    La actividad física puede modificar la composición corporal y la mineralización ósea. Se compararon ambas variables en gimnastas femeninas de competición y controles apareados por edad y sexo que realizaban gimnasia recreativa (n = 12 en cada grupo; edades 9 a 14 años). La composición corporal se evaluó por métodos antropométricos y densitometría de rayos X (DXA). El contenido mineral y la densidad mineral óseas se midieron por DXA en cuerpo entero y columna lumbar.     Impacts of the Mount Pinatubo eruption on ENSO in the GEOS seasonal-to-subseasonal forecasting system

    The eruption of Mount Pinatubo in June 1991 introduced a perturbation of the Earth's global energy budget by increasing the stratospheric aerosol loading by an order of magnitude, with effects on the global climate. In this presentation we analyze the effects of the Mt. Pinatubo eruption on the seasonal forecast performed with Goddard Earth Observing System Seasonal-to Subseasonal (GEOS-S2S) system, an Earth System Model that includes an interactive ocean and a bulk aerosol model coupled to radiation. We performed 10-member ensembles for the year after the eruption (June 1991-May 1992) at ~0.5 horizontal resolution, with and without the inclusion of the Mt. Pinatubo eruption. In GEOS-S2S, the eruption leads to ta strengthening of El Nino peaking in January 1992. The strengthening is mainly due to the weakening of the trade winds, which is caused by a attening of the temperature gradient across the Pacic due to a differential response to the volcanic forcing between the central and eastern Pacic (ocean-dynamical thermostat). This response largely depends on the assumed size for the volcanic aerosols. Indeed, we performed simulations assuming a volcanic aerosol effective radius of 0.35 m (similar to tropospheric aerosol, and the default in GEOS) and 0.6 m (closer to observations of volcanic aerosol from Pinatubo-sized eruptions). We nd that in the latter case the tropical radiative forcing is lower, since smaller aerosols scatter shortwave radiation more eciently than larger ones. Accordingly, the impact on ENSO is not statistically signicant when a larger and more realistic particle radius is assumed

    Optical constants of magnetron-sputtered magnesium films in the 25-1300 eV energy range

    The transmittance of dc magnetron-sputtered Mg thin films was measured in the 25-1300 eV spectral range. Freestanding Mg films protected with Al layers were characterized ex situ. Transmittance measurements were used to obtain the extinction coefficient k of Mg films. The obtained k values along with the data available in the literature, and with interpolations and extrapolations for the rest of the spectrum, were used to obtain the real part of the index of refraction n by the Kramers-Krönig analysis. Sum-rule tests indicated a good consistency of the data. © 2010 American Institute of Physics.Peer Reviewe

    Evidence that low doses of Taxol enhance the functional transactivatory properties of p53 on p21 waf promoter in MCF-7 breast cancer cells

    AbstractIn the present study, we evidence how in breast cancer cells low doses of Taxol for 18h determined the upregulation of p53 and p21 waf expression concomitantly with a decrease of the anti-apoptotic Bcl-2. P53 and its gene product, the mdm2 protein, in treated cells exhibits a prevalent nuclear compartmentalization, thus potentiating p53 transactivatory properties. Indeed, the most important finding of this study consists with the evidence that Taxol at lower concentrations is able to produce the activation of p21 promoter via p53. Prolonged exposure of MCF-7 cells to Taxol (48h) resulted in an increased co-association between p21 and PCNA compared to control and this well fits with the simultaneous block of cell cycle into the G2/M phase

    We search for signals of Weak Annihilation in inclusive semileptonic D decays. We consider both the widths and the lepton energy moments, which are quite sensitive probes.     Biogenesis of the mitochondrial phosphate carrier

    The mitochondrial phosphate carrier (PiC) is a member of the family of inner-membrane carrier proteins which are generally synthesized without a cleavable presequence. Surprisingly, the cDNA sequences of bovine and rat PiC suggested the existence of an amino-terminal extension sequence in the precursor of PiC. By expressing PiC in vitro, we found that PiC is indeed synthesized as a larger precursor. This precursor was imported and proteolytically processed by mitochondria, whereby the correct amino-terminus of the mature protein was generated. Import of PiC showed the characteristics of mitochondrial protein uptake, such as dependence on ATP and a membrane potential and involvement of contact sites between mitochondrial outer and inner membranes. The precursor imported in vitro was correctly assembled into the functional form, demonstrating that the authentic import and assembly pathway of PiC was reconstituted when starting with the presequence-carrying precursor. These results are discussed in connection with the recently postulated role of PiC as an import receptor located in the outer membrane

    Introduction Progesterone Receptor (PR) positivity is associated with a good prognosis and better response to breast cancer treatment. Conversely, cyclin D1 (CD1) is retained a marker of poor outcome since it has been associated with breast cancer metastasis in clinical studies. Material and methods 17-Hydroxyprogesterone (OHPg) was from Sigma-Aldrich. Antibodies and Protein A/GPLUS-Agarose were from Santa Cruz Biotechnology. T47-D, MCF-7 and MDA-MB-231 human breast cancer cells from the American Type Culture Collection; Total real-time RT-PCR assay; Western blotting and immunoprecipitation; Transfections and luciferase assays; Lipid-Mediated Transfection of siRNA Duplexes; Chromatin immunoprecipitation (ChIP) assays and realtime ChIP; Wound-healing assays; Transmigration assays; Cell invasion assay; Phalloidin staining. Results and discussions Herein we provide evidences that OHPg through PR-B isoform, reduces motility and invasion of T47-D and MCF-7 breast cancer cells, by targeting the cytoplasmic CD1. Specifically, OHPg reduces CD1 expression through a transcriptional mechanism due to the occupancy of CD1 promoter at a canonical half progesterone responsive element by PR-B. This allows the recruitment of HDAC1 influencing a less permissive chromatin conformation for gene transcription and the release of RNA Pol II. CD1 has an active role in the control of cell migration and metastasis through the interaction with key components of focal adhesion such as Paxillin (Pxn). In untreated T47-D and MCF-7 cells a specific co-immunoprecipitation of endogenous cytoplasmic CD1 with Pxn was detected. Interestingly, OHPg exposure reduced the interaction between these proteins although total Pxn expression was substantially unaffected. Moreover a concomitant reduction of p-Pxn levels was observed and these effects were required for OHPg/PR-B dependent delay in cell invasion, as evidenced by assays carried out with the phoshomimetic mutants of Pxn. Conclusion Collectively these findings support the importance of PR-B expression in breast cancer cells behaviour, suggesting potentiating of PR-B signalling as a prospective useful strategy to restrict breast tumour cells invasion and metastasis
