    The possibility of producing high carbon steel/Inconel 718 bimetallic parts via Fused Filament Fabrication and Sintering is explored. Compatibility of the two alloys with particular attention to elements interdiffusion through the interface as well as the effect of the deposition strategy were analyzed. Microstructural features, relative density and parts shrinkage were investigated, as well. Although first-tentative process parameters values were not sufficient to reach an acceptable material densification, a good bonding between Inconel 718 and carbon steel was observed, suggesting the potential to obtain sound bimetallic parts with a great range of material properties. Due to a difference in densification kinetics, sintering temperature was revealed to be the most critical process parameter to optimize to minimize porosity

    Inconel 625 is a nickel-based superalloy that finds application in many industrial sectors thanks to its high strength, excellent fabricability, and  outstanding corrosion resistance. It is characterized by a good weldability and often used in the as-welded conditions. In the following, a metallurgical and thermal analysis of Inconel 625 Electron-Beam welded joints is described. The thermal analysis was supported by a numerical model that uses a superimposition of a spherical and a conical shape heat source with Gaussian power density distribution in order to reproduce the nail-shape of the fusion zone. The heat source parameters were calibrated by using experimental data coming from metallurgical observations and temperature measurements.     Immobilization of Lathyrus cicera Amine Oxidase on Magnetic Microparticles for Biocatalytic Applications

    Amine oxidases are enzymes belonging to the class of oxidoreductases that are widespread, from bacteria to humans. The amine oxidase from Lathyrus cicera has recently appeared in the landscape of biocatalysis, showing good potential in the green synthesis of aldehydes. This enzyme catalyzes the oxidative deamination of a wide range of primary amines into the corresponding aldehydes but its use as a biocatalyst is challenging due to the possible inactivation that might occur at high product concentrations. Here, we show that the enzyme’s performance can be greatly improved by immobilization on solid supports. The best results are achieved using amino-functionalized magnetic microparticles: the immobilized enzyme retains its activity, greatly improves its thermostability (4 h at 75 ◦C), and can be recycled up to 8 times with a set of aromatic ethylamines. After the last reaction cycle, the overall conversion is about 90% for all tested substrates, with an aldehyde production ranging between 100 and 270 mg depending on the substrate used. As a proof concept, one of the aldehydes thus produced was successfully used for the biomimetic synthesis of a non-natural benzylisoquinoline alkaloid

    Unveiling the impact of laser power variations on microstructure, corrosion, and stress-assisted surface crack initiation in laser powder bed fusion-processed Ni-Fe-Cr alloy 718

    Corrosion and stress-corrosion related failures often compromise the integrity of critical metallic components during their service, raising significant concerns. It is crucial to comprehend the crack initiation mechanism and the impact of alloy microstructure on this crack initiation process. It is known that the introduction of unique microstructures through metal additive manufacturing brings new challenges. This study aims to investigate, for the first time, the effects of microstructural alterations resulting from fluctuations in laser power during laser powder bed fusion on the surface cracking initiation mechanism and electrochemical behaviour of Ni-Fe-Cr alloy 718, which is widely used in applications that require exceptional strength and corrosion resistance. To carry out this investigation, microcapillary electrochemical methods were combined with high-resolution techniques (TEM, SEM, AFM). The findings emphasize the existence of an optimal range of process parameters that effectively mitigate corrosion and crack initiation susceptibility. This work demonstrated that slight deviations in laser power from this optimal value result in diverse alterations at the micro and submicron scales. These alterations include increased subgrain width, porosity, dislocation density, density of nanovoids, and distribution of carbides. Importantly, these changes, particularly in dislocation and nanovoid densities caused by minor variations in process parameters, significantly affect the material's susceptibility to corrosion initiation and stress-assisted surface cracking

    Increasing the Liquidus Temperature by Employing the Controlled Diffusion Solidification (CDS) Process: A Potential Route to Improved Castings

    Abstract Recent theories suggest the existence of an incubation time, over which a liquid alloy prepares for nucleation by decomposing into compositional fluctuations. Accordingly, in a recent work by the present authors, the solidification path of a Controlled Diffusion Solidification (CDS) mixture was calculated. The calculated CDS path begins at a higher liquidus temperature comparing to conventional solidification and the fraction solid values are achieved at a relatively higher temperature. To provide information on the CDS mechanism and physical structure of the CDS mixture in the mushy zone, Al-7.8Zn-2.6Mg-2Cu alloy was solidified, in this study, via conventional and CDS process in the presence and absence of recalescence. Typical grain structures obtained via the two solidification conditions is characterized using Electron Back Scattered Diffraction. Results showed that the nucleation continues to occur in the presence of recalescence, while it is suppressed in its absence. According to the two step nucleation theory, the increase in the nucleation temperature causes sufficient recalescence in the mixture, allowing the unnucleated liquid phase to decompose into chemical fluctuations and prepares for further nucleation. As a result, in the presence of recalescence, nucleation in a CDS mixture is not as readily halted as during the conventional solidification, which is in contradiction with the recent theories developed based on the classical theory of nucleation

    Rare mutations in SQSTM1 modify susceptibility to frontotemporal lobar degeneration

