    International audienceAccurate path length measurements in absorption cells are recurrent requirements in quantitative molecular absorption spectroscopy. A new twin path laser interferometer for length measurements in a simple direct path absorption geometry is presented, along with a full uncertainty budget. The path in an absorption cell is determined by measuring the optical path length change due to the diminution of the refractive index when the cell originally filled with nitrogen gas is evacuated. The performance of the instrument based on a stabilized HeNe laser is verified by comparison with the results of direct mechanical length measurements of a roughly 45 mm long, specially designed absorption cell

    Effect of Lipid Headgroup Charge and pH on the Stability and Membrane Insertion Potential of Calcium Condensed Gene Complexes

To access the final edited and published work see http://doi.org/10.1021/la504970n.Noncovalently condensed complexes of genetic material, cell penetrating peptides (CPPs), and calcium chloride present a nonviral route to improve transfection efficiency of nucleic acids (e.g., pDNA and siRNA). However, the exact mechanisms of membrane insertion and delivery of macromolecule complexes to intracellular locations as well as their stability in the intracellular environment are not understood. We show that calcium condensed gene complexes containing different hydrophilic (i.e., dTAT, K9, R9, and RH9) and amphiphilic (i.e., RA9, RL9, and RW9) CPPs formed stable cationic complexes of hydrodynamic radii 100 nm at neutral pH. However, increasing the acidity caused the complexes to become neutral or anionic and increase in size. Using zwitterionic and anionic phospholipid monolayers as models that mimic the membrane composition of the outer leaflet of cell membranes and intracellular vesicles and pHs that mimic the intracellular environment, we study the membrane insertion potential of these seven gene complexes (CPP/pDNA/Ca2+ complexes) into model membranes. At neutral pH, all gene complexes demonstrated the highest insertion potential into anionic phospholipid membranes, with complexes containing amphiphilic peptides showing the maximum insertion. However, at acidic pH, the gene complexes demonstrated maximum monolayer insertion into zwitterionic lipids, irrespective of the chemical composition of the CPP in the complexes. Our results suggest

    The electrochemical and chemical oxidation of extended TTF 4 and 5 are analysed by cyclic voltammetry, Visible/NIR and ESR spectroscopies, and the X-ray structures of the new salts 5·BF4(CH2Cl2) and 4·ClO4(THF)1/2 are presented. The effects of structural factors on the π-dimerization or the disproportionation reaction of the cation radical are shown. The oxidation of compound 4 presents the successive formation of stable cation radical and dication species both in dichloromethane (DCM) and in a CH3CN/THF mixture. In contrast, for compound 5, the stability of the oxidation states strongly depends on the nature of the solvent. In DCM, the oxidation of 5 proceeds by two close one-electron transfers while in CH3CN/THF the dication is directly formed via a two-electron process. The X-ray structures of the two salts reveal the formation of pi-dimers of cation radical. While the dimer (5(2))2+ is due mainly to π–π interactions between the conjugating spacer, the multiplication of the sulfur atoms in compound 4 contributes to stabilize the dimer by the combined effects of S–S and π–π interactions. Visible/NIR and ESR experiments confirm the higher tendency of 4+· to dimerize with the occurrence of dimer and monomer in solution, while for 5+· only the monomer is detected in DCM. On the other hand, by dissolution of 5·BF4(CH2Cl2) in CH3CN, only the neutral and the dicationic states of compounds 5 are observed owing to the disproportionation reaction

    Les auteurs rapportent une observation rare d'un jeune adolescent de 17 ans qui a présenté une fracture du processus coronoïde du coude survenue suite à un accident de sport. Il s'agit d'une observation décrivant un abord particulier par voie antérieure transversale permettant un contrôle direct et vissage en compression du fragment déplacé; l'évolution radio clinique était  satisfaisante. Nous discuterons à la lumière de la littérature, notre attitude thérapeutique, et l'évolution de ce cadre nosologique à travers l'analyse de cette observation

    A mortality event of the venerid bivalve Callista chione (Linnaeus, 1758) in a hatchery system. A case study

    Abnormal mortality of the smooth venus clam (Callista chione) was encountered when conditioning these clams in a hatchery system. A histopathological analysis was performed to establish the causes of this mortality episode. Our results showed an increase in rickettsia-like bacteria infection intensity between the individuals collected at the start of the conditioning in the hatchery and those collected during the mortality episode. Husbandry stress most likely increased disease susceptibility and progression in these clams. Rickettsia-like colonies were observed in large numbers in the gills of all individuals examined. Nematopsis sp. spores and rod-shaped basophilic bacteria could also been seen in some of the individuals examined. Microbiological analysis of clam tissue did not reveal the presence of any potentially pathogenic bacteria and all the clams were shown to be free of Perkinsus sp. parasites. The conditioning protocol was adapted from those used for other venerid clams due to the lack of data on this species. These findings highlight the need to perform further studies to evaluate the optimal parameters for C. chione broodstock conditioning

    Understanding the relationship between cerebellar structure and social abilities

    Background The cerebellum contains more than 50% of all neurons in the brain and is involved in a broad range of cognitive functions, including social communication and social cognition. Inconsistent atypicalities in the cerebellum have been reported in individuals with autism compared to controls suggesting the limits of categorical case control comparisons. Alternatively, investigating how clinical dimensions are related to neuroanatomical features, in line with the Research Domain Criteria approach, might be more relevant. We hypothesized that the volume of the “cognitive” lobules of the cerebellum would be associated with social difficulties. Methods We analyzed structural MRI data from a large pediatric and transdiagnostic sample (Healthy Brain Network). We performed cerebellar parcellation with a well-validated automated segmentation pipeline (CERES). We studied how social communication abilities—assessed with the social component of the Social Responsiveness Scale (SRS)—were associated with the cerebellar structure, using linear mixed models and canonical correlation analysis. Results In 850 children and teenagers (mean age 10.8 ± 3 years; range 5–18 years), we found a significant association between the cerebellum, IQ and social communication performance in our canonical correlation model. Limitations Cerebellar parcellation relies on anatomical boundaries, which does not overlap with functional anatomy. The SRS was originally designed to identify social impairments associated with autism spectrum disorders. Conclusion Our results unravel a complex relationship between cerebellar structure, social performance and IQ and provide support for the involvement of the cerebellum in social and cognitive processes
