    High Ice Water Content at Low Radar Reflectivity near Deep Convection: Part II. Evaluation of Microphysical Pathways in Updraft Parcel Simulations

    The aeronautics industry has established that a threat to aircraft is posed by atmospheric conditions of substantial ice water content (IWC) where equivalent radar reflectivity (Ze) does not exceed 20-30 dBZ and supercooled water is not present; these conditions are encountered almost exclusively in the vicinity of deep convection. Part 1 (Fridlind et al., 2015) of this two-part study presents in situ measurements of such conditions sampled by Airbus in three tropical regions, commonly near 11 km and -43 C, and concludes that the measured ice particle size distributions are broadly consistent with past literature with profiling radar measurements of Z(sub e) and mean Doppler velocity obtained within monsoonal deep convection in one of the regions sampled. In all three regions, the Airbus measurements generally indicate variable IWC that often exceeds 2 gm (exp -3) with relatively uniform mass median area-equivalent diameter (MMD(sub eq) of 200-300 micrometers. Here we use a parcel model with size-resolved microphysics to investigate microphysical pathways that could lead to such conditions. Our simulations indicate that homogeneous freezing of water drops produces a much smaller ice MMD(sub eq) than observed, and occurs only in the absence of hydrometeor gravitational collection for the conditions considered. Development of a mass mode of ice aloft that overlaps with the measurements requires a substantial source of small ice particles at temperatures of about -10 C or warmer, which subsequently grow from water vapor. One conceivable source in our simulation framework is Hallett-Mossop ice production; another is abundant concentrations of heterogeneous ice freezing nuclei acting together with copious shattering of water drops upon freezing. Regardless of the production mechanism, the dominant mass modal diameter of vapor-grown ice is reduced as the ice-multiplication source strength increases and as competition for water vapor increases. Both mass and modal diameter are reduced by entrainment and by increasing aerosol concentrations. Weaker updrafts lead to greater mass and larger modal diameters of vapor-grown ice, the opposite of expectations regarding lofting of larger ice particles in stronger updrafts. While stronger updrafts do loft more dense ice particles produced primarily by raindrop freezing, we find that weaker updrafts allow the warm rain process to reduce competition for diffusional growth of the less dense ice expected to persist in convective outflow

    Occurrences of jet engine power loss and damage have been associated with flight through fully glaciated deep convection at -10 to -50 degrees Centigrade. Power loss events commonly occur during flight through radar reflectivity (Zeta (sub e)) less than 20-30 decibels relative to Zeta (dBZ - radar returns) and no more than moderate turbulence, often overlying moderate to heavy rain near the surface. During 2010-2012, Airbus carried out flight tests seeking to characterize the highest ice water content (IWC) in such low-radar-reflectivity regions of large, cold-topped storm systems in the vicinity of Cayenne, Darwin, and Santiago. Within the highest IWC regions encountered, at typical sampling elevations (circa 11 kilometers), the measured ice size distributions exhibit a notably narrow concentration of mass over area-equivalent diameters of 100-500 micrometers. Given substantial and poorly quantified measurement uncertainties, here we evaluate the consistency of the Airbus in situ measurements with ground-based profiling radar observations obtained under quasi-steady, heavy stratiform rain conditions in one of the Airbus-sampled locations. We find that profiler-observed radar reflectivities and mean Doppler velocities at Airbus sampling temperatures are generally consistent with those calculated from in situ size-distribution measurements. We also find that column simulations using the in situ size distributions as an upper boundary condition are generally consistent with observed profiles of radar reflectivity (Ze), mean Doppler velocity (MDV), and retrieved rain rate. The results of these consistency checks motivate an examination of the microphysical pathways that could be responsible for the observed size-distribution features in Ackerman et al. (2015)

    GILZ inhibits the mTORC2/AKT pathway in BCR-ABL+ cells

    The malignant phenotype of chronic myeloid leukemia (CML) is due to the abnormal tyrosine kinase activity of the BCR-ABL oncoprotein, which signals several downstream cell survival pathways, including phosphoinositide 3-kinase/AKT, signal transducer and activator of transcription 5 and extracellular signal-regulated kinase 1/2. In patients with CML, tyrosine kinase inhibitors (TKIs) are used to suppress the BCR-ABL tyrosine kinase, resulting in impressive response rates. However, resistance can occur, especially in acute-phase CML, through various mechanisms. Here, we show that the glucocorticoid-induced leucine zipper protein (GILZ) modulates imatinib and dasatinib resistance and suppresses tumor growth by inactivating the mammalian target of rapamycin complex-2 (mTORC2)/AKT signaling pathway. In mouse and human models, GILZ binds to mTORC2, but not to mTORC1, inhibiting phosphorylation of AKT (at Ser473) and activating FoxO3a-mediated transcription of the pro-apoptotic protein Bim; these results demonstrate that GILZ is a key inhibitor of the mTORC2 pathway. Furthermore, CD34+ stem cells isolated from relapsing CML patients underwent apoptosis and showed inhibition of mTORC2 after incubation with glucocorticoids and imatinib. Our findings provide new mechanistic insights into the role of mTORC2 in BCR-ABL+ cells and indicate that regulation by GILZ may influence TKI sensitivity