    Mutations in the gene coding for Sequestosome 1 (SQSTM1) have been genetically associated with amyotrophic lateral sclerosis (ALS) and Paget disease of bone. In the present study, we analyzed the SQSTM1 coding sequence for mutations in an extended cohort of 1,808 patients with frontotemporal lobar degeneration (FTLD), ascertained within the European Early-Onset Dementia consortium. As control dataset, we sequenced 1,625 European control individuals and analyzed whole-exome sequence data of 2,274 German individuals (total n = 3,899). Association of rare SQSTM1 mutations was calculated in a meta-analysis of 4,332 FTLD and 10,240 control alleles. We identified 25 coding variants in FTLD patients of which 10 have not been described. Fifteen mutations were absent in the control individuals (carrier frequency < 0.00026) whilst the others were rare in both patients and control individuals. When pooling all variants with a minor allele frequency < 0.01, an overall frequency of 3.2 % was calculated in patients. Rare variant association analysis between patients and controls showed no difference over the whole protein, but suggested that rare mutations clustering in the UBA domain of SQSTM1 may influence disease susceptibility by doubling the risk for FTLD (RR = 2.18 [95 % CI 1.24-3.85]; corrected p value = 0.042). Detailed histopathology demonstrated that mutations in SQSTM1 associate with widespread neuronal and glial phospho-TDP-43 pathology. With this study, we provide further evidence for a putative role of rare mutations in SQSTM1 in the genetic etiology of FTLD and showed that, comparable to other FTLD/ALS genes, SQSTM1 mutations are associated with TDP-43 pathology

    Prescription appropriateness of anti-diabetes drugs in elderly patients hospitalized in a clinical setting: evidence from the REPOSI Register

    Diabetes is an increasing global health burden with the highest prevalence (24.0%) observed in elderly people. Older diabetic adults have a greater risk of hospitalization and several geriatric syndromes than older nondiabetic adults. For these conditions, special care is required in prescribing therapies including anti- diabetes drugs. Aim of this study was to evaluate the appropriateness and the adherence to safety recommendations in the prescriptions of glucose-lowering drugs in hospitalized elderly patients with diabetes. Data for this cross-sectional study were obtained from the REgistro POliterapie-Società Italiana Medicina Interna (REPOSI) that collected clinical information on patients aged ≥ 65 years acutely admitted to Italian internal medicine and geriatric non-intensive care units (ICU) from 2010 up to 2019. Prescription appropriateness was assessed according to the 2019 AGS Beers Criteria and anti-diabetes drug data sheets.Among 5349 patients, 1624 (30.3%) had diagnosis of type 2 diabetes. At admission, 37.7% of diabetic patients received treatment with metformin, 37.3% insulin therapy, 16.4% sulfonylureas, and 11.4% glinides. Surprisingly, only 3.1% of diabetic patients were treated with new classes of anti- diabetes drugs. According to prescription criteria, at admission 15.4% of patients treated with metformin and 2.6% with sulfonylureas received inappropriately these treatments. At discharge, the inappropriateness of metformin therapy decreased (10.2%, P &lt; 0.0001). According to Beers criteria, the inappropriate prescriptions of sulfonylureas raised to 29% both at admission and at discharge. This study shows a poor adherence to current guidelines on diabetes management in hospitalized elderly people with a high prevalence of inappropriate use of sulfonylureas according to the Beers criteria

    Antidiabetic Drug Prescription Pattern in Hospitalized Older Patients with Diabetes

    Objective: To describe the prescription pattern of antidiabetic and cardiovascular drugs in a cohort of hospitalized older patients with diabetes. Methods: Patients with diabetes aged 65 years or older hospitalized in internal medicine and/or geriatric wards throughout Italy and enrolled in the REPOSI (REgistro POliterapuie SIMI—Società Italiana di Medicina Interna) registry from 2010 to 2019 and discharged alive were included. Results: Among 1703 patients with diabetes, 1433 (84.2%) were on treatment with at least one antidiabetic drug at hospital admission, mainly prescribed as monotherapy with insulin (28.3%) or metformin (19.2%). The proportion of treated patients decreased at discharge (N = 1309, 76.9%), with a significant reduction over time. Among those prescribed, the proportion of those with insulin alone increased over time (p = 0.0066), while the proportion of those prescribed sulfonylureas decreased (p &lt; 0.0001). Among patients receiving antidiabetic therapy at discharge, 1063 (81.2%) were also prescribed cardiovascular drugs, mainly with an antihypertensive drug alone or in combination (N = 777, 73.1%). Conclusion: The management of older patients with diabetes in a hospital setting is often sub-optimal, as shown by the increasing trend in insulin at discharge, even if an overall improvement has been highlighted by the prevalent decrease in sulfonylureas prescription

    The “Diabetes Comorbidome”: A Different Way for Health Professionals to Approach the Comorbidity Burden of Diabetes

    (1) Background: The disease burden related to diabetes is increasing greatly, particularly in older subjects. A more comprehensive approach towards the assessment and management of diabetes’ comorbidities is necessary. The aim of this study was to implement our previous data identifying and representing the prevalence of the comorbidities, their association with mortality, and the strength of their relationship in hospitalized elderly patients with diabetes, developing, at the same time, a new graphic representation model of the comorbidome called “Diabetes Comorbidome”. (2) Methods: Data were collected from the RePoSi register. Comorbidities, socio-demographic data, severity and comorbidity indexes (Cumulative Illness rating Scale CIRS-SI and CIRS-CI), and functional status (Barthel Index), were recorded. Mortality rates were assessed in hospital and 3 and 12 months after discharge. (3) Results: Of the 4714 hospitalized elderly patients, 1378 had diabetes. The comorbidities distribution showed that arterial hypertension (57.1%), ischemic heart disease (31.4%), chronic renal failure (28.8%), atrial fibrillation (25.6%), and COPD (22.7%), were the more frequent in subjects with diabetes. The graphic comorbidome showed that the strongest predictors of death at in hospital and at the 3-month follow-up were dementia and cancer. At the 1-year follow-up, cancer was the first comorbidity independently associated with mortality. (4) Conclusions: The “Diabetes Comorbidome” represents the perfect instrument for determining the prevalence of comorbidities and the strength of their relationship with risk of death, as well as the need for an effective treatment for improving clinical outcomes
