    Magnetic and relaxation properties of vanadium(iv) complexes: an integrated H-1 relaxometric, EPR and computational study

    We report a detailed study of the magnetic and relaxation properties of a series of oxovanadium(IV) complexes comprising the aqua ion [VO(H2O)5]2+ and [VO(ox)2]2- (ox = oxalate), [VO(nta)]- (nta = nitrilotriacetate), [VO(dtpa)] 3- (dtpa = diethylenetriaminepentaacetate) and [VO(acac)2] (acac = acetylacetonato) in solution. The complexes were characterized using continuous wave (X-band) and pulsed (Q-band) EPR measurements and 1H nuclear magnetic relaxation dispersion (NMRD) studies in the 0.01-120 MHz 1H Larmor frequency range. The 51V A-tensor parameters obtained from the analysis of EPR spectra are in good agreement with those obtained using theoretical calculations at the DFT and coupled-cluster levels (DLPNO-CCSD), while g-tensors were obtained with CASSCF/NEVPT2 calculations. EPR measurements reveal significant differences in the electronic Te1 and Te m relaxation times, with [VO(acac)2] showing a markedly different behaviour due to the trans coordination geometry. The NMRD profiles measured at different temperatures have contributions from both the outer- and inner-sphere mechanisms, with the latter showing contributions from the dipolar and scalar mechanisms. The rotational correlation times ( tR) obtained from the fitting of NMRD and EPR data are in good mutual agreement. The scalar mechanism depends on the hyperfine coupling constants of the coordinated water molecule Haiso, which were obtained from the fitting of the NMRD profiles and DFT calculations. Finally, the analysis of the data provided information on the exchange rate of coordinated water molecules, which display mean residence times of similar to 7-17 mu s at 298 K

    Effect of surface roughness and sterilization on bacterial adherence to ultra-high molecular weight polyethylene

    AbstractSterilization with ethylene oxide (EO) and gas plasma (GP) are well-known methods applied to ultra-high molecular weight polyethylene (UHMWPE) surfaces in the belief that they prevent major material changes caused by gamma irradiation. However, the influence of these surface sterilization methods on bacterial adherence to UHMWPE is unknown. UHMWPE samples with various degrees of roughness (0.3, 0.8 and 2.0 µm) were sterilized with either GP or EO. The variations in hydrophobicity, surface free energy and surface functional groups were investigated before and after sterilization. Sterilized samples were incubated with either Staphylococcus aureus or Staphylococcus epidermidis in order to study bacterial adherence to these materials. Fewer bacteria adhered to UHMWPE after sterilization with EO than after sterilization with GP, especially to the smoothest surfaces. No changes in chemical composition of the UHMWPE surface due to sterilization were observed using X-ray photoemission spectroscopy analysis. The decreased bacterial adherence to UHMWPE found at the smoothest surfaces after sterilization with EO was not directly related to changes in chemical composition. Increased bacterial adherence to rougher surfaces was associated with increased polar surface energy of EO-sterilized surfaces

    Development of a Retrospective Measure of Child Abuse for Heterosexual and LGBTQ Mexican Adults

    Child abuse is a phenomenon that affects a large proportion of the world’s population and has important effects on their mental health. Although several instruments exist to measure it, they present some difficulties that require improvement. The present study started from an instrument previously developed for Mexican population (Esparza-Del Villar et al., Journal of Interpersonal Violence, 2020) to construct a new retrospective measure of abuse: the Child Abuse Scale for Adults (EAIA, Spanish initials). We worked with a total sample of 810 individuals, consisting mainly of women (> 70%) and young adults (mean age around 23 years). We conducted both traditional (exploratory and confirmatory factor analysis) and advanced (exploratory graph analysis, item response theory) procedures to obtain a short self-report measure of child abuse experiences. The EAIA evidenced a clear factor structure (CFI = .96, RSMEA = .06), as well as adequate reliability for its three subscales: sexual (ω = .91), physical (ω = .88) and emotional abuse (ω = .93). In addition, it showed partial scalar invariance between sexes. At the item level, it was observed that the items of the sexual and physical abuse subscales had greater informative capacity at high levels of maltreatment, while the items of the emotional abuse subscale showed better psychometric quality at average levels of the construct. Finally, associations were found in the expected direction between the three subscales of the EAIA and a set of psychopathological variables (depression, anxiety and suicidal ideation). In conclusion, the EAIA constitutes a promising alternative to retrospectively measure child abuse in the Mexican adult population

    A noninterventional study evaluating the effectiveness of rotigotine and levodopa combination therapy in younger versus older patients with Parkinson\u27s disease

    Background: PD0013 was a 6-month non-interventional study in clinical-practice comparing effectiveness and tolerability of rotigotine+levodopa in younger (<70years) vs. older (≥70years) Parkinson’s disease (PD) patients.Methods: Patients previously received levodopa for ≥6-months as monotherapy or in combination with another dopamine-agonist (DA). Primary variable: Unified PD Rating Scale (UPDRS) Part-II change from baseline to end-of-observation-period (EOP).Results: 91 younger/99 older patients started rotigotine; 68 younger/62 older patients completed the study. Most switched from levodopa+another DA. Addition of rotigotine as first DA was more common in older patients (20.2% vs.15.4%). Mean\ub1SD rotigotine-exposure: 6.1\ub13.4mg/24h younger vs. 4.9\ub12.4mg/24h older. Eleven patients changed levodopa dose during the study.At EOP, improvement in mean UPDRS-II was greater in younger patients (p=0.0289). UPDRS-II responder-rate (≥20% decrease in UPDRS-II score) was higher in younger patients (42.3% vs. 25.9%). Improvement across age-groups was similar on PD Sleep Scale-2 and Clinical Global Impressions-Improvement Scale. Adverse-drug-reactions (ADRs), and discontinuations because of ADRs, were more common among older patients. There were no new safety-signals.Conclusions: Despite low rotigotine doses, when added to levodopa or switched from levodopa+another DA, rotigotine led to greater improvement in UPDRS-II in younger patients (<70years). Assessment of individual patient data revealed clinically-meaningful improvements in UPDRS-II in both age-groups

