    The Late Time Light Curve of SN 1998bw Associated with GRB980425

    We report 139 photometric observations through the B, V, and I filters of the supernova SN 1998bw, an object which is associated with the Gamma-Ray Burst GRB 980425. Detailed light curves of this unique supernova can be compared to theoretical models, so we report here our light curve for 123 days between 27 June 1998 and 28 October 1998. The light curve of SN 1988bw is consistent with those of the Type Ic class. We find that the magnitude-versus-time relation for this supernova is linear to within 0.05 mags in all colors over the entire duration of our study. Our measured uniform decline rates are 0.0141±0.00020.0141 \pm 0.0002, 0.0184±0.00030.0184 \pm 0.0003, and 0.0181±0.00030.0181 \pm 0.0003 magnitudes per day in the B, V, and I bands. The linear decline and the rate of that decline suggest that late time light curve is powered by the radioactive decay of cobalt with some leakage of the gamma rays.Comment: 15 pages, 1 figure, 1 table, Accepted for publication in PAS

    Quality of Postoperative Pain Management after Midfacial Fracture Repair—An Outcome-oriented Study

    Objectives There is a lack of literature regarding the procedure-specific quality of acute postoperative pain management after midfacial fracture repair. The purpose of the presented prospective clinical study was to evaluate postoperative pain management after surgical repair of midfacial fractures. Materials and methods Eighty-five adults were evaluated on the first postoperative day following midfacial repair using the questionnaire of the Quality Improvement in Postoperative Pain Management (QUIPS) project. The main outcome measures were patients’ characteristics and clinical- and patient-reported outcome parameters. Results Overall, pain on the first postoperative day was moderate. A significant correlation between process and outcome parameters could be shown. Duration of surgery above the calculated median was significantly associated with higher maximum pain intensity (p = 0.017). Patients requiring opioids in the recovery room presented significantly higher pain on activity (p = 0.029) and maximum pain (p = 0.035). Sleeping impairment (p = 0.001) and mood disturbance (p = 0.008) were significantly more prevalent in patients undergoing repair of a centrolateral midfacial fracture. Conclusions QUIPS is a simple and qualified tool to evaluate the procedure specific quality of acute postoperative pain management. Pain on the first postoperative day following midfacial fracture repair seems overall to be moderate. Nearly a third of the patients showed inadequate postoperative pain management. To prevent inadequate postoperative pain management, it is necessary to establish a continued procedure-specific outcome measurement

    The Data Analysis Pipeline for the SDSS-IV MaNGA IFU Galaxy Survey: Emission-Line Modeling

    SDSS-IV MaNGA (Mapping Nearby Galaxies at Apache Point Observatory) is the largest integral-field spectroscopy survey to date, aiming to observe a statistically representative sample of 10,000 low-redshift galaxies. In this paper we study the reliability of the emission-line fluxes and kinematic properties derived by the MaNGA Data Analysis Pipeline (DAP). We describe the algorithmic choices made in the DAP with regards to measuring emission-line properties, and the effect of our adopted strategy of simultaneously fitting the continuum and line emission. The effect of random errors are quantified by studying various fit-quality metrics, idealized recovery simulations and repeat observations. This analysis demonstrates that the emission lines are well-fit in the vast majority of the MaNGA dataset and the derived fluxes and errors are statistically robust. The systematic uncertainty on emission-line properties introduced by the choice of continuum templates is also discussed. In particular, we test the effect of using different stellar libraries and simple stellar-population models on the derived emission-line fluxes and the effect of introducing different tying prescriptions for the emission-line kinematics. We show that these effects can generate large (>> 0.2 dex) discrepancies at low signal-to-noise and for lines with low equivalent width (EW); however, the combined effect is noticeable even for Hα\alpha EW >> 6~\AA. We provide suggestions for optimal use of the data provided by SDSS data release 15 and propose refinements on the \DAP\ for future MaNGA data releases.Comment: accepted on A