    Inflammation and ER stress differentially regulate STAMP2 expression and localization in adipocytes

    Background Chronic ER stress and dysfunction is a hallmark of obesity and a critical contributor to metaflammation, abnormal hormone action and altered substrate metabolism in metabolic tissues, such as liver and adipocytes. Lack of STAMP2 in lean mice induces inflammation and insulin resistance on a regular diet, and it is dysregulated in the adipose tissue of obese mice and humans. We hypothesized that the regulation of STAMP2 is disrupted by ER stress. Methods 3T3-L1 and MEF adipocytes were treated with ER stress inducers thapsigargin and tunicamycin, and inflammation inducer TNFα. The treatments effect on STAMP2 expression and enzymatic function was assessed. In addition, 3T3-L1 adipocytes and HEK cells were utilized for Stamp2 promoter activity investigation performed with luciferase and ChIP assays. Results ER stress significantly reduced both STAMP2 mRNA and protein expression in cultured adipocytes whereas TNFα had the opposite effect. Concomitant with loss of STAMP2 expression during ER stress, intracellular localization of STAMP2 was altered and total iron reductase activity was reduced. Stamp2 promoter analysis by reporter assays and chromatin immunoprecipitation, showed that induction of ER stress disrupts C/EBPα-mediated STAMP2 expression. Conclusion These data suggest a clear link between ER stress and quantitative and functional STAMP2-deficiency

    The Measured Relationship between Ice Water Content and Cloud Radar Reflectivity in Tropical Convective clouds.

    International audienceIn this paper we use unprecedented bulk measurements of ice water content (IWC) up to approximately 5 gm-3 and 95 GHz radar reflectivities (Z95) to analyze the statistical relationship between these two quantities and its variability. The unique aspect of this study is that these IWC – Z95 relationships do not use assumptions on cloud microphysics or backscattering calculations. IWCs greater than 2 gm-3 are also included for the first time in such analysis, owing to improved bulk IWC probe technology and a flight program targeting high ice water content. Using a single IWC – Z95 relationship allows for the retrieval of IWC from radar reflectivities with less than 30% bias and 40-70% rms difference. These errors can be reduced further down to 10-20% bias over the whole IWC range using the temperature variability of this relationship. IWC errors largely increase for Z95> 15-16 dBZ, due to the distortion of the IWC – Z95 relationship by non-Rayleigh scattering effects. A non-linear relationship is proposed to reduce these errors down to 20% bias and 20-35% rms differences. This non-linear relationship also outperforms the temperature-dependent IWC – Z95 relationship for convective profiles. The joint frequency distribution of IWC and temperature within and around deep tropical convective cores shows that at the -50°C ± 5°C level – the cruise altitude of many commercial jet aircraft – IWCs greater than 1.5 gm-3 were found exclusively in convective profiles

    Artificial cloud test confirms volcanic ash detection using infrared spectral imaging

    Airborne volcanic ash particles are a known hazard to aviation. Currently, there are no means available to detect ash in flight as the particles are too fine (radii<30 µm) for on-board radar detection and, even in good visibility, ash clouds are difficult or impossible to detect by eye. The economic cost and societal impact of the April/May 2010 Icelandic eruption of Eyjafjallajokull generated renewed interest in finding ways to identify airborne volcanic ¨ ash in order to keep airspace open and avoid aircraft groundings. We have designed and built a bi-spectral, fast-sampling, uncooled infrared camera device (AVOID) to examine its ability to detect volcanic ash from commercial jet aircraft at distances of more than 50 km ahead. Here we report results of an experiment conducted over the Atlantic Ocean, off the coast of France, confirming the ability of the device to detect and quantify volcanic ash in an artificial ash cloud created by dispersal of volcanic ash from a second aircraft. A third aircraft was used to measure the ash in situ using optical particle counters. The cloud was composed of very fine ash (mean radii ∼10 µm) collected from Iceland immediately after the Eyjafjallajokull eruption and had a vertical thickness of ¨ ∼200 m, a width of ∼2 km and length of between 2 and 12 km. Concentrations of ∼200 µg m−3 were identified by AVOID at distances from ∼20 km to ∼70 km. For the first time, airborne remote detection of volcanic ash has been successfully demonstrated from a long-range flight test aircraft
