El tratamiento con relaxina estimula la diferenciación de células madre mesenquimales en osteoblastos
Resumen Antecedentes/objetivo: Varios estudios han determinado que la relaxina estimula la diferenciación y regula la actividad de los osteoclastos maduros, pero se sabe poco sobre su efecto en la diferenciación de las células mesenquimales hacia el linaje osteogénico. Por lo tanto, este estudio tuvo como objetivo determinar el efecto de la relaxina sobre la proliferación y diferenciación del linaje osteoblástico de células mesenquimales derivadas de pulpa dental humana (hDPSC). Materiales y métodos: En este estudio in vitro, las hDPSC fueron caracterizadas y tratadas con relaxina a diferentes dosis (10e80 ng/ml) y tiempos (1e21 días). La morfología se evaluó mediante microscopía y la proliferación mediante un ensayo de resazurina. La diferenciación osteoblástica se evaluó mediante tinción con rojo alizarina, marcaje con fosfatasa alcalina (ALP) y cambios en la expresión de los genes de diferenciación osteoblástica RUNX2 y BMP2. Resultados: El tratamiento con relaxina no indujo cambios en la proliferación o viabilidad de las hDPSCs; sin embargo, se observaron células más grandes y una mayor prolongación citoplasmática. El tratamiento con relaxina (20 y 80 ng/ml) aumentó significativamente la formación de nódulos calcificados en los días 14 y 21. Las señales citoquímicas para la expresión de los genes ALP, RUNX2 y BMP2 aumentaron significativamente (P < 0,05) con el tratamiento con relaxina.Abstract Background/purpose: Several studies have determined that relaxin stimulates differentiation and regulates the activity of mature osteoclasts, but little is known about its effect on the differentiation of mesenchymal cells towards the osteogenic lineage. Therefore, this study aimed to determine the effect of relaxin on the proliferation and differentiation of the osteoblastic lineage of mesenchymal cells derived from human dental pulp (hDPSC). Materials and methods: In this in vitro study, hDPSC were characterized and treated with relaxin at different doses (10e80 ng/ml) and times (1e21 days). Morphology was assessed by microscopy, and proliferation was assessed using a resazurin assay. Results: Relaxin treatment did not induce changes in the proliferation or viability of hDPSCs; however, larger cells and increased cytoplasmic prolongation were observed. Relaxin treatment (20 and 80 ng/ml) significantly increased calcified nodule formation on days 14 and 21. Photocatalytic activity of nitrogen-doped and undoped titanium dioxide sputtered thin films
In the present work titanium dioxide (TiO2) thin films were grown by d.c. reactive magnetron sputtering process, systematically varying the Ar/O2 ratio in the gas mixture, in order to study the influence of the oxygen partial pressure on the crystallographic structure and photocatalytic activity of the TiO2 thin films. After the sputtering process the TiO2 coatings were nitrided in a microwave (f= 2.45 GHz) Electron Cyclotron Resonance (ECR) plasma discharge in pure nitrogen, to compare the photocatalytic activity of undoped and nitrogen-doped TiO2 thin films. The crystal structure of the TiO2 grown samples was studied by x-ray diffraction (XRD) and the presence of the anatase phase in these films were corroborated by Raman spectroscopy. On the other hand, X-ray photoelectron spectroscopy (XPS) measurements carried out in the nitrogen-doped TiO2 samples, showed that the nitrogen was incorporated to the films with an average concentration of 18 at% of N. The UV-Vis optical spectroscopy allowed calculating the band gap. A narrowing of the optical band gap from 3.2 eV for the undoped films to 2.5 eV for the N-doped films was observed. Photocatalytic activity tests were done using a methylene blue (MB) dye solution. The irradiation of the films in the MB dye solution was carried out with an emission lamp in the UV and in the visible range for undoped and N-doped TiO2 films, respectively. The results showed that the N-doped TiO2 films had a higher photocalytic activity in the visible range, reaching a greater MB degradation in comparison with undoped samples, which were subjected to a higher energy radiation.Fil: Franco Arias, Lina Maria. Penile Cancer in Cali, Colombia: 10 Years of Casuistry in a Tertiary Referral Center of a Middle-Income Country
Background: Penile cancer is a rare disease in Colombia; in Cali, it represents 0.7% of all cancers. Penile cancer has been associated with old age, bad hygiene, smoking and lack of circumcision. This study aimed to describe the sociodemographic and clinica characteristics of patients with penile cancer who consulted to a tertiary referral hospital. Methods: A case series of all penile cance cases at a reference institution in Cali during 2001-2010. Socioeconomic, demographic and clinical features of patients were described, and bivariate analyses were carried out. Results: There were 46 penile cancer cases. The average age was 60 ± 16.9 years. The main reason for consultation was an exophytic mass on the penis (75.0%). The most common location was the glans (69.6%), and the more frequent histology type was the squamous cell carcinoma (95.7%). With regard to risk factors, 65.5% of the patients had history of smoking and90.9% did not have circumcision. Patients who underwent radical amputation had higher rates of positive nodes (55% vs. 13.5%, p=0.015) and ulcerative lesions (77.8% vs. 29.7%, p=0.018) than those who did not have the procedure done. Recurrence was associated with the presence of lymphadenopathy (p=0.02) and history of circumcision (p=0.015). Conclusion: Most of the patients with penile cancer found in this study had old age, history of tobacco use and lack of circumcision. Patients who presented with lymph node metastasis had to undergo more radical procedures and suffered a greater rate of recurrence compared with those without lymph node involvement. Robust ..
