    Chronicles of nature calendar, a long-term and large-scale multitaxon database on phenology

    We present an extensive, large-scale, long-term and multitaxon database on phenological and climatic variation, involving 506,186 observation dates acquired in 471 localities in Russian Federation, Ukraine, Uzbekistan, Belarus and Kyrgyzstan. The data cover the period 1890-2018, with 96% of the data being from 1960 onwards. The database is rich in plants, birds and climatic events, but also includes insects, amphibians, reptiles and fungi. The database includes multiple events per species, such as the onset days of leaf unfolding and leaf fall for plants, and the days for first spring and last autumn occurrences for birds. The data were acquired using standardized methods by permanent staff of national parks and nature reserves (87% of the data) and members of a phenological observation network (13% of the data). The database is valuable for exploring how species respond in their phenology to climate change. Large-scale analyses of spatial variation in phenological response can help to better predict the consequences of species and community responses to climate change.Peer reviewe

    Phenological shifts of abiotic events, producers and consumers across a continent

    Ongoing climate change can shift organism phenology in ways that vary depending on species, habitats and climate factors studied. To probe for large-scale patterns in associated phenological change, we use 70,709 observations from six decades of systematic monitoring across the former Union of Soviet Socialist Republics. Among 110 phenological events related to plants, birds, insects, amphibians and fungi, we find a mosaic of change, defying simple predictions of earlier springs, later autumns and stronger changes at higher latitudes and elevations. Site mean temperature emerged as a strong predictor of local phenology, but the magnitude and direction of change varied with trophic level and the relative timing of an event. Beyond temperature-associated variation, we uncover high variation among both sites and years, with some sites being characterized by disproportionately long seasons and others by short ones. Our findings emphasize concerns regarding ecosystem integrity and highlight the difficulty of predicting climate change outcomes. The authors use systematic monitoring across the former USSR to investigate phenological changes across taxa. The long-term mean temperature of a site emerged as a strong predictor of phenological change, with further imprints of trophic level, event timing, site, year and biotic interactions.Peer reviewe

    Design and baseline characteristics of the finerenone in reducing cardiovascular mortality and morbidity in diabetic kidney disease trial

    Background: Among people with diabetes, those with kidney disease have exceptionally high rates of cardiovascular (CV) morbidity and mortality and progression of their underlying kidney disease. Finerenone is a novel, nonsteroidal, selective mineralocorticoid receptor antagonist that has shown to reduce albuminuria in type 2 diabetes (T2D) patients with chronic kidney disease (CKD) while revealing only a low risk of hyperkalemia. However, the effect of finerenone on CV and renal outcomes has not yet been investigated in long-term trials. Patients and Methods: The Finerenone in Reducing CV Mortality and Morbidity in Diabetic Kidney Disease (FIGARO-DKD) trial aims to assess the efficacy and safety of finerenone compared to placebo at reducing clinically important CV and renal outcomes in T2D patients with CKD. FIGARO-DKD is a randomized, double-blind, placebo-controlled, parallel-group, event-driven trial running in 47 countries with an expected duration of approximately 6 years. FIGARO-DKD randomized 7,437 patients with an estimated glomerular filtration rate >= 25 mL/min/1.73 m(2) and albuminuria (urinary albumin-to-creatinine ratio >= 30 to <= 5,000 mg/g). The study has at least 90% power to detect a 20% reduction in the risk of the primary outcome (overall two-sided significance level alpha = 0.05), the composite of time to first occurrence of CV death, nonfatal myocardial infarction, nonfatal stroke, or hospitalization for heart failure. Conclusions: FIGARO-DKD will determine whether an optimally treated cohort of T2D patients with CKD at high risk of CV and renal events will experience cardiorenal benefits with the addition of finerenone to their treatment regimen. The analysed body of work of this wordsmith represents a natural phase of evolution of national, Russian, foreign literatures which, absorbing several currents, arises as a new phenomenon in the existing artistic system, reflecting both certain characteristics of non-classical rhetoric and recent trends in public conscience, where search for the meaning is transferred from social and philosophical areas into spheres of intuition and aesthetic. The current interest to the non-classical rhetoric dictates the need to make sense of nuances in Aygy’s lyrics, who was able to present through his own literary world the main idea of Chuvash ethnic community – aspiration towards light

    Fluorescent ratiometric pH indicator SypHer2: applications in neuroscience and regenerative biology

    AbstractBackground SypHer is a genetically encoded fluorescent pH-indicator with a ratiometric readout, suitable for measuring fast intracellular pH shifts. However, a relatively low brightness of the indicator limits its use. Methods Here we designed a new version of pH-sensor - SypHer-2, that has up to three times brighter fluorescence signal in cultured mammalian cells compared to the SypHer. Results Using the new indicator we registered activity-associated pH oscillations in neuronal cell culture. We observed prominent temporal neuronal cytoplasm acidification that occurs in parallel with calcium entry. Furthermore, we monitored pH in presynaptic and postsynaptic termini by targeting SypHer-2 directly to these compartments and revealed marked differences in pH dynamics between synaptic boutons and dendritic spines. Finally, we were able to reveal for the first time the intracellular pH drop which occurs within an extended region of the amputated tail of the Xenopus laevis tadpole before it begins to regenerate. Conclusions SypHer2 is suitable for quantitative monitoring of pH in biological systems of different scales, from small cellular subcompartments to animal tissues in vivo. General significance The new pH-sensor will help to investigate pH-dependent processes in both in vitro and in vivo studies

    Synthesis and Characterization of Silver-Containing Nanocomposites Based on 1-Vinyl-1,2,4-triazole and Acrylonitrile Copolymer

    Insoluble silver-containing nanocomposites based on 1-vinyl-1,2,4-triazole and acrylonitrile copolymer was synthesized and characterized by scanning electron microscopy, Fourier transform infrared spectroscopy, atomic absorption, X-ray diffraction, and thermogravimetric data analysis. The silver content of the nanocomposites varied from 8.0 to 23.5% (the silver content depends on the ratio of the copolymer-silver salt). Synthesis of silver nanocomposites was carried out by thermal treatment of a polymer complex of copolymer-silver salt without the use of an additional reducing agent. X-ray diffraction was used to control the formation of silver nanoparticles at different temperatures (210°С, 220°С, and 250°С). Synthesis of nanocomposites shows a developed porous structure with a channel diameter in most cases of 1-4 microns. The EPR monitoring of the nanoparticle germination and growth reveals that the generation of the nanocomposites proceeds via a stage of complex formation between the polymeric matrix and silver ions. The synthesized nanocomposites possess paramagnetic properties and show a narrow EPR symmetric signal with the concentration of unpaired electrons of 1020 spin/g, a g-factor 2.0050, and signal width 5-6 Gs. Thermal stability of silver-containing nanocomposites is up to 270-320°C
