesticide exposure was reviewed. This systematic review reveals that the effects of pesticide exposure on body weight are mostly inconclusive, finding conflicting results in both humans and experimental animals. The outcomes reviewed are dependent on many factors, including dosage and route of administration, species, sex, and treatment duration. Comparison of four steady-state models of increasing complexity for assessing the leaching requirement in agricultural salt-threatened soils
Irrigation scheduling in salt-threatened soils must include an estimation of the leaching requirement (LR). Many models have been developed over the last 40 years for assessing the LR, and they should be compared on common grounds to guide potential users. The LR for salts (LR gamma), chloride (LRCI) and SAR (LRSAR) and therefore the eventual LR was assessed with simple equations and three steady-state computer models of increasing complexity, WATSUIT, SALSODIMAR and SALTIRSOIL. These models were assessed in 30 scenarios characterised by different crops and water qualities in the irrigated area of the Vega Baja del Segura (SE Spain). The simple equations, WATSUIT and SALTIRSOIL calculated quite similar eventual LRs, which were between 0.99 depending on crop species and water quality. The SALSODIMAR gave remarkably higher eventual LRs (between 0.31 and > 0.99). This occurred because SALSODIMAR uses the hypothesis that the saturation extract is more concentrated than the drainage water, contrary to what is assumed by the simple equations or calculated by WATSUIT and SALTIRSOIL. Rainfall, which is not taken into account by the simple equations and WATSUIT, and soil calcite weathering, which is not taken into account by SALSODIMAR, were revealed, respectively, as important and very important aspects to be included in steady-state models. Although the SALTIRSOIL appears to be the most complete model, the simple equations give acceptably similar irrigation doses for many of the situations considered in this study. Iridium-(K2-NSi) catalyzed dehydrogenation of formic acid: effect of auxiliary ligands on the catalytic performance
IThe iridium(III) complexes [Ir(H)(Cl)(κ2-NSitBu2)(κ2-bipyMe2)] (2) and [Ir(H)(OTf)(κ2-NSitBu2)(κ2-bipyMe2)] (3) (NSitBu2 = {4-methylpyridine-2-yloxy}ditertbutylsilyl) have been synthesized and characterized including X-ray studies of 3. A comparative study of the catalytic activity of complexes 2, 3, [Ir(H)(OTf)(κ2-NSitBu2)(coe)] (4), and [Ir(H)(OTf)(κ2-NSitBu2)(PCy3)] (5) (0.1 mol%) as catalysts precursors for the solventless formic acid dehydrogenation (FADH) in the presence of Et3N (40 mol%) at 353 K has been performed. The highest activity (TOF5 min ≈ 3260 h−1) has been obtained with 3 at 373 K. However, at that temperature the FTIR spectra show traces of CO together with the desired products (H2 and CO2). Thus, the best performance was achieved at 353 K (TOF5 min ≈ 1210 h−1 and no observable CO). Kinetic studies at variable temperature show that the activation energy of the 3-catalyzed FADH process is 16.76 kcal mol−1. Kinetic isotopic effect (5 min) values of 1.6, 4.5, and 4.2 were obtained for the 3-catalyzed dehydrogenation of HCOOD, DCOOH, and DCOOD, respectively, at 353 K. The strong KIE found for DCOOH and DCOOD evidenced that the hydride transfer from the C–H bond of formic acid to the metal is the rate-determining step of the process
Maximising embryo production in endangered sheep breeds: in vitro procedures that complement in vivo techniques
This study investigated the use of previously superovulated ovaries as a source of oocytes, assessing the competence of them for in vitro embryo production. Two superovulatory treatments were performed: equine Chorionic Gonadotrophin (eCG) plus porcine Follicle-Stimulating Hormone (pFSH) in a single dose or the conventional protocol of six decreasing doses of pFSH. Thirty donor ewes of the endangered Ojalada breed were given either the simplified (group S; n=15) or the decreasing-dose (group D; n=15) treatments three times at intervals of ≥50 days. Ovaries were recovered on day 7 after the oestrus following the third treatment, just after embryo flushing, and the oocytes were collected to assess in vitro maturation, fertilisation and development to the blastocyst stage. The two superovulatory treatments did not differ in the mean number of oocytes selected for maturation (7.1±1.2 and 8.5±1.5 per ewe in the D and S groups, respectively). The oocytes recovered from ewes in Group D (87.5%) had a significantly (p<0.05) higher maturation rate than did those recovered from ewes in group S (75%), but no differences were found in fertilisation rate (94% and 94.6% in the D and S groups, respectively); both groups did not differ in their blastocyst rates and the total number of cells in in vitro-produced blastocysts. In the two experimental groups, 1.7 (D) and 1.8 (S) in vitro-produced blastocysts were generated per ewe, which indicate that it is feasible to combine in vivo and in vitro techniques to maximise embryo production in endangered sheep breeds.EEA ChubutFil: Forcada, Fernando. Correlation between cytologic features and histologic grades in cutaneous and subcutaneous soft tissue sarcomas in dogs—A pilot study
Abstract: Currently, canine soft tissue sarcoma (STS) grading is based on histopathology. In humans, several studies have demonstrated concordance between cytologic grading systems for STS and histologic grade. The aim of this study was to correlate several cytologic parameters (smear cellularity, anisokaryosis, nucleolar malignancy score, multinucleation, and the number of mitotic figures per 200 cells) that form part of a human STS cytologic grading system, with histologic grades of canine cutaneous and subcutaneous STS. Three observers (blinded) reviewed the cytologic preparations independently from cases with confirmed histologic diagnoses of STS. A cytologic grading score was assigned for each parameter. Correlations between cytologic grading scores (averaged between observers) and histologic grades were assessed using Spearman's correlation coefficient, with statistical significance defined as P < .05. Twenty‐one cases were included in the study (10 Grade I STS, nine Grade II STS, and two Grade III STS). The number of mitotic figures (≥3) per 200 cells was the only parameter that showed a significant but weak, positive correlation with histologic grade (rs = .469; P = .032). No Grade I tumors had ≥3 mitotic figures per 200 cells; however, ≥3 mitotic figures per 200 cells were only observed in 33% of Grade II tumors and 50% (one out of two) of the Grade III tumors. This pilot study suggests that an increased number of mitotic figures seen on cytology might correlate with higher grade STS; however, the sensitivity of this parameter for grading STS appears to be low
