    Selective laser melting–enabled electrospinning: Introducing complexity within electrospun membranes

    Additive manufacturing technologies enable the creation of very precise and well-defined structures that can mimic hierarchical features of natural tissues. In this article, we describe the development of a manufacturing technology platform to produce innovative biodegradable membranes that are enhanced with controlled microenvironments produced via a combination of selective laser melting techniques and conventional electrospinning. This work underpins the manufacture of a new generation of biomaterial devices that have significant potential for use as both basic research tools and components of therapeutic implants. The membranes were successfully manufactured and a total of three microenvironment designs (niches) were chosen for thorough characterisation. Scanning electron microscopy analysis demonstrated differences in fibre diameters within different areas of the niche structures as well as differences in fibre density. We also showed the potential of using the microfabricated membranes for supporting mesenchymal stromal cell culture and proliferation. We demonstrated that mesenchymal stromal cells grow and populate the membranes penetrating within the niche-like structures. These findings demonstrate the creation of a very versatile tool that can be used in a variety of tissue regeneration applications including bone healing

    Computational methodology to determine fluid related parameters on non regular three-dimensional scaffolds

    The application of three-dimensional (3D) biomaterials to facilitate the adhesion, proliferation, and differentiation of cells has been widely studied for tissue engineering purposes. The fabrication methods used to improve the mechanical response of the scaffold produce complex and non regular structures. Apart from the mechanical aspect, the fluid behavior in the inner part of the scaffold should also be considered. Parameters such as permeability (k) or wall shear stress (WSS) are important aspects in the provision of nutrients, the removal of metabolic waste products or the mechanically-induced differentiation of cells attached in the trabecular network of the scaffolds. Experimental measurements of these parameters are not available in all labs. However, fluid parameters should be known prior to other types of experiments. The present work compares an experimental study with a computational fluid dynamics (CFD) methodology to determine the related fluid parameters (k and WSS) of complex non regular poly(L-lactic acid) scaffolds based only on the treatment of microphotographic images obtained with a microCT (lCT). The CFD analysis shows similar tendencies and results with low relative difference compared to those of the experimental study, for high flow rates. For low flow rates the accuracy of this prediction reduces.     Emergence of 3D Printed Dosage Forms: Opportunities and Challenges

    The recent introduction of the first FDA approved 3D-printed drug has fuelled interest in 3D printing technology, which is set to revolutionize healthcare. Since its initial use, this rapid prototyping (RP) technology has evolved to such as extent that it is currently being used in a wide range of applications including in tissue engineering, dentistry, construction, automotive and aerospace. However, in the pharmaceutical industry this technology is still in its infancy and its potential yet to be fully explored. This paper presents various 3D printing technologies such as stereolithographic, powder based, selective laser sintering, fused deposition modelling and semi-solid extrusion 3D printing. It also provides a comprehensive review of previous attempts at using 3D printing technologies on the manufacturing dosage forms with a particular focus on oral tablets. Their advantages particularly with adaptability in the pharmaceutical field have been highlighted, including design flexibility and control and manufacture which enables the preparation of dosage forms with complex designs and geometries, multiple actives and tailored release profiles. An insight into the technical challenges facing the different 3D printing technologies such as the formulation and processing parameters is provided. Light is also shed on the different regulatory challenges that need to be overcome for 3D printing to fulfil its real potential in the pharmaceutical industry

    Combining regenerative medicine strategies to provide durable reconstructive options: auricular cartilage tissue engineering

    Recent advances in regenerative medicine place us in a unique position to improve the quality of engineered tissue. We use auricular cartilage as an exemplar to illustrate how the use of tissue-specific adult stem cells, assembly through additive manufacturing and improved understanding of postnatal tissue maturation will allow us to more accurately replicate native tissue anisotropy. This review highlights the limitations of autologous auricular reconstruction, including donor site morbidity, technical considerations and long-term complications. Current tissue-engineered auricular constructs implanted into immune-competent animal models have been observed to undergo inflammation, fibrosis, foreign body reaction, calcification and degradation. Combining biomimetic regenerative medicine strategies will allow us to improve tissue-engineered auricular cartilage with respect to biochemical composition and functionality, as well as microstructural organization and overall shape. Creating functional and durable tissue has the potential to shift the paradigm in reconstructive surgery by obviating the need for donor sites

    Powder bed generation in integrated modelling of additive layer manufacturing of orthopaedic implants

    This paper presents an original model of powder bed generation developed within the frame of an integrated modelling approach for studying the interaction of physical mechanisms in additive layer manufacturing (ALM) of orthopaedic implants. The model is based on cellular automata (CA) approach and describes the relationship between moving particles of different sizes during deposition on a surface in three dimensions. The surface is defined by the horizontal two-dimensional CA on which particles fall and irreversibly stick to a growing deposit. The model allows for consideration of different restructuring cases when particles are allowed to rotate as often as necessary until achievement of a local minimum position. Changes in the packing density of the powder bed have been investigated numerically depending on technological parameters, such as particle size distribution, deposition rate and sequence of powder deposition. The model has been developed with the aim of merging to the finite element (FE)-based integrated model and is applicable to a different ranges of materials including metals and also non-metals

    Abstract The electronic industry has room for improvement in adopting cleaner strategies, both in production processes (often energy-intensive and polluting) and in waste management. Many small components like security tags are routinely disposed of via general waste, which could be reduced adopting biodegradable polymers. In this work, a method for selective deposition of metallic micro-tracks on polycaprolactone (PCL) for circuitry integration is presented. The polymer is biodegradable, flexible, suitable for 3D printing, and can be obtained from sustainable sources. Photoreduction of Ag ions was used to generate seeds for subsequent selective electroless copper (Cu) plating in a process that avoids common but undesirable compounds such as cyanides and palladium. Two different photopatterning methods were successfully used to achieve selective Cu plating: flood exposure with a 460 nm light-emitting diode (LED) and direct laser writing (DLW) using a 405 nm laser, achieving 47 ± 11 μm wide tracks. The deposition of uniform Cu layers on PCL substrates is demonstrated, with thicknesses of up to 14 μm and electrical conductivities of up to 2.06 × 107 S m−1, which is near the conductivity of bulk Cu (5.89 × 107 S m−1). Cu-plated interconnects were demonstrated to be fully functional for powering a 5 SMD LEDs circuit. Furthermore, DLW enabled the interconnect manufacturing on an uneven substrate. This method is flexible, selective, low-cost, vacuum-free and of minimized environmental impact, and it provides a new route towards the manufacturing of biodegradable electronics.</jats:p
