Binding of an antibody mimetic of the human low density lipoprotein receptor to apolipoprotein E is governed through electrostatic forces. Studies using site-directed mutagenesis and molecular modeling.
Monoclonal antibody 2E8 is specific for an epitope that coincides with the binding site of the low density lipoprotein receptor (LDLR) on human apoE. Its reactivity with apoE variants resembles that of the LDLR: it binds well with apoE3 and poorly with apoE2. The heavy chain complementarity-determining region (CDRH) 2 of 2E8 shows homology to the ligand-binding domain of the LDLR. To define better the structural basis of the 2E8/apoE interaction and particularly the role of electrostatic interactions, we generated and characterized a panel of 2E8 variants. Replacement of acidic residues in the 2E8 CDRHs showed that Asp52, Glu53, and Asp56 are essential for high-affinity binding. Although Asp31 (CDRH1), Glu58 (CDRH2), and Asp97 (CDRH3) did not appear to be critical, the Asp97 â Ala variant acquired reactivity with apoE2. A Thr57 â Glu substitution increased affinity for both apoE3 and apoE2. The affinities of wild-type 2E8 and variants for apoE varied inversely with ionic strength, suggesting that electrostatic forces contribute to both antigen binding and isoform specificity. We propose a model of the 2E8·apoE immune complex that is based on the 2E8 and apoE crystal structures and that is consistent with the apoE-binding properties of wild-type 2E8 and its variants. Given the similarity between the LDLR and 2E8 in terms of specificity, the LDLR/ligand interaction may also have an important electrostatic component

Early Observations And Analysis Of The Type Ia SN 2014J In M82
We present optical and near infrared (NIR) observations of the nearby Type Ia SN 2014J. Seventeen optical and 23 NIR spectra were obtained from 10 days before (-10d) to 10 days after (+10d) the time of maximum B-band brightness. The relative strengths of absorption features and their patterns of development can be compared at one day intervals throughout most of this period. Carbon is not detected in the optical spectra, but we identify C I lambda 1.0693 in the NIR spectra. Mg II lines with high oscillator strengths have higher initial velocities than other Mg II lines. We show that the velocity differences can be explained by differences in optical depths due to oscillator strengths. The spectra of SN 2014J show that it is a normal SN Ia, but many parameters are near the boundaries between normal and high-velocity subclasses. The velocities for OI, Mg II, Si II, S Ca a, and Fell suggest that SN 2014J has a layered structure with little or no mixing. That result is consistent with the delayed detonation explosion models. We also report photometric observations, obtained from -10d to +29d, in the UBVRIJH and K-s bands. The template fitting package SNooPy is used to interpret the light curves and to derive photometric parameters. Using R-v = 1.46, which is consistent with previous studies, SNooPy finds that A(v) = 1.80 for E(B - V)(host) = 1.23 +/- 0.06 mag. The Advanced Compton Telescope
The Advanced Compton Telescope (ACT), the next major step in gamma-ray astronomy, will probe the fires where chemical elements are formed by enabling high-resolution spectroscopy of nuclear emission from supernova explosions. During the past two years, our collaboration has been undertaking a NASA mission concept study for ACT. This study was designed to (1) transform the key scientific objectives into specific instrument requirements, (2) to identify the most promising technologies to meet those requirements, and (3) to design a viable mission concept for this instrument. We present the results of this study, including scientific goals and expected performance, mission design, and technology recommendations

Rapid improvements in sequencing and array-based platforms are resulting in a flood of diverse genome-wide data, including data from exome and whole-genome sequencing, epigenetic surveys, expression profiling of coding and noncoding RNAs, single nucleotide polymorphism (SNP) and copy number profiling, and functional assays. Analysis of these large, diverse data sets holds the promise of a more comprehensive understanding of the genome and its relation to human disease. Experienced and knowledgeable human review is an essential component of this process, complementing computational approaches. This calls for efficient and intuitive visualization tools able to scale to very large data sets and to flexibly integrate multiple data types, including clinical data. A Comparison between Four Analytical Methods for the Measurement of Fe(II) at Nanomolar Concentrations in Coastal Seawater
Dissolved Fe(II) in seawater is deemed an important micronutrient for microbial organisms, but its analysis is challenging due to its transient nature. We conducted a series of Fe(II) method comparison experiments, where spikes of 5 to 31 nM Fe(II) were added to manipulated seawaters with varying dissolved oxygen (37 to 156 ÎŒM) concentrations. The observed Fe(II) concentrations from four analytical methods were compared: spectrophotometry with ferrozine, stripping voltammetry, and flow injection analysis using luminol (with, and without, a pre-concentration column). Direct comparisons between the different methods were undertaken from the derived apparent Fe(II) oxidation rate constant (k1). Whilst the two luminol based methods produced the most similar concentrations throughout the experiments, k1 was still subject to a 20â30% discrepancy between them. Contributing factors may have included uncertainty in the calibration curves, and different responses to interferences from Co(II) and humic/fulvic organic material. The difference in measured Fe(II) concentrations between the luminol and ferrozine methods, from 10 minâ2 h after the Fe(II) spikes were added, was always relatively large in absolute terms (>4 nM) and relative to the spike added (>20% of the initial Fe(II) concentration). k1 derived from ferrozine observed Fe(II) concentrations was 3â80%, and 4â16%, of that derived from luminol observed Fe(II) with, and without, pre-concentration respectively. The poorest comparability of k1 was found after humic/fulvic material was added to raise dissolved organic carbon to 120 ÎŒM. A luminol method without pre-concentration then observed Fe(II) to fall below the detection limit (<0.49 nM) within 10 min of a 17 nM Fe(II) spike addition, yet other methods still observed Fe(II) concentrations of 2.7 to 3.7 nM 30 min later. k1 also diverged accordingly with the ferrozine derived value 4% of that derived from luminol without pre-concentration. These apparent inconsistencies suggest that some inter-dataset differences in measured Fe(II) oxidation rates in natural waters may be attributable to differences in the analytical methods used rather than arising solely from substantial shifts in Fe(II) speciation

