    Seeing a drummer’s performance modulates the subjective experience of groove when listening to popular music drum patterns

    Spontaneous rhythmical movements, like foot-tapping and head-bobbing, often emerge when people listen to music, promoting the sensation of being in the ‘groove’: a psychological construct that additionally incorporates positive affect. Here we report the first study to investigate if seeing the music maker modulates this subjective experience of groove. Across trials participants (n = 36) listened to high and low groove drum beats, while concurrently observing a task-irrelevant point-light display (PLD) of the drummer. The PLD was either fully-corresponding with the audio or was incompatible across three other visual display conditions: a static PLD, a corresponding but asynchronous PLD (0.5s time shifted); or a non-corresponding PLD (e.g. low groove audio paired with high groove PLD). Participants rated: (a) their desire to move; and (b) the perceived groove in response to the auditory beats only, using 8-point Likert scales. In both measurements there were significant main effects of groove level and of visual display. Ratings were higher for high compared to low groove audio, and for the fully-corresponding condition compared to the other three visual conditions. The participants’ desire to move was also rated higher in the static condition compared to the non-corresponding condition, while the two-way interaction was also significant: ratings were higher for the high compared to low groove audio in the fully-corresponding, static and asynchronous conditions, but not in the non-corresponding condition. These findings identify the importance of seeing as well as hearing the musician for an enhanced listening experience, which necessitates a multimodal account of music perception

    Community-based control of a neglected tropical disease: the mossy foot treatment and prevention association

    Podoconiosis (endemic non-filarial elephantiasis, also known as mossy foot) is a non-communicable disease now found exclusively in the tropics, caused by the conjunction of environmental, genetic, and economic factors. Silicate particles formed by the disintegration of lava in areas of high altitude (over 1,000 m) and seasonal rainfall (over 1,000 mm per annum) penetrate the skin of barefoot subsistence farmers, and in susceptible individuals cause lymphatic blockage and subsequent elephantiasis [1]. Although an estimated one million Ethiopians (of a total population of 77 million) are afflicted with podoconiosis [2], which creates a huge economic burden in endemic areas [3], no national policy has yet been developed to control or prevent the condition, and most affected communities remain unaware of treatment options

    In Vivo RNAi Screening Identifies Regulators of Actin Dynamics as Key Determinants of Lymphoma Progression

    April 1, 2010Mouse models have markedly improved our understanding of cancer development and tumor biology. However, these models have shown limited efficacy as tractable systems for unbiased genetic experimentation. Here, we report the adaptation of loss-of-function screening to mouse models of cancer. Specifically, we have been able to introduce a library of shRNAs into individual mice using transplantable Eμ-myc lymphoma cells. This approach has allowed us to screen nearly 1,000 genetic alterations in the context of a single tumor-bearing mouse. These experiments have identified a central role for regulators of actin dynamics and cell motility in lymphoma cell homeostasis in vivo. Validation experiments confirmed that these proteins represent bona fide lymphoma drug targets. Additionally, suppression of two of these targets, Rac2 and twinfilin, potentiated the action of the front-line chemotherapeutic vincristine, suggesting a critical relationship between cell motility and tumor relapse in hematopoietic malignancies.National Institutes of Health (U.S.) (RO1 CA128803-01)Massachusetts Institute of Technology.     Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

    PANC Study (Pancreatitis: A National Cohort Study): national cohort study examining the first 30 days from presentation of acute pancreatitis in the UK

    Abstract Background Acute pancreatitis is a common, yet complex, emergency surgical presentation. Multiple guidelines exist and management can vary significantly. The aim of this first UK, multicentre, prospective cohort study was to assess the variation in management of acute pancreatitis to guide resource planning and optimize treatment. Methods All patients aged greater than or equal to 18 years presenting with acute pancreatitis, as per the Atlanta criteria, from March to April 2021 were eligible for inclusion and followed up for 30 days. Anonymized data were uploaded to a secure electronic database in line with local governance approvals. Results A total of 113 hospitals contributed data on 2580 patients, with an equal sex distribution and a mean age of 57 years. The aetiology was gallstones in 50.6 per cent, with idiopathic the next most common (22.4 per cent). In addition to the 7.6 per cent with a diagnosis of chronic pancreatitis, 20.1 per cent of patients had a previous episode of acute pancreatitis. One in 20 patients were classed as having severe pancreatitis, as per the Atlanta criteria. The overall mortality rate was 2.3 per cent at 30 days, but rose to one in three in the severe group. Predictors of death included male sex, increased age, and frailty; previous acute pancreatitis and gallstones as aetiologies were protective. Smoking status and body mass index did not affect death. Conclusion Most patients presenting with acute pancreatitis have a mild, self-limiting disease. Rates of patients with idiopathic pancreatitis are high. Recurrent attacks of pancreatitis are common, but are likely to have reduced risk of death on subsequent admissions. </jats:sec

    Effects of exposing shrimp larvae (Pandalus borealis) to aquaculture pesticides at field relevant concentrations, with and without food limitation

    Anti-parasitic drugs used in the aquaculture industry are discharged to the sea after treatment of salmon. In this study, the effects of azamethiphos (AZA) in the Salmosan® formulation and deltamethrin (DEL) in the Alpha Max® formulation, have been assessed in Northern shrimp larvae (Pandalus borealis) when administered both separately and in combination. The exposure concentrations were 100 ng/L for AZA and 2 ng/L for DEL, each representing a 1000-fold dilution of the prescribed concentrations for salmon. These two chemicals were combined at these concentrations to give a third treatment (AZA + DEL). When larvae were exposed for two hours on the first, second and third days post hatch (dph), significantly increased mortality and reduced swimming activity were observed for larvae from the DEL and combined AZA + DEL treatments 4 dph, though not in larvae from the AZA treatment. A single pulse exposure, delivered on the first day post hatch, caused similar effects on mortality and swimming activity 4 dph as the three-pulse exposure. Mortality was driven by the presence of DEL in both experiments, with no amplification or reduction of effects observed when DEL and AZA were combined. Larvae were observed for 13 days following the single pulse exposure, with food limitation introduced as an additional stressor on day 4. In the DEL and AZA + DEL treatments mortality continued to increase regardless of food level, with no larvae completing development to stage II. The overriding toxicity of DEL masked any potential effects the reduced food ration may have exerted. Swimming activity was lower for AZA treated larvae than Control larvae 13 dph, when both groups were fed daily, though no other significant changes to mortality, development to stage II, feeding rate or gene expression were observed. Food limited Control and AZA larvae had lower swimming activity and feeding rate than daily fed Control larvae, with expression of pyruvate kinase and myosin genes also downregulated. However, there was no negative effect on survival or successful development to stage II in these treatments. In addition, mesencephalic astrocyte-derived neurotropic factor was downregulated in food limited Control larvae when compared with the daily fed Controls. Results from this study together with reported estimates of dispersion plume concentrations of discharged pesticides indicate that toxic concentrations of deltamethrin could reach shrimp larvae several kilometers from a treated salmon farm.publishedVersio
