    Tracking the timing of Neotethyan oceanic slab break-off: Geochronology and geochemistry of the quartz diorite porphyries, NE Turkey

    The initiation of the break-off of the northern branch of the Neotethyan oceanic lithosphere is an important but poorly understood event in the geology of the Sakarya Zone (SZ) in northeastern Turkey. Although it is well-known that Latest Cretaceous intrusives (~70 Ma) and early Eocene adakitic magmatic rocks are present in the eastern SZ, the outcrops of the early Eocene non-adakitic rocks are very limited, and their tectono-magmatic evolution has not been studied. We describe a small outcrop of non-adakitic quartz diorite porphyry in the Kov area of the Gümüşhane region in northeastern Turkey. The genesis of these porphyries is significant in evaluating the syn- to post-collision-related magmatism. The LA-ICP-MS zircon U-Pb dating revealed that the Kov quartz diorite porphyries (KQDP) formed at ca. 50 Ma, coeval with adakitic rocks, and ~20 Myr later than the slab roll-back-related intrusive rocks. The KQDPs are calc-alkaline in composition and enriched in large ion lithophile elements (LILEs) and light rare earth elements (LREEs) and depleted in high field strength elements (HFSEs; e.g., Nb, Ta, Ti), with significant negative anomalies of Nb, Ta, and Ti but positive anomalies of Th, U, and Pb. Isotopic compositions of the samples show limited range of variation and slight enrichment of 87Sr/86Sr(t) (0.70489 to 0.70555), εNd(t) (−1.4 to −1.2) with TDM of 1.11 to 1.61 Ga. Pb isotopic ratios of the samples point to an enriched mantle source. They were likely crystallized from the melt that originated from an EM2-type spinel-facies subcontinental lithospheric mantle (SCLM), followed by the fractionation with insignificant crustal assimilation. The SCLM was metasomatically enriched, and the metasomatic agent was likely H2O-rich fluids rather than sediments released from subducting oceanic crust during the Late Cretaceous closure of the Neotethyan oceanic lithosphere. In conjunction with the geological background and previous data, we propose that the generation of the KQDPs resulted from a slab break-off event that caused ascending or infiltration of hot asthenosphere, triggering mantle melting. Such sporadic occurrences of the KQDPs, with coeval adakitic rocks in the SZ, are likely associated with the onset of extensional tectonics due to the earlier stage of slab break-off along the region during the early Eocene period.publishe

    Targeting quiescent leukemic stem cells using second generation autophagy inhibitors

    In chronic myeloid leukemia (CML), tyrosine kinase inhibitor (TKI) treatment induces autophagy that promotes survival and TKI-resistance in leukemic stem cells (LSCs). In clinical studies hydroxychloroquine (HCQ), the only clinically approved autophagy inhibitor, does not consistently inhibit autophagy in cancer patients, so more potent autophagy inhibitors are needed. We generated a murine model of CML in which autophagic flux can be measured in bone marrow-located LSCs. In parallel, we use cell division tracing, phenotyping of primary CML cells, and a robust xenotransplantation model of human CML, to investigate the effect of Lys05, a highly potent lysosomotropic agent, and PIK-III, a selective inhibitor of VPS34, on the survival and function of LSCs. We demonstrate that long-term haematopoietic stem cells (LT-HSCs: Lin−Sca-1+c-kit+CD48−CD150+) isolated from leukemic mice have higher basal autophagy levels compared with non-leukemic LT-HSCs and more mature leukemic cells. Additionally, we present that while HCQ is ineffective, Lys05-mediated autophagy inhibition reduces LSCs quiescence and drives myeloid cell expansion. Furthermore, Lys05 and PIK-III reduced the number of primary CML LSCs and target xenografted LSCs when used in combination with TKI treatment, providing a strong rationale for clinical use of second generation autophagy inhibitors as a novel treatment for CML patients with LSC persistence

    A comprehensive analysis of the genetic diversity and environmental adaptability in worldwide Merino and Merino-derived sheep breeds

