IF/00225/2015The multistate of species originated by the compound 5-hydroxy-4′-dimethylaminoflavylium was studied in the presence of sodium dodecyl sulfate micelles 0.1 M. A series of pH jumps obtained by addition of base to the equilibrated solutions of the flavylium cation results in a metastable equilibrium between the flavylium cation and the quinoidal base with pKa = 7.6. In neutral to moderately basic medium, only at 60 °C and after 5 days a new distribution involving the flavylium cation and trans-chalcones is achieved. On the other hand, the anionic trans-chalcone can be formed in few minutes in very basic medium at room temperature and from this, another meta-stable state constituted by protonated, neutral and anionic trans-chalcones, pKCt+/Ct = 2.6 and pKCt/Ct- = 10.7 is reached. The equilibrium is apparently attained between flavylium cation, neutral and anionic trans-chalcones respectively for pKAH+/Ct = 4.1 and pKCt/Ct- = 10.7. Irradiation of the neutral (stable or metastable) and protonated (meta-stable) trans-chalcones yields to the appearance of the quinoidal base or the flavylium cation in an extended pH region 1 < pH < 10. The peculiar properties of this flavylium based photochromic system are explained by means of an energy level diagram where cis-chalcone and hemiketal lay above the quinoidal base.authorsversionpublishe

O risco de consumo desses alimentos foi maior entre crianças de famÃlias de menor posição socioeconômica, filhas de mães de baixa escolaridade, de cor da pele preta, mais jovens e de baixa renda.OBJECTIVE Assessing the regular consumption of ultra-processed foods by children at 24 months of age from the 2015 Pelotas Birth Cohort and the main demographic, socioeconomic, and behavioral factors related to the consumption of these products. METHODS Population-based cohort in the city of Pelotas, RS, where 4,275 children were assessed at birth and 95.4% of them were followed up until 24 months of age. Food consumption was assessed by a questionnaire on regular consumption of ultra-processed foods, which collected information regarding sex, household income, maternal skin color, schooling level, and age, the child attending day care and having siblings, breastfeeding status, and obesity. The outcome was the sum of ultra-processed foods regularly consumed by a child. A multivariate Poisson regression analysis was used to calculate the association between the regular consumption of ultra-processed foods and exposure variables. RESULTS The mean number of ultra-processed foods consumed was 4.8 (SD = 2.3). The regular consumption of ultra-processed foods was positively associated with black skin color and having siblings, and negatively associated with household income and maternal schooling level and age. CONCLUSION The mean regular consumption of ultra-processed foods by children from the 2015 Pelotas Birth Cohort is high, which can negatively affect the children’s diet. The risk of consuming this kind of food was higher among children from families of lower socioeconomic status, whose mothers present lower education level, black skin color, and younger age
Pyrazoles as potential modulators of inflammation through the inhibition of COX2 activity and human leukocytes' oxidative burst
The inflammatory process is a complex and tightly regulated cascade of events that involves the production of prostaglandins (PG) by the inducible isoform cyclooxygenase 2 (COX-2) and the production of reactive pro-oxidant species. When the production of these mediators becomes excessive, it can lead to chronic inflammation and associated diseases such as diabetes, rheumatoid arthritis, and cancer. Unfortunately, many existing anti-inflammatory agents are associated with unwanted side effects. Therefore, there is a critical need to discover new and effective compounds that can modulate the inflammatory cascade. In this study, an extensive panel of structurally related pyrazoles holding diverse structures and substitutions were tested in vitro against human COX-2, and ex vivo in human whole blood, through the measurement of prostaglandin E2 (PGE2) production. Their potential inhibitory effect against human leukocytes’ oxidative burst was also studied. The results showed that some of the tested compounds had a significant inhibitory effect on COX2 activity, and pyrazoles 4 and 11 (Figure 1) excelled as the most potent inhibitors, with IC50 < 25 µM. Nonetheless, among the tested compounds only 1 was able to inhibit both the COX-2 activity and the PGE2 production. The tested pyrazoles, namely pyrazole 4, also demonstrated a potential inhibitory effect (IC50 < 5 µM) against human leukocytes’ oxidative burst. These results represent a significant contribution for the design and development of new anti-inflammatory molecules.info:eu-repo/semantics/publishedVersio
Tricoblastoma trabecular em gato doméstico (Felis catus domesticus, Linnaeus 1758): relato de caso
Trichoblastoma is characterized as a benign cutaneous neoplasm that originates in the trichoblastic epithelium of the hair bulb. The present report describes the case of a 4-year-old cross breed neutered male cat that had a history of a slow, freely movable, ulcerated, slow growing, solitary cutaneous nodule located on the left scapular region. Histopathological evaluation showed neoplastic cells arranged in cohesive nests or palisades with eosinophilic and scant cytoplasm. Based on the microscopic findings, a diagnosis of trabecular trichoblastoma was made. The treatment of choice consisted of total surgical excision of the mass with safe surgical margins.
