    Clinical measurement of tooth wear: tooth Wear Indices

    Attrition, erosion, and abrasion result in alterations to the tooth and manifest as tooth wear. Each classification corresponds to a different process with specific clinical features. Classifications made so far have no accurate prevalence data because the indexes do not necessarily measure a specific etiology, or because the study populations can be diverse in age and characteristics. Tooth wears (attrition, erosion and abrasion) is perceived internationally as a growing problem. However, the interpretation and comparison of clinical and epidemiological studies, it is increasingly difficult because of differences in terminology and the large number of indicators/indices that have been developed for the diagnosis, classification and monitoring of the loss of dental hard tissue. These indices have been designed to identify increasing severity and are usually numerical, none have universal acceptance, complicating the evaluation of the true increase in prevalence reported. This article considers the ideal requirements for an erosion index. A literature review is conducted with the aim of analyzing the evolution of the indices used today and discuss whether they meet the clinical needs and research in dentistr

    La práctica especulativa como modelo pedagógico para la arquitectura

    Introducción: uno de los mayores desafíos de la arquitectura como disciplina después de la crisis, ha sido repensar en cómo tendría que enfocarse la formación de los (futuros) arquitectos. En un momento en el que parece que la práctica arquitectónica -cada vez más supeditada a los humores de los mercados-, haya quedado en segundo plano, un (re)encuentro con la teoría puede ser el camino para reconfigurar su cometido en los próximos años. Objetivos: el objetivo principal será describir la práctica especulativa como modelo pedagógico para la arquitectura. Gracias a ello, se quieren alcanzar dos objetivos específicos: por un lado, proporcionar claves de entendimiento para superar el antagonismo entre teoría y práctica; y por el otro, plantear su potencial para problemas sociales y políticos a gran escala. Método: el estudio se llevará a cabo a través de la comparación de pedagogías que utilizan actualmente la práctica especulativa en la arquitectura y en el diseño, y en interacción con otras prácticas culturales. Conclusiones: una vez que se consiga alejar la disciplina arquitectónica de los aspectos comerciales o de las demandas del mercado, resituándola en un contexto más amplio y global. La práctica especulativa, en este sentido, es lo que permite enfocar estrategias para futuros posibles, y repensar, a la vez, el presente en el que vivimos

    Green discoloration of the crown after internal root resorption treatment with grey mineral trioxide aggregate (MTA)

    Root resorption (RR) is the loss of dental hard tissues as a result of clastic activities. Internal inflammatory root resorption (IRR) is a type of RR characterized by progressive loss of tooth substance starting from the root canal wall. IRR is usually asymptomatic, slowly progressing, and detectable upon routine radiographic examination or by the clinical sign of a ‘pink spot’ when the IRR involves the crown or the coronal third of the root canal. Mineral trioxide aggregate (MTA) is a biocompatible cement that has been used successfully in pulp capping, pulpotomy, treatment of traumatized teeth with immature apices, and for treatment of root resorption. The treatment and follow-up of a maxillary central incisor with perforating IRR managed by root canal treatment and grey MTA repair is reported. IRR affected tooth 11 (FDI World Dental Federation) in a 29-year-old female patient, with a history of previous trauma. Follow-up radiographs over 24 months demonstrated the maintenance of a functional tooth. However, the tooth showed a green discoloration of the crown after MTA treatment

    La patología implanto-endodóncica (PIE) está descrita en la literatura implantológica como una de las causas de periimplantitis apical, entendida como la lesión osteolítica en la región apical del implante, con normal osteointegración de su porción coronal, provocada por la infección por contigüidad a partir de la lesión periapical del diente adyacente. Pero el concepto de PIE no sólo abarca la periimplantitis retrógrada por contaminación diente-a-implante, sino también los procesos inflamatorios periapicales en dientes adyacentes al implante por contaminación implante-adiente, cuando la colocación del implante provoca la necrosis del diente adyacente y la consiguiente periodontitis apical. Incluso podríamos incluir dentro de la PIE los casos de periimplantitis apical en implantes postextracción provocada por la infección residual presente en el alvéolo de un diente extraído con periodontitis apical. En definitiva, la PIE incluye las lesiones endodóncicas e implantarias apicales que son el resultado de infecciones residuales o por contigüidad entre diente e implante. En esta revisión bibliográfica se define y clasifica la PIE, repasándose la casuística publicada así como su influencia en el resultado del tratamiento implantológico

    Nuevas metodologías para el fomento del aprendizaje autónomo y colaborativo del estudiante en el conocimiento teórico-práctico de la arquitectura

    Tradicionalmente el concepto de patrimonio se ha considerado lo opuesto a la sostenibilidad, sin embargo, hoy día, debido a la crisis económica y a la falta de recursos, es el binomio patrimonio-sostenibilidad la apuesta clave para implicar a los arquitectos en un desarrollo urbano sostenible. Asimismo la incorporación de distintas disciplinas en el abordaje patrimonial facilita al arquitecto la gestión patrimonial desde un punto de vista holístico e inclusivo. El objetivo principal es proporcionar una serie de herramientas a los estudiantes que les permita abordar un problema patrimonial desde un enfoque sostenible y con un refuerzo teórico. Gracias a ello se quieren alcanzar dos objetivos específicos, por un lado, fomentar la resolución de problemas en equipo, y por el otro facilitar la comprensión del objeto patrimonial con una dimensión contemporánea inserto en la ciudad. El estudio se llevará a cabo a través del análisis de los textos y cartas internacionales que existen en materia de patrimonio. En paralelo, el estudiante debe ir identificando los nuevos valores patrimoniales generados al caso de estudio. La comprensión de la evolución del concepto de patrimonio aplicado al objeto de estudio permite al estudiante comprender la dimensión global del bien patrimonial, así como la cantidad de actores y disciplinas implicadas en su gestión