    The Mid-Infrared Emission of M87

    We discuss Subaru and Spitzer Space Telescope imaging and spectroscopy of M87 in the mid-infrared from 5-35 um. These observations allow us to investigate mid-IR emission mechanisms in the core of M87 and to establish that the flaring, variable jet component HST-1 is not a major contributor to the mid-IR flux. The Spitzer data include a high signal-to-noise 15-35 μ\mum spectrum of the knot A/B complex in the jet, which is consistent with synchrotron emission. However, a synchrotron model cannot account for the observed {\it nuclear} spectrum, even when contributions from the jet, necessary due to the degrading of resolution with wavelength, are included. The Spitzer data show a clear excess in the spectrum of the nucleus at wavelengths longer than 25 um, which we model as thermal emission from cool dust at a characteristic temperature of 55 \pm 10 K, with an IR luminosity \sim 10^{39} {\rm ~erg ~s^{-1}}. Given Spitzer's few-arcsecond angular resolution, the dust seen in the nuclear spectrum could be located anywhere within ~5'' (390 pc) of the nucleus. In any case, the ratio of AGN thermal to bolometric luminosity indicates that M87 does not contain the IR-bright torus that classical unified AGN schemes invoke.     An initial assessment of SMAP soil moisture retrievals using high-resolution model simulations and in situ observations

    At the end of its first year of operation, we compare soil moisture retrievals from the Soil Moisture Active Passive (SMAP) mission to simulations from a land surface model with meteorological forcing downscaled from observations/reanalysis and in situ observations from sparse monitoring networks within continental United States (CONUS). The radar failure limits the duration of comparisons for the active and combined products (~3 months). Nevertheless, the passive product compares very well against in situ observations over CONUS. On average, SMAP compares to the in situ data even better than the land surface model and provides significant added value on top of the model and thus good potential for data assimilation. At large scale, SMAP is in good agreement with the model in most of CONUS with less-than-expected degradation over mountainous areas. Lower correlation between SMAP and the model is seen in the forested east CONUS and significantly lower over the Canadian boreal forests.United States.     A TALEN Genome-Editing System for Generating Human Stem Cell-Based Disease Models

    SummaryTranscription activator-like effector nucleases (TALENs) are a new class of engineered nucleases that are easier to design to cleave at desired sites in a genome than previous types of nucleases. We report here the use of TALENs to rapidly and efficiently generate mutant alleles of 15 genes in cultured somatic cells or human pluripotent stem cells, the latter for which we differentiated both the targeted lines and isogenic control lines into various metabolic cell types. We demonstrate cell-autonomous phenotypes directly linked to disease—dyslipidemia, insulin resistance, hypoglycemia, lipodystrophy, motor-neuron death, and hepatitis C infection. We found little evidence of TALEN off-target effects, but each clonal line nevertheless harbors a significant number of unique mutations. Given the speed and ease with which we were able to derive and characterize these cell lines, we anticipate TALEN-mediated genome editing of human cells becoming a mainstay for the investigation of human biology and disease

    Academic detailing to increase colorectal cancer screening by primary care practices in Appalachian Pennsylvania

    <p>Abstract</p> <p>Background</p> <p>In the United States, colorectal cancer (CRC) is the third most frequently diagnosed cancer and second leading cause of cancer death. Screening is a primary method to prevent CRC, yet screening remains low in the U.S. and particularly in Appalachian Pennsylvania, a largely rural area with high rates of poverty, limited health care access, and increased CRC incidence and mortality rates. Receiving a physician recommendation for CRC screening is a primary predictor for patient adherence with screening guidelines. One strategy to disseminate practice-oriented interventions is academic detailing (AD), a method that transfers knowledge or methods to physicians, nurses or office staff through the visit(s) of a trained educator. The objective of this study was to determine acceptability and feasibility of AD among primary care practices in rural Appalachian Pennsylvania to increase CRC screening.</p> <p>Methods</p> <p>A multi-site, practice-based, intervention study with pre- and 6-month post-intervention review of randomly selected medical records, pre- and post-intervention surveys, as well as a post-intervention key informant interview was conducted. The primary outcome was the proportion of patients current with CRC screening recommendations and having received a CRC screening within the past year. Four practices received three separate AD visits to review four different learning modules.</p> <p>Results</p> <p>We reviewed 323 records pre-intervention and 301 post-intervention. The prevalence of being current with screening recommendation was 56% in the pre-intervention, and 60% in the post-intervention (p = 0. 29), while the prevalence of having been screened in the past year increased from 17% to 35% (p < 0.001). Colonoscopies were the most frequently performed screening test. Provider knowledge was improved and AD was reported to be an acceptable intervention for CRC performance improvement by the practices.</p> <p>Conclusions</p> <p>AD appears to be acceptable and feasible for primary care providers in rural Appalachia. A ceiling effect for CRC screening may have been a factor in no change in overall screening rates. While the study was not designed to test the efficacy of AD on CRC screening rates, our evidence suggests that AD is acceptable and may be efficacious in increasing recent CRC screening rates in Appalachian practices which could be tested through a randomized controlled study.</p

