    Characterization of Subcellular Organelles in Cortical Perisynaptic Astrocytes

    Perisynaptic astrocytic processes (PAPs) carry out several different functions, from metabolite clearing to control of neuronal excitability and synaptic plasticity. All these functions are likely orchestrated by complex cellular machinery that resides within the PAPs and relies on a fine interplay between multiple subcellular components. However, traditional transmission electron microscopy (EM) studies have found that PAPs are remarkably poor of intracellular organelles, failing to explain how such a variety of PAP functions are achieved in the absence of a proportional complex network of intracellular structures. Here, we use serial block-face scanning EM to reconstruct and describe in three dimensions PAPs and their intracellular organelles in two different mouse cortical regions. We described five distinct organelles, which included empty and full endosomes, phagosomes, mitochondria, and endoplasmic reticulum (ER) cisternae, distributed within three PAPs categories (branches, branchlets, and leaflets). The majority of PAPs belonged to the leaflets category (~60%), with branchlets representing a minority (~37%). Branches were rarely in contact with synapses (<3%). Branches had a higher density of mitochondria and ER cisternae than branchlets and leaflets. Also, branches and branchlets displayed organelles more frequently than leaflets. Endosomes and phagosomes, which accounted for more than 60% of all the organelles detected, were often associated with the same PAP. Likewise, mitochondria and ER cisternae, representing ~40% of all organelles were usually associated. No differences were noted between the organelle distribution of the somatosensory and the anterior cingulate cortex. Finally, the organelle distribution in PAPs did not largely depend on the presence of a spine apparatus or a pre-synaptic mitochondrion in the synapse that PAPs were enwrapping, with some exceptions regarding the presence of phagosomes and ER cisternae, which were slightly more represented around synapses lacking a spine apparatus and a presynaptic mitochondrion, respectively.     Effects of light intensity on the distribution of anthocyanins in Kalanchoe brasiliensis Camb. and Kalanchoe pinnata (Lamk.) Pers

    This paper compares two medicinal species of Kalanchoe, which are often used interchangeably by the population, regarding the distribution of anthocyanins under the influence of four luminosity levels for 6 months. For the morphoanatomical analysis, the 6th stem node of each plant was sectioned. Usual histochemical tests revealed the presence of anthocyanins by cross sections of the stems, petioles and leaf blades. The petioles and leaf blades were submitted to the extraction with acidified methanol, and the anthocyanins were quantified by spectrophotometric readings. At the macroscopic level, it was noticed for both species a higher presence of anthocyanins in stems and petioles of plants under full sunlight. The microscopy of K. brasiliensis stems evidenced the deposition of anthocyanins in the subjacent tissue to the epidermis and cortex, which increased with light intensity. In K. pinnata a subepidermal collenchyma was observed, which interfered in the visualization of anthocyanins. In petioles and leaf blades of K. brasiliensis the deposition of anthocyanins was peripheral, and in K. pinnata it was also throughout the cortex. The quantification of anthocyanins in petioles showed in 70% of light higher averages than in 25%, but in leaf blades there were no significant results. This study contributes to the pharmacognosy of Kalanchoe and it is sustained by the description of flavonoids as biological markers of the genus.Este trabalho compara duas espécies medicinais de Kalanchoe utilizadas muitas vezes de forma indiferenciada pela população, quanto à distribuição de antocianinas sob influência de quatro níveis de luminosidade por 6 meses. Para a análise morfoanatômica foi seccionado o 6 º nó do caule de cada planta. Testes histoquímicos clássicos evidenciaram a presença de antocianinas em cortes transversais dos caules, pecíolos e lâminas foliares. Os pecíolos e lâminas foliares foram submetidos à extração com metanol acidificado e as antocianinas foram quantificadas através de leituras espectrofotométricas. Observou-se em nível macroscópico nas duas espécies, maior presença de antocianinas nos caules e pecíolos das plantas sob luz plena. A microscopia de caules de K. brasiliensis evidenciou a deposição de antocianinas no tecido subjacente à epiderme e córtex, o que aumentou com a intensidade luminosa. Em K. pinnata, observou-se colênquima subepidérmico, o que interferiu na visualização de antocianinas. Nos pecíolos e lâminas foliares de K. brasiliensis, a deposição de antocianinas foi periférica e em K. pinnata, se deu também por todo o córtex. A quantificação de antocianinas dos pecíolos mostrou, em 70% de luminosidade, teores superiores aos obtidos em 25%, porém nas lâminas foliares não foram obtidos resultados significativos. Este trabalho contribui para a farmacognosia de Kalanchoe e sustenta-se pela descrição de flavonóides como marcadores biológicos do gênero

    It is shown that the breaking of the conformal invariance in quantum electrodynamics due to the trace anomaly results in the generation of long wave electromagnetic fields during inflationary stage of the universe evolution.     É cada vez maior o número de empresas que estão distribuindo seus processos de produção de software ao redor do mundo, visando ganhos de produtividade, redução de custos e melhorias na qualidade. Engenheiros de software têm reconhecido a grande influência desta forma de trabalho e estão em busca de processos que facilitem projetos distribuídos. Além dos engenheiros também os gerentes têm enfrentado desafios em diferentes níveis no desenvolvimento distribuído de software (DDS), em especial no monitoramento do projeto. Desta forma, este trabalho tem como objetivo comprovar a seguinte hipótese: Um projeto distribuído de software pode ter o cronograma de suas atividades monitoradas, por meio de um processo que aplique a técnica do Kanban e a modelagem BPMN. O método utilizado para validação do modelo proposto foi o experimental, que simulou um ambiente real de DDS, mobilizando 82 desenvolvedores. Para uma aplicação inicial, o processo apresentou resultados positivos, portanto é eficaz para o monitoramento do cronograma das atividades de um projeto distribuído

    Seasonlity of Kawasaki Disease: A global perspective

    The authors are for the Kawasaki Disease Global Climate ConsortiumBACKGROUND: Understanding global seasonal patterns of Kawasaki disease (KD) may provide insight into the etiology of this vasculitis that is now the most common cause of acquired heart disease in children in developed countries worldwide. METHODS: Data from 1970-2012 from 25 countries distributed over the globe were analyzed for seasonality. The number of KD cases from each location was normalized to minimize the influence of greater numbers from certain locations. The presence of seasonal variation of KD at the individual locations was evaluated using three different tests: time series modeling, spectral analysis, and a Monte Carlo technique. RESULTS: A defined seasonal structure emerged demonstrating broad coherence in fluctuations in KD cases across the Northern Hemisphere extra-tropical latitudes. In the extra-tropical latitudes of the Northern Hemisphere, KD case numbers were highest in January through March and approximately 40% higher than in the months of lowest case numbers from August through October. Datasets were much sparser in the tropics and the Southern Hemisphere extra-tropics and statistical significance of the seasonality tests was weak, but suggested a maximum in May through June, with approximately 30% higher number of cases than in the least active months of February, March and October. The seasonal pattern in the Northern Hemisphere extra-tropics was consistent across the first and second halves of the sample period. CONCLUSION: Using the first global KD time series, analysis of sites located in the Northern Hemisphere extra-tropics revealed statistically significant and consistent seasonal fluctuations in KD case numbers with high numbers in winter and low numbers in late summer and fall. Neither the tropics nor the Southern Hemisphere extra-tropics registered a statistically significant aggregate seasonal cycle. These data suggest a seasonal exposure to a KD agent that operates over large geographic regions and is concentrated during winter months in the Northern Hemisphere extra-tropics.published_or_final_versio