Is painless synovitis different from painful synovitis? A controlled, ultrasound, radiographic, clinical trial
OBJECTIVE: This study compares the clinical, ultrasonography, radiography, and laboratory outcomes of painless and painful chronic synovitis in patients with established rheumatoid arthritis. METHODS: This cross-sectional study involved 60 patients with rheumatoid arthritis and synovitis in the metacarpophalangeal joints; 30 of the patients did not experience pain, and 30 had experienced pain for at least 6 months prior to the study. The radiocarpal, distal radioulnar, and metacarpophalangeal joints were evaluated using the ultrasound gray scale, power Doppler, and radiography. Past and present clinical and laboratory findings were also evaluated. RESULTS: There were no statistically significant differences between the groups for most of the outcomes. The group with pain scored worse on the disease activity indices (e.g., DAS 28 and SDAI), function questionnaires (HAQ and Cochin), and pinch strength test. A logistic regression analysis revealed that the use of an immunobiological agent was associated with a 3-fold greater chance of belonging to the group that experienced pain. The painless group had worse erosion scores in the second and fifth metacarpophalangeal with odd ratios (ORs) of 6.5 and 3.5, respectively. The painless group had more cartilage with grade 4 damage in the third metacarpophalangeal. CONCLUSIONS: The rheumatoid arthritis patients with both painless and painful synovitis exhibited similar disease histories and radiographic and ultrasound findings. However, the ultrasonography evaluation revealed worse scores in the second and fifth metacarpophalangeal of the synovitis patients who did not experience pain.Fundacao de Amparo a Pesquisa do Estado de Sao PauloUNIFESP communityHospital do Servidor Publico Estadual (IAMSPE)Universidade Federal de São Paulo (UNIFESP) Escola Paulista de Medicina Disciplina de ReumatologiaUniversidade Federal de São Paulo (UNIFESP) Escola Paulista de Medicina Departamento de Diagnostico por ImagemUNIFESP, EPM, Disciplina de ReumatologiaUNIFESP, EPM, Depto. de Diagnostico por ImagemFAPESP - 2012/04867-6SciEL
Effect of green tea extract on bonding durability of an etch-and-rinse adhesive system to caries-affected dentin
Objective Green tea extract has been advocated as a matrix metalloproteinase (MMP) inhibitor; however, its effect on bond durability to caries-affected dentin has never been reported. Thus, the aim of this in vitro study was to evaluate the effect of two MMP inhibitors (2% chlorhexidine and 2% green tea extract), applied after acid etching, on bond durability of an etch-and-rinse adhesive system to caries-affected dentin. Material and Methods Occlusal enamel was removed from third molars to expose the dentin surface, and the molars were submitted to a caries induction protocol for 15 days. After removal of infected dentin, specimens were conditioned with 37% phosphoric acid (15 seconds) and randomly divided into three groups, according to the type of dentin pretreatment (n=10): NT: no treatment; GT: 2% green tea extract; CLX: 2% chlorhexidine. The etch-and-rinse adhesive system (Adper™ Single Bond 2, 3M ESPE, St. Paul, MN, USA) was applied according to the manufacturer's instructions, and composite resin restorations were built on the dentin. After 24 hours, at 37°C, the resin-tooth blocks were sectioned perpendicularly to the adhesive interface in the form of sticks (0.8 mm2 of adhesive area) and randomly subdivided into two groups according to when they were to be submitted to microtensile bond strength (μTBS) testing: immediately or 6 months after storage in distilled water. Data were reported in MPa and submitted to two-way ANOVA for completely randomized blocks, followed by Tukey’s test (α=0.05). Results After 24 hours, there was no significant difference in the μTBS of the groups. After 6 months, the GT group had significantly higher μTBS values. Conclusion It was concluded that the application of 2% green tea extract was able to increase bond durability of the etch-and-rinse system to dentin. Neither the application of chlorhexidine nor non-treatment (NT - control) had any effect on bond strength after water storage
