    Temperature-sensitive Tien Shan tree ring chronologies show multi-centennial growth trends

    Two millennia-length juniper ring width chronologies, processed to preserve multi-centennial growth trends, are presented for the Alai Range of the western Tien Shan in Kirghizia. The chronologies average the information from seven near-timberline sampling sites, and likely reflect summer temperature variation. For comparison, chronologies are also built using standard dendrochronological techniques. We briefly discuss some qualities of these "inter-decadal” records, and show the low frequency components removed by the standardization process include a long-term negative trend in the first half of the last millennium and a long-term positive trend since about AD 1800. The multi-centennial scale Alai Range chronologies, where these trends are retained, are both systematically biased (but in an opposite sense) in their low frequency domains. Nevertheless, they represent the best constraints and estimates of long-term summer temperature variation, and reflect the Medieval Warm Period, the Little Ice Age, and a period of warming since about the middle of the nineteenth centur

    Vision system and electronic tongue application to detect coffee adulteration with barley

    Time consuming and expensive methods have been applied for detection of coffee adulteration based on the literature. In the present work, an optical method (vision system) and the application of an electronic tongue is proposed to reveal the addition of barley in different proportion to coffee in ground and brewed forms. In a range of 1 to 80% (w/w) Robusta coffee was blended with roasted barley. Principal Component Analysis (PCA) accomplished on vision system image data showed a good discrimination of the adulterated samples. The results of Polar Qualification System (PQS) data reduction method revealed even small differences in the right barley content order by point method approach. With Partial Least Squares (PLS) regression the amount of barley in Robusta was predicted with high R2 (0.996) and relatively low RMSEP (∼2%) values in case of vision system data processing. Considering electronic tongue measurements, PCA results showed a good discrimination of samples with higher barley concentration. Misclassification was found in the low concentrated area by Lienar Discriminant Analgsis (LDA). To obtain an accurate model for barley content prediction in coffee, the most sensitive sensor signals were used to apply PLS regression successfully (R2=0.97, RMSEP=3.99% (w/w))

    Efficient Identification of HIV Serodiscordant Couples by Existing HIV Testing Programs in South Brazil.

    ObjectiveTo examine the feasibility of identifying HIV negative at risk individuals in HIV serodiscordant couples, during voluntary HIV testing in South Brazil.MethodsWe surveyed HIV testers at 4 public testing sites in Rio Grande do Sul. We obtained information on risk behaviors and sexual partnerships. HIV testing and testing for recent infection were performed; HIV prevalence and risk behaviors were assessed among subjects who reported having a steady partner who was HIV positive (serodiscordant group) and compared with the general testing population.ResultsAmong 3100 patients, 490 (15.8%) reported being in a steady relationship with an HIV positive partner. New HIV infections were diagnosed in 23% of the serodiscordant group (vs. 13% in the general population, p = 0.01); among newly positive subjects, recent HIV infections were more frequent (23/86, 26.7%) among testers with positive partners than among the general testing group (52/334; 15.6%; p = 0.016). Less than half of the serodiscordant testers reported having used a condom during the last sexual intercourse with their HIV-positive partner. Participants with inconsistent condom use with steady partner were four times more likely to test positive for HIV compared to those who reported always using condoms with the steady partner (OR: 4.2; 95% CI: 2.3 to 7.5).ConclusionIt is highly feasible to identify large numbers of HIV susceptible individuals who are in HIV serodiscordant relationships in South Brazil testing sites. Condom use within HIV serodiscordant couples is low in this setting, suggesting urgent need for biomedical prevention strategies to reduce HIV transmission

    Lactate Dehydrogenase-Elevating Virus Induces Systemic Lymphocyte Activation via TLR7-Dependent IFNα Responses by Plasmacytoid Dendritic Cells

    BACKGROUND:Lactate dehydrogenase-elevating virus (LDV) is a natural infectious agent of mice. Like several other viruses, LDV causes widespread and very rapid but transient activation of both B cells and T cells in lymphoid tissues and the blood. The mechanism of this activation has not been fully described and is the focus of the current studies. PRINCIPAL FINDINGS:A known inducer of early lymphocyte activation is IFNalpha, a cytokine strongly induced by LDV infection. Neutralization of IFNalpha in the plasma from infected mice ablated its ability to activate lymphocytes in vitro. Since the primary source of virus-induced IFNalpha in vivo is often plasmacytoid dendritic cells (pDC's), we depleted these cells prior to LDV infection and tested for lymphocyte activation. Depletion of pDC's in vivo eradicated both the LDV-induced IFNalpha response and lymphocyte activation. A primary receptor in pDC's for single stranded RNA viruses such as LDV is the toll-like receptor 7 (TLR7) pattern recognition receptor. Infection of TLR7-knockout mice revealed that both the IFNalpha response and lymphocyte activation were dependent on TLR7 signaling in vivo. Interestingly, virus levels in both TLR7 knockout mice and pDC-depleted mice were indistinguishable from controls indicating that LDV is largely resistant to the systemic IFNalpha response. CONCLUSION:Results indicate that LDV-induced activation of lymphocytes is due to recognition of LDV nucleic acid by TLR7 pattern recognition receptors in pDC's that respond with a lymphocyte-inducing IFNalpha response

    The role of smoking and body mass index in mortality risk assessment for geriatric hip fracture patients

    Background Smoking, obesity, and being below a healthy body weight are known to increase all-cause mortality rates and are considered modifiable risk factors. The purpose of this study is to assess whether adding these risk factors to a validated geriatric inpatient mortality risk tool will improve the predictive capacity for hip fracture patients. We hypothesize that the predictive capacity of the Score for Trauma Triage in the Geriatric and Middle-Aged (STTGMA) tool will improve. Methodology Between October 2014 and August 2021, 2,421 patients \u3e55-years-old treated for hip fractures caused by low-energy mechanisms were analyzed for demographics, injury details, hospital quality measures, and mortality.
