    Microbial biomass, nutrient availability and nutrient uptake by wheat in two soils with organic amendments

    A 72-day greenhouse pot experiment was conducted with a sandy loam or a silt loam soil to examine the effects of farmyard manure (FYM), poultry litter (PL) and biogenic waste compost (BWC) at 10 g dw kg-1 soil on microbial biomass and activity and growth and nutrient uptake by wheat. Soil samples were collected at days 0, 14, 28, 42, 56 and 72 after planting. Growth and nutrient uptake by wheat were determined on day 72. All three amendments increased microbial biomass C, N and P, dehydrogenase activity, plant growth and nutrient uptake with a greater effect by FYM and PL than by BWC. All amendments increased microbial biomass C, N and P and enzyme activity particularly on day 0. These microbial parameters decreased after day 0 indicating microbial biomass turnover. All amendments increased plant growth and nutrient uptake. It is concluded that organicamendments can stimulate microbial growth and nutrient uptake as well as plant growth and nutrient uptake. Microbes can increase plant nutrient availability by nutrient mobilisation but also because nutrients taken up by the microbial biomass initially could become available to plants when the microbial biomass turns over as the easily available C is depleted.M.A. Malik, K.S.     Role of Values and Environmental Concerns for the Sustainable Purchasing Behavior: Evidence From Pakistan

    Sustainable purchasing is an attempt to purchase greener, healthier, and more economical items from greener, more sustainable organizations. It is based on the idea that each and every buy has shrouded human wellbeing through natural and social effects and that it is possible to decrease unfavourable effects by purchasing better items. The motive of the present study was to identify the role of values and environmental concerns for sustainable purchasing behaviour of buyers in the Pakistani green market. With the help of reliable and valid Likert type self-administered questionnaires, data was collected from the sample of the people who claim to live green lifestyles and purchase green products. Results of Smart PLS path model confirm that values and environmental concerns have an influence on sustainable purchasing behaviour in the Pakistani green market. This study provides a greater insight to improve purchaser behaviour in the green market of Pakistan and will help to increase the trust of buyers towards the green market

    Growth rhythms in sunflower (Helianthus annuus L.) in response to environmental disparity

    Sunflower hybrids maintain high level of stability in a variety of environments. Environmental variations affect and modify plant attributes like growth, development and assimilation through physio-morphicfunctions, thus modifying plant phenology. Field experiments, one each in spring and autumn, were conducted at Pir Mehr Ali Shah, Arid Agriculture University, Rawalpindi, Pakistan for two years (2007 and 2008) to document the effect of environmental variations on growth rhythms of sunflower hybrids. Four Sunflower hybrids, Alisson-RM, Parasio-24, MG-2 and S-278 were planted in randomized complete block design with four replications. The data based on physiological attributes like leaf area (LA),specific leaf area (SLA), crop growth rate (CGR) and net assimilation rate (NAR) at 10 days interval after complete emergence to 60 days after emergence (DAE) were recorded. Overall higher values of LA, SLA, CGR and NAR were recorded during spring as compared to autumn for both years. LA, CGR and NAR of all the hybrids followed a sigmoid curve pattern during both seasons; however, at the start of the season lesser values were observed during spring as compared to autumn. SLA exhibited anopposite trend and continuously decreased till the end. Sigmoid curve pattern of LA, CGR and NAR may be related to environmental variables like temperature and sunshine hours. The decrease after a peak may be due to senescence of older leaves and shifting of crop from vegetative to reproductive phase

    Seed storage protein polymorphism in ten elite rice (Oryza sativa L.) genotypes of Sindh

    Seed protein profiling is the most promising tool in determining the molecular polymorphism and genetic homology. Seed storage proteins help in cultivar identification by avoiding the external environmental influences. Electrophoretically detectable proteins in rice grains possess the potential of characterizing the germplasms by their taxonomic and evolutionary aspects. This study was aimed at exploiting the genetic variations among ten elite rice genotypes of Sindh through  electrophoretical separation of grain proteins by sodium dodecyl sulphate polyacryamide gel electrophoresis (SDSPAGE). In conclusion, the investigation revealed negligible polymorphism, with reference to the total seed protein profiles, among the rice genotypes of Sindh, Pakistan used in this study. Hence, it is highly important to include a significant number of rice genotypes to explore their existing genotypic diversity for future rice breeding programs. The SDS-PAGE in combination with 2-D electrophoresis is further suggested for documenting contrasting variations of isoforms of protein peptides.Key words: Seed storage proteins, polymorphism, Oryza sativa, Sindh, SDS-PAG

    Base catalyzed transesterification of sunflower oil biodiesel

    In this study, sunflower oil was investigated for biodiesel production. Sunflower is one of the leading oil seed crop, cultivated for the production of oil in the world. It has also been considered as an important crop for biodiesel production. Seeds for biodiesel production were procured from local farmers of Attock and Rawalpidni divisions. Electric oil expeller was used for the extraction of crude oil. Base catalyzed transesterifiction process is applied for optimum yield (80%) of biodiesel. Fuel properties of sunflower oil biodiesel were compared with American Society for Testing and Materials (ASTM). Engine efficiency of biodiesel with reference to power, efficiency and consumption of biodiesel blends (B100, B20 and B5) were determined.     In vitro regeneration of Pakistani peanut (Arachis hypogea L.) varieties using de-embryonated coteledonary explants