    Macroscopic brain gray matter staining: historical protocol overview and neuroanatomy learning applications in second-year medical students

    Macroscopic staining in anatomical samples of the central nervous system is a technique that has been used for decades to achieve better differentiation of multiple gray matter structures, such as the cortex, basal ganglia, and cerebellar nuclei. Staining methods are based on using the different components of the brain, mainly the lipids present in the white matter. These techniques have been progressively forgotten while computer renderings are increasing; however, as a primary exposure to surgical anatomy, stained brain specimens are considered a helpful tool. We aim to summarize different staining techniques, their principles, and their current applications for neuroanatomy learning purposes. In total, four gray matter staining protocol descriptions (Mulligan's, Roberts's, Alston's, and Prussian Blue) were performed, as well as Likert scale surveys of second-year medical students about their perceptions of the stained sections. The results showed that the different macroscopic stains for brain tissue are based on lipid and reactant interactions, intending to increase the white matter (WM) and gray matter (GM) contrast. The search also showed that most staining protocols would take 2 days to develop. Efficient preservation options include submerging the sections in formaldehyde solutions, formaldehyde-free solutions, ethanol, or applying plastination techniques. Based on the student's perspective, the stained slices seem to be a valuable alternative to facilitate the study and identification of the basal ganglia and their relationships with the white matter (from 51.2 to 72% based on the Likert scale) compared with the non-stained sections. In conclusion, macroscopic staining of brain tissue continues to be a valuable tool for comprehensively studying the brain. Further research is needed to determine the efficacy of stained specimens as teaching tools

    Exposure to Bisphenol A and Phthalates during Pregnancy and Ultrasound Measures of Fetal Growth in the INMA-Sabadell Cohort

    Background: Prenatal exposure to bisphenol A (BPA) and phthalates may affect fetal growth; however, previous findings are inconsistent and based on few studies. Objectives: We assessed whether prenatal exposure to BPA and phthalates was associated with fetal growth in a Spanish birth cohort of 488 mother–child pairs. Methods: We measured BPA and eight phthalates [four di(2-ethylhexyl) phthalate metabolites (DEHPm), mono-benzyl phthalate (MBzP), and three low-molecular-weight phthalate metabolites (LMWPm)] in two spot-urine samples collected during the first and third trimester of pregnancy. We estimated growth curves for femur length (FL), head circumference (HC), abdominal circumference (AC), biparietal diameter (BPD), and estimated fetal weight (EFW) during pregnancy (weeks 12–20 and 20–34), and for birth weight, birth length, head circumference at birth, and placental weight. Results: Overall, results did not support associations of exposure to BPA or DEHPm during pregnancy with fetal growth parameters. Prenatal MBzP exposure was positively associated with FL at 20–34 weeks, resulting in an increase of 3.70% of the average FL (95% CI: 0.75, 6.63%) per doubling of MBzP concentration. MBzP was positively associated with birth weight among boys (48 g; 95% CI: 6, 90) but not in girls (–27 g; 95% CI: –79, 25) (interaction p-value = 0.04). The LMWPm mono-n-butyl phthalate (MnBP) was negatively associated with HC at 12–20 pregnancy weeks [–4.88% of HC average (95% CI: –8.36, –1.36%)]. Conclusions: This study, one of the first to combine repeat exposure biomarker measurements and multiple growth measures during pregnancy, finds little evidence of associations of BPA or phthalate exposures with fetal growth. Phthalate metabolites MBzP and MnBP were associated with some fetal growth parameters, but these findings require replication

    A phase II dose-escalation trial of perioperative desmopressin (1-desamino-8-d-arginine vasopressin) in breast cancer patients

    Desmopressin (dDAVP) is a well-known peptide analog of the antidiuretic hormone vasopressin, used to prevent excessive bleeding during surgical procedures. dDAVP increases hemostatic mediators, such as the von Willebrand factor (vWF), recently considered a key element in resistance to metastasis. Studies in mouse models and veterinary trials in dogs with locally-advanced mammary tumors demonstrated that high doses of perioperative dDAVP inhibited lymph node and early blood-borne metastasis and significantly prolonged survival. We conducted a phase II dose-escalation trial in patients with breast cancer, administering a lyophilized formulation of dDAVP by intravenous infusion in saline, 30–60 min before and 24 h after surgical resection. Primary endpoints were safety and tolerability, as well as selection of the best dose for cancer surgery. Secondary endpoints included surgical bleeding, plasma levels of vWF, and circulating tumor cells (CTCs) as measured by quantitative PCR of cytokeratin-19 transcripts. Only 2 of a total of 20 patients experienced reversible adverse events, including hyponatremia (grade 4) and hypersensitivity reaction (grade 2). Reactions were adequately managed by slowing the infusion rate. A reduced intraoperative bleeding was noted with increasing doses of dDAVP. Treatment was associated with higher vWF plasma levels and a postoperative drop in CTC counts. At the highest dose level evaluated (2 μg/kg) dDAVP appeared safe when administered in two slow infusions of 1 μg/kg, before and after surgery. Clinical trials to establish the effectiveness of adjunctive perioperative dDAVP therapy are warranted. This trial is registered on www.clinicaltrials.gov (NCT01606072).Fil: Weinberg, Ruth S..