    BACKGROUND: To enhance and extend the knowledge about the global historical and phylogenetic relationships between Merino and Merino-derived breeds, 19 populations were genotyped with the OvineSNP50 BeadChip specifically for this study, while an additional 23 populations from the publicly available genotypes were retrieved. Three complementary statistical tests, Rsb (extended haplotype homozygosity between-populations), XP-EHH (cross-population extended haplotype homozygosity), and runs of homozygosity (ROH) islands were applied to identify genomic variants with potential impact on the adaptability of Merino genetic type in two contrasting climate zones. RESULTS: The results indicate that a large part of the Merino's genetic relatedness and admixture patterns are explained by their genetic background and/or geographic origin, followed by local admixture. Multi-dimensional scaling, Neighbor-Net, Admixture, and TREEMIX analyses consistently provided evidence of the role of Australian, Rambouillet and German strains in the extensive gene introgression into the other Merino and Merino-derived breeds. The close relationship between Iberian Merinos and other South-western European breeds is consistent with the Iberian origin of the Merino genetic type, with traces from previous contributions of other Mediterranean stocks. Using Rsb and XP-EHH approaches, signatures of selection were detected spanning four genomic regions located on Ovis aries chromosomes (OAR) 1, 6 and 16, whereas two genomic regions on OAR6, that partially overlapped with the previous ones, were highlighted by ROH islands. Overall, the three approaches identified 106 candidate genes putatively under selection. Among them, genes related to immune response were identified via the gene interaction network. In addition, several candidate genes were found, such as LEKR1, LCORL, GHR, RBPJ, BMPR1B, PPARGC1A, and PRKAA1, related to morphological, growth and reproductive traits, adaptive thermogenesis, and hypoxia responses. CONCLUSIONS: To the best of our knowledge, this is the first comprehensive dataset that includes most of the Merino and Merino-derived sheep breeds raised in different regions of the world. The results provide an in-depth picture of the genetic makeup of the current Merino and Merino-derived breeds, highlighting the possible selection pressures associated with the combined effect of anthropic and environmental factors. The study underlines the importance of Merino genetic types as invaluable resources of possible adaptive diversity in the context of the occurring climate changes

    The usability of recycled carbon fibres in short fibre thermoplastics: interfacial properties

    The objective of this study was to investigate the feasibility of combining discontinuous recycled carbon fibres with polypropylene, to produce a low-cost, high specific stiffness material for high-volume applications. The inherent low affinity of carbon fibre and polypropylene motivated a detailed study of the surface characteristics of carbon fibre and interfacial behaviour between the two materials, using the microbond test. The effects of removing the sizing from the fibres, as well as introducing a maleic anhydride-grafted polypropylene coupling agent, were extensively investigated. Polypropylene was found to degrade when prepared under atmospheric conditions; therefore, it was necessary to form droplets under nitrogen. Removal of the sizing from the fibre using pyrolysis and solvolysis techniques altered the surface morphology of the fibre and increased the interfacial shear strength (IFSS) by 4 and 33 %, respectively. A more significant improvement in the fibre–matrix adhesion was achieved by adding a maleic anhydride coupling agent at 2 wt%, which increased the IFSS by 320 %

    Transcriptomic profiles of muscle, heart, and spleen in reaction to circadian heat stress in Ethiopian highland and lowland male chicken

    Temperature stress impacts both welfare and productivity of livestock. Global warming is expected to increase the impact, especially in tropical areas. We investigated the biological mechanisms regulated by temperature stress due to the circadian temperature cycle in temperature adapted and non-adapted chicken under tropical conditions. We studied transcriptome profiles of heart, breast muscle, and spleen tissues of Ethiopian lowland chicken adapted to high circadian temperatures and non-adapted Ethiopian highland chicken under lowland conditions at three points during the day: morning, noon, and evening. Functional annotations and network analyses of genes differentially expressed among the time points of the day indicate major differences in the reactions of the tissues to increasing and decreasing temperatures, and also the two chickens lines differ. However, epigenetic changes of chromatin methylation and histone (de)acetylation seemed to be central regulatory mechanisms in all tissues in both chicken lines. Finally, all tissues showed differentially expressed genes between morning and evening times indicating biological mechanisms that need to change during the night to reach morning levels again the next day.</p

    Developing worksheet based on science process skills: Factors affecting solubility

    The purpose of this study is to develop a worksheet about the factors affecting solubility, which could be useful for the prospective science teachers (PST) to remind and regain their science process skills (SPS). The pilot study of the WS was carried out with 32 first grade PST during the 2007-2008 academic year in the education department at Giresun University, Turkey. Action research methodology was used in this study. Thoughts from experts were received during the development of the worksheet. The study had some limitations in providing concrete evidence as to how the WS based on SPS effects the PST SPS, since the paper is a suggestion.