The Emerald International Handbook of Feminist Perspectives on Womenâs Acts of Violence
Over the past twenty years, interest in womenâs violence has grown as an area of academic research and teaching across disciplines such as criminology, sociology, history, international relations, public health and film and literary studies. This handbook makes a timely contribution by acting as a comprehensive introduction to a wide range of international, interdisciplinary scholarship which applies feminist perspectives to the phenomenon of womenâs violence. Violence is enabled and enacted by individuals, organisations and states with interconnections between these different levels (Collins, 1998, 2017). We adopt this multilevel understanding of violence in the handbook, bringing together contributions on interpersonal and intimate violence by women, womenâs violence as agents of institutions, and womenâs political violence as state and non-state actors. The handbook is international in scope with contributions from scholars across countries in the Global South and North. Womenâs acts of violence are rarer than menâs and frequently perceived as more shocking. Violence by women is regularly sensationalised and stigmatised, especially in media discourses. This sensationalisation and stigmatisation relies on and reproduces misogynistic tropes about violent women as âevilâ, âunnaturalâ and masculinised. The chapters in this handbook are written from a feminist perspective and eschew or deconstruct stereotypical portrayals in favour of more considered and complex analyses of the violence women enact and the relevance of their social and political positioning, as well as cultural understandings of womanhood and how these inform understandings of and responses to this violence. Previous literature has emphasised that womenâs violence is frequently understood as deviant and transgressive, violating norms of ideal femininity. Such ideas are explored throughout the handbook in contributions which consider, for instance, the vilification and monsterisation of women who kill. These chapters tease out the ways in which violent women are profiled as unnatural and abject. The handbook therefore retains a focus on scholarship which considers the âabnormalityâ of violent women while also including contributions which demonstrate that womenâs violent acts can be normalised and made invisible, for example when perpetrated during a professional role. Adding further nuance, various chapters address how, due to marginalisation across axes of race and class, certain women are not always presumed to be non-violent or perceived through norms of ideal femininity. The handbook explores how these assumptions can lead to overcriminalisation and harsh treatment within the criminal justice system. The significance of womenâs intersectional identities is a consistent theme throughout the handbook. Running through the chapters too is the seemingly intractable problem of agency â including the obstacles to fully assigning agency to violent women as well as the frequently unwanted consequences when they are considered to have acted with agency and are punished more harshly. Throughout the handbook, authors grapple with questions of womenâs volitional capacity, considering difficult questions of how far we should consider the contexts in which women commit violence, which include structural oppression, domestic- and gender-based violence, and cultural norms. The contributions reveal the necessity of abandoning a binary view of victim-perpetrator, agency/non-agency and evolving a more complex framework in which to gauge questions of intention and deliberation. The handbook is divided into eight sections: historical perspectives; understanding womenâs acts of violence; women as perpetrators of interpersonal and intimate violence; power and womenâs violence; women and non-state political violence; cultural interpretations of violent women; fictional representations of violent women; and violent women and girls in the criminal justice system. The Rapidly Flaring Afterglow of the Very Bright and Energetic GRB 070125
We report on multi-wavelength observations, ranging from the X-ray to radio
wave bands, of the IPN-localized gamma-ray burst GRB 070125. Spectroscopic
observations reveal the presence of absorption lines due to O I, Si II, and C
IV, implying a likely redshift of z = 1.547. The well-sampled light curves, in
particular from 0.5 to 4 days after the burst, suggest a jet break at 3.7 days,
corresponding to a jet opening angle of ~7.0 degrees, and implying an intrinsic
GRB energy in the 1 - 10,000 keV band of around E = (6.3 - 6.9)x 10^(51) erg
(based on the fluences measured by the gamma-ray detectors of the IPN network).
GRB 070125 is among the brightest afterglows observed to date. The spectral
energy distribution implies a host extinction of Av < 0.9 mag. Two
rebrightening episodes are observed, one with excellent time coverage, showing
an increase in flux of 56% in ~8000 seconds. The evolution of the afterglow
light curve is achromatic at all times. Late-time observations of the afterglow
do not show evidence for emission from an underlying host galaxy or supernova.
Any host galaxy would be subluminous, consistent with current GRB host-galaxy
samples. Evidence for strong Mg II absorption features is not found, which is
perhaps surprising in view of the relatively high redshift of this burst and