    Incidence and determinants of new AIDS-defining illnesses after HAART initiation in a Senegalese cohort

    <p>Abstract</p> <p>Background</p> <p>Although a dramatic decrease in AIDS progression has been observed after Highly Active Anti Retroviral Therapy (HAART) in both low- and high-resource settings, few data support that fact in low-resource settings.</p> <p>This study describes the incidence of AIDS-defining illnesses (ADI) after HAART initiation and analyzes their risk factors in a low-resource setting. A focus was put on CD4 cell counts and viral load measurements.</p> <p>Methods</p> <p>404 HIV-1-infected Senegalese adult patients were enrolled in a prospective observational cohort and data censored as of April 2008. A Poisson regression was used to model the incidence of ADIs over two periods and to assess its association with baseline variables, current CD4, current viral load, CD4 response, and virological response.</p> <p>Results</p> <p>ADI incidence declined from 20.5 ADIs per 100 person-years, 95% CI = [16.3;25.8] during the first year to 4.3, 95% CI = [2.3;8.1] during the fourth year but increased afterwards. Before 42 months, the decrease was greater in patients with clinical stage CDC-C at baseline and with a viral load remaining below 1000 cp/mL but was uniform across CD4 strata (p = 0.1). After 42 months, 293 patients were still at risk. The current CD4 and viral load were associated with ADI incidence (decrease of 21% per 50 CD4/mm<sup>3 </sup>and of 61% for patients with a viral load < 1000 cp/mL).</p> <p>Conclusions</p> <p>During the first four years, a uniform decline of ADI incidence was observed even in patients with low CD4-cell counts at HAART initiation as long as the viral load remained undetectable. An increase was noted later in patients with immunologic and virological failures but also in patients with only virological failure.</p

    Co-Conserved Features Associated with cis Regulation of ErbB Tyrosine Kinases

    BACKGROUND: The epidermal growth factor receptor kinases, or ErbB kinases, belong to a large sub-group of receptor tyrosine kinases (RTKs), which share a conserved catalytic core. The catalytic core of ErbB kinases have functionally diverged from other RTKs in that they are activated by a unique allosteric mechanism that involves specific interactions between the kinase core and the flanking Juxtamembrane (JM) and COOH-terminal tail (C-terminal tail). Although extensive studies on ErbB and related tyrosine kinases have provided important insights into the structural basis for ErbB kinase functional divergence, the sequence features that contribute to the unique regulation of ErbB kinases have not been systematically explored. METHODOLOGY/PRINCIPAL FINDINGS: In this study, we use a Bayesian approach to identify the selective sequence constraints that most distinguish ErbB kinases from other receptor tyrosine kinases. We find that strong ErbB kinase-specific constraints are imposed on residues that tether the JM and C-terminal tail to key functional regions of the kinase core. A conserved RIxKExE motif in the JM-kinase linker region and a glutamine in the inter-lobe linker are identified as two of the most distinguishing features of the ErbB family. While the RIxKExE motif tethers the C-terminal tail to the N-lobe of the kinase domain, the glutamine tethers the C-terminal tail to hinge regions critical for inter-lobe movement. Comparison of the active and inactive crystal structures of ErbB kinases indicates that the identified residues are conformationally malleable and can potentially contribute to the cis regulation of the kinase core by the JM and C-terminal tail. ErbB3, and EGFR orthologs in sponges and parasitic worms, diverge from some of the canonical ErbB features, providing insights into sub-family and lineage-specific functional specialization. CONCLUSION/SIGNIFICANCE: Our analysis pinpoints key residues for mutational analysis, and provides new clues to cancer mutations that alter the canonical modes of ErbB kinase regulation