Resultados clínicos en receptores de trasplante renal posterior a la conversión a ImTOR
Introducción: los ImTOR, sirolimus y everolimus son una alternativa de inmunosupresión en personas que han recibido transplantes rena-les. En este artículo, se describe la experiencia de pacientes que han experimentado una conversión a ImTOR, y a los que se les ha hecho un seguimiento por más de cinco años.Materiales y métodos: se incluyeron pacientes con transplantes renales desde 1995 hasta 2013, quienes tuvieron indicación de suspensión del inhibidor de calcineurina (ICN) después del tercer mes posterior al trasplante. Todos los pacientes fueron sometidos a biopsia renal antes de la administración de ImTOR. Ningún paciente tuvo diagnóstico de nefropatía crónica, IFTA >40 % o proteinuria >350 mg/24h. Se elaboró un análisis descriptivo para todas las variables. Para estudiar la supervivencia del paciente y del injerto, y la incidencia de rechazo agudo, se usó el método de Kaplan-Meier.Resultados: de 1273 trasplantes renales, la conversión de ICN a ImTOR se realizó en 166 casos (13 %). Al 78 % (n=129) se le administró sirolimus. El 13 % de los pacientes perdió la función del injerto y 7 pacientes (4,2 %) fallecieron. En el 37 % de los casos, se retiró el ImTOR. La principal causa de retiro fue el hallazgo de proteinuria patológica. La incidencia de rechazo agudo después del cambio a ImTOR fue de 9,6 %. La supervivencia del injerto tras uno y cinco años fue de 96,6 % y 83,5 %, respectivamente; y la supervivencia del paciente a uno y cinco años fue de 98 % y 97 %, respectivamente.Conclusiones: el uso de inhibidores ImTOR parece ser seguro en este grupo de pacientes trasplantados, pues hubo una baja tasa de rechazo y buena supervivencia del injert
Implicaciones metodológicas e inconsistencias de la Tercera Comunicación Nacional sobre Cambio Climático de Colombia
Las Comunicaciones Nacionales sobre Cambio Climático (CNCC) son un mecanismo para que los países informen sus avances en mitigación y adaptación, y constituyen uno de los elementos de base para la política sobre cambio climático a escala nacional. Colombia ha emitido tres CNCC. La tercera plantea un escenario que considera las proyecciones de diversos modelos incluidos en la quinta fase del Proyecto de Comparación de Modelos Acoplados (Coupled Model Intercomparison Project, CMIP), el cual se estima como el promedio de las proyecciones correspondientes a las cuatro trayectorias de concentración representativa (Representative Concentration Pathways,RCP) presentadas en el quinto reporte de evaluación del Panel Intergubernamental sobre Cambio Climático. Cada una de estas RCP representa una trayectoria de concentración de gases de efecto invernadero (GEI) para un escenario particular de crecimiento poblacional, económico y tecnológico que conduce a una posible trayectoria de evolución del sistema climático. En este estudio se comparan las proyecciones presentadas en la Tercera CNCC con las obtenidas directamente de los modelos empleados. Nuestros resultados demuestran que al utilizarse un promedio de RCP se pierden escenarios alternos que podrían ser importantes a la hora de considerar posibles futuros diferentes y anulan la utilidad de plantear diversas trayectorias de emisiones de GEI. Más aun, una comparación entre la Segunda y la Tercera CNCC muestra proyecciones de precipitación opuestas para diferentes regiones del país, lo cual es de particular importancia, pues el escenario de cambio climático planteado en la Tercera CNCC sirve de referencia para la toma de decisiones en materia de cambio climático a nivel nacional
High incidence of Noonan syndrome features including short stature and pulmonic stenosis in patients carrying NF1 missense mutations affecting p.Arg1809: genotype-phenotype correlation
Neurofibromatosis type 1 (NF1) is one of the most frequent genetic disorders, affecting 1:3,000 worldwide. Identification of genotype-phenotype correlations is challenging because of the wide range clinical variability, the progressive nature of the disorder, and extreme diversity of the mutational spectrum. We report 136 individuals with a distinct phenotype carrying one of five different NF1 missense mutations affecting p.Arg1809. Patients presented with multiple cafe-au-lait macules (CALM) with or without freckling and Lisch nodules, but no externally visible plexiform neurofibromas or clear cutaneous neurofibromas were found. About 25% of the individuals had Noonan-like features. Pulmonic stenosis and short stature were significantly more prevalent compared with classic cohorts (P<0.0001). Developmental delays and/or learning disabilities were reported in over 50% of patients. Melanocytes cultured from a CALM in a segmental NF1-patient showed two different somatic NF1 mutations, p.Arg1809Cys and a multi-exon deletion, providing genetic evidence that p.Arg1809Cys is a loss-of-function mutation in the melanocytes and causes a pigmentary phenotype. Constitutional missense mutations at p.Arg1809 affect 1.23% of unrelated NF1 probands in the UAB cohort, therefore this specific NF1 genotype-phenotype correlation will affect counseling and management of a significant number of patients
Seasonality of mortality under climate change: a multicountry projection study
A warming climate can substantially change the seasonality of mortality in the future. Our projections suggest that health-care systems should consider preparing for a potentially increased demand during warm seasons and sustained high demand during cold seasons, particularly in regions characterised by arid, temperate, and continental climates
