    Předmětem diplomové práce je analýza současného stavu nákupní strategie firmy vedoucí k identifikaci prostorů pro zlepšení nákupu firmy. Cílem práce je návrh nové strategie nákupu se zaměřením na zakázky. Na základě analýzy práce obsahuje návrh na rozšíření portfolia nabízených produktů o vhodný výrobek, návrh vypořádání se se specifickými požadavky zákazníků a návrh na zavedení strategie just-in-time. Návrhy přispívají k efektivnímu řízení nákupu.The subject of the thesis is to analyze the current state of company’s purchasing strategy, leading to identify areas for improvement of company’s purchase. The aim is to draft a new purchasing strategy while focusing on contracts. Based on the analysis, the thesis includes a proposal to expand the portfolio of products with a proper product, a proposal to deal with the specific customers’ requirements and a proposal of implementation of the strategy just-in-time. Proposals contribute to the effective management of purchasing.

    Accumulation of heavy metals in the ecosystem and their toxic effects through food chain can cause serious ecological and health problems. In the present study, experiments were performed to understand how the addition of magnetite (Fe3O4) nanoparticles reduces the toxicity caused by Cd, Pb, Cu, and Zn in cucumber plants. Plant growth parameters, lipid peroxidation, and antioxidant enzymes were measured in seedling samples treated with either metals or metals supplemented with Fe3O4 to demonstrate the reduction in metal-induced oxidative stress conferred by Fe3O4. Results showed that the toxic effect of metals on seedling growth parameters can be arranged in the rank order of inhibition as follows: Cu > Cd > Zn > Pb. Exposure to metals significantly decreased the seedlings growth, the activities of superoxide dismutase (SOD) and peroxidases (POD), while the malondialdehyde (MDA) content significantly increased in cucumber seedlings. The reducing activity of nano-Fe3O4 against heavy metals stresses was confirmed in this study by the decrease in MDA content. The correlation between the decrease of MDA concentration and the increase in SOD and POD activities in the presence of nano-Fe3O4 suggest that the MDA reduction in the tested seedlings can result from the increased enzyme activity

    Assessment of phenolic herbicide toxicity and mode of action by different assays

    A phytotoxicity assay based on seed germination/root elongation has been optimized and used to evaluate the toxic effects of some phenolic herbicides. The method has been improved by investigating the influence of experimental conditions. Lepidium sativum was chosen as the most suitable species, showing high germinability, good repeatability of root length measurements, and low sensitivity to seed pretreatment. DMSO was the most appropriate solvent carrier for less water-soluble compounds. Three dinitrophenols and three hydroxybenzonitriles were tested: dinoterb, DNOC, 2,4-dinitrophenol, chloroxynil, bromoxynil, and ioxynil. Toxicity was also determined using the Vibrio fischeri MicrotoxA (R) test, and a highly significant correlation was found between EC50 values obtained by the two assays. Dinoterb was the most toxic compound. The toxicity of hydroxybenzonitriles followed the order: ioxynil > bromoxynil > chloroxynil; L. sativum exhibited a slightly higher sensitivity than V. fischeri to these compounds. A QSAR analysis highlighted the importance of hydrophobic, electronic, and hydrogen-bonding interactions, in accordance with a mechanism of toxic action based on protonophoric uncoupling of oxidative phosphorylation. The results suggest that the seed germination/root elongation assay with L. sativum is a valid tool for the assessment of xenobiotic toxicity and can be recommended as part of a test battery

    Plant stress activated by chlorine from disinfectants prepared on the base of sodium hypochlorite

    In this study, the phytotoxicity of disinfectants prepared on the base of sodium hypochlorite was determined. For our tests two commercial products, Savo and Dom Amor, as well as 10% NaClO solution were used. While Savo contained only NaClO, Dom Amor contained NaClO and earthworm enzymes. Products on the base of NaClO are used in households for cleaning and disinfection of floors, furniture, sanitary and kitchen equipment. Savo may be used for the disinfection of drinking waters as well. Products with NaClO are also used for bacteria, algae and pathogens reduction in irrigation waters. As a subject, young seedlings of mustard Sinapis alba L. were used for the study of chronic toxicity. The observed parameters of the inhibition of roots and shoots growth, dry (DM) and fresh (FM) mass as well as photosynthetic pigments production (chlorophyll a, b, carotenoids) and water content in the plants were determined. The results point out that Dom Amor was the most toxic for S. alba seedlings growth and the rank order of the FAC contents for both plant parts was arranged as: Dom Amor > Savo > NaClO. All disinfectants reduced the DM and FM of roots; however, they stimulated biomass production in the shoots. On the basis of the obtained results it could be concluded, that disinfectants stimulated photosynthetic pigments production and reduced water content mainly in the roots. Dom Amor did not significantly reduced the water content in the shoots and for this parameter the following rank orders of inhibition for roots and shoots could be arranged as NaClO > Dom Amor > Savo and NaClO > Savo > Dom Amor, respectively. All commercial products increased chlorophyll a (Chla) and the carotenoids (Car) content in the shoots. As significant increase was confirmed first for Chla whose content in the presence of NaClO at concentration 24 mL/L overextended that in the control by 3.5 times. The rank orders of stimulation for Chla and Car were NaClO > Savo > Dom Amor and Dom Amor > NaClO > Savo, respectively