    Peanut (Arachis hypogaea L.) belongs to the family leguminosae and is one of the world’s largest oilseed crops. This study is the first report on peanut regeneration from Pakistan using four commercially released peanut varieties, that is, Golden, BARI-2000, BARD-479 and BARD-92. Longitudinally, halved cotyledons with removed embryos were employed as explants. Among various tested combinations of BAP and NAA, the best combination was 4 and 0.1 mg/L respectively. BARI-2000 proved to be the best responsive variety for in vitro regeneration in terms of number of shoots/explant (133.3%) and number of rooted plants/explant (124.99%). The optimized protocol, which is the first one in the country, would be used to incorporate important traits in future breeding programmes of peanut in Pakistan.Key words: Arachis hypogea, de-embryonated cotyledons, BAP, NAA, in vitro regeneration, Pakistan

    ESA SENTINEL 2 IMAGERY AND GBGEOAPP: INTEGRATED TOOLS FOR THE DEOSAI NATIONAL PARK MANAGEMENT PLAN

    Deosai plateau, in the Gilgit-Baltistan Province of Pakistan, for its average elevation of 4,114 meters, is the second highest plateau in the world after Changtang Tibetan Plateau. Two biogeographically important mountain ranges merge in Deosai: the Himalayan and Karakorum–Pamir highlands. The Deosai National Park, with its first recognition in 1993, encompasses an area of about 1620 km2, with the altitude ranging from 3500 to 5200 meters a.s.l. It is known and visited by tourists for the presence of brown bear, but a large number of species of fauna and flora leave, and can be seen during the summer season. This high-altitude ecosystem is particularly fragile and can be considered a sentinel for the effects of climate changes. Due to its geographic position and high altitude, the area of Deosai has never been studied in all its ecosystem components, producing high resolution maps. The first land cover map of Deosai with 10 meters of resolution is discussed in this study. This map has been obtained from Sentinel-2 imagery and improved through the new tool developed in this study: the GBGEOApp. This application for mobile has been done with three main ambitions: the validation of the new land cover map, its improvement with land use information, and the collection of new data in the field. On the basis of the results, the use of the GBGEOApp, as a tool for validation and increasing of environmental data collection, seems to be completely applicable involving the local technicians in a process of data sharing

    Isolated spin qubits in SiC with a high-fidelity infrared spin-to-photon interface

    The divacancies in SiC are a family of paramagnetic defects that show promise for quantum communication technologies due to their long-lived electron spin coherence and their optical addressability at near-telecom wavelengths. Nonetheless, a mechanism for high-fidelity spin-to-photon conversion, which is a crucial prerequisite for such technologies, has not yet been demonstrated. Here we demonstrate a high-fidelity spin-to-photon interface in isolated divacancies in epitaxial films of 3C-SiC and 4H-SiC. Our data show that divacancies in 4H-SiC have minimal undesirable spin-mixing, and that the optical linewidths in our current sample are already similar to those of recent remote entanglement demonstrations in other systems. Moreover, we find that 3C-SiC divacancies have millisecond Hahn-echo spin coherence time, which is among the longest measured in a naturally isotopic solid. The presence of defects with these properties in a commercial semiconductor that can be heteroepitaxially grown as a thin film on shows promise for future quantum networks based on SiC defects.Comment: 26 pages, 4 figure

    Stark Tuning and Electrical Charge State Control of Single Divacancies in Silicon Carbide

    Neutrally charged divacancies in silicon carbide (SiC) are paramagnetic color centers whose long coherence times and near-telecom operating wavelengths make them promising for scalable quantum communication technologies compatible with existing fiber optic networks. However, local strain inhomogeneity can randomly perturb their optical transition frequencies, which degrades the indistinguishability of photons emitted from separate defects, and hinders their coupling to optical cavities. Here we show that electric fields can be used to tune the optical transition frequencies of single neutral divacancy defects in 4H-SiC over a range of several GHz via the DC Stark effect. The same technique can also control the charge state of the defect on microsecond timescales, which we use to stabilize unstable or non-neutral divacancies into their neutral charge state. Using fluorescence-based charge state detection, we show both 975 nm and 1130 nm excitation can prepare its neutral charge state with near unity efficiency.Comment: 12 pages, 4 figure

    Detection and diversity of a putative novel heterogeneous polymorphic proline-glycine repeat (Pgr) protein in the footrot pathogen Dichelobacter nodosus

    Dichelobacter nodosus, a Gram-negative anaerobic bacterium, is the essential causative agent of footrot in sheep. Currently, depending on the clinical presentation in the field, footrot is described as benign or virulent; D. nodosus strains have also been classified as benign or virulent, but this designation is not always consistent with clinical disease. The aim of this study was to determine the diversity of the pgr gene, which encodes a putative proline-glycine repeat protein (Pgr). The pgr gene was present in all 100 isolates of D. nodosus that were examined and, based on sequence analysis had two variants, pgrA and pgrB. In pgrA, there were two coding tandem repeat regions, R1 and R2: different strains had variable numbers of repeats within these regions. The R1 and R2 were absent from pgrB. Both variants were present in strains from Australia, Sweden and the UK, however, only pgrB was detected in isolates from Western Australia. The pgrA gene was detected in D. nodosus from tissue samples from two flocks in the UK with virulent footrot and only pgrB from a flock with no virulent or benign footrot for >10 years. Bioinformatic analysis of the putative PgrA protein indicated that it contained a collagen-like cell surface anchor motif. These results suggest that the pgr gene may be a useful molecular marker for epidemiological studies