    Monitoring of emerging water stress situations by thermal and vegetation indices in different almond cultivars

    In recent years, the area dedicated to modern irrigated almond plantations has increased significantly in Spain. However, the legal irrigation allocations are lower than the maximum water requirements of the crop in most cases. Therefore, almond growers are forced to implement regulated deficit irrigation strategies on their farms, applying water stress in certain resistant phenological periods and avoiding it in sensitive periods. Given the need to monitor the water status of the crop, especially in the most sensitive periods to water stress, the objective of this work was to evaluate the sensitivity of two UAV-based crop water status indicators to detect early water stress conditions in four almond cultivars. The field trial was conducted during 2020 in an experimental almond orchard, where two irrigation strategies were established: full irrigation (FI), which received 100% of irrigation requirements (IR), and regulated deficit irrigation (RDI), which received 70% of IR during the whole irrigation period except during the kernel-filling stage when received 40% IR. The UAV flights were performed on four selected dates of the irrigation season. The Crop Water Status Index (CWSI) and the Normalized Difference Vegetation Index (NDVI) were derived from thermal and multispectral images, respectively, and compared to classical water status indicators, i.e., stem water potential (Ψstem ), stomatal conductance (gs ), and photosynthetic rate (AN ). Of the four flights performed, three corresponded to mild water stress conditions and a single flight was performed under moderate water stress conditions. Under mild water stress, CWSI was not able to capture the differences between FI and RDI trees that were observed with Ψstem . Under moderate stress conditions, CWSI was sensitive to the water deficit reached in the trees and showed significant differences among both irrigation treatments. No differences were observed in the CWSI and NVDI response to water stress among cultivars. Although NDVI and CWSI were sensitive to water stress, the low signal intensity observed in NDVI makes this index less robust than CWSI to monitor crop water stress. It can be concluded that UAV-based CWSI measurements are reliable to monitor almond water status, although for early (mild) levels of water stress, Ψstem seems to be the preferred option.Junta de Andalucía AVA.AVA2019.05

    Effect of the methacrylate-based endodontic sealer Epiphany on rat peritoneal macrophages viability

    Objective: To evaluate the effect of the endodontic sealer Epiphany on rat peritoneal macrophages viability. Materials and methods: Peritoneal macrophages were obtained from Wistar rats and resuspended in RPMI- 1640 medium. Undiluted (crude extract) and diluted extracts to 10%, 1%, 0.1%, 0.01%, 0.001% and 0.0001% of Epiphany, AH 26 and AH Plus sealers on RPMI-1640 medium were tested for cytotoxicity to rat peritoneal macrophages using the trypan blue dye exclusion assay. Data were analyzed statistically by the Kruskal-Wallis and Mann-Whitney tests at 5% significance level. Results: Crude extract of Epiphany killed 51% of cells, but was less cytotoxic that crude extracts of AH Plus and AH 26, which killed 81% and 86% of cells, respectively. Ten-fold dilutions of Epiphany, AH Plus and AH 26 killed 44%, 56%, 62% of macrophages, respectively. A hundred dilution of Epiphany only killed 7% of macrophages, but the same dilution of AH Plus and AH 26 killed 10% and 31% of macrophages, respectively. Lower dilutions of sealer extracts caused minimal cell death as compared to the control groups (p>0.05). Conclusions: The methacrylate-based endodontic sealer Epiphany showed lower cytotoxicity on macrophages than resin-based sealers AH Plus and AH 26.     Sleep-disordered breathing, circulating exosomes, and insulin sensitivity in adipocytes

    Background: Sleep-disordered-breathing (SDB), which is characterized by chronic intermittent hypoxia (IH) and sleep fragmentation (SF), is a prevalent condition that promotes metabolic dysfunction, particularly among patients suffering from obstructive hypoventilation syndrome (OHS). Exosomes are generated ubiquitously, are readily present in the circulation, and their cargo may exert substantial functional cellular alterations in both physiological and pathological conditions. However, the effects of plasma exosomes on adipocyte metabolism in patients with OHS or in mice subjected to IH or SF mimicking SDB are unclear. Methods: Exosomes from fasting morning plasma samples from obese adults with polysomnographically-confirmed OSA before and after 3 months of adherent CPAP therapy were assayed. In addition, C57BL/6 mice were randomly assigned to (1) sleep control (SC), (2) sleep fragmentation (SF), and (3) intermittent hypoxia (HI) for 6 weeks, and plasma exosomes were isolated. Equivalent exosome amounts were added to differentiated adipocytes in culture, after which insulin sensitivity was assessed using 0 nM and 5 nM insulin-induced pAKT/AKT expression changes by western blotting. Results: When plasma exosomes were co-cultured and internalized by human naive adipocytes, significant reductions emerged in Akt phosphorylation responses to insulin when compared to exosomes obtained after 24 months of adherent CPAP treatment (n = 24; p < 0.001), while no such changes occur in untreated patients (n = 8). In addition, OHS exosomes induced significant increases in adipocyte lipolysis that were attenuated after CPAP, but did not alter pre-adipocyte differentiation. Similarly, exosomes from SF- and IH-exposed mice induced attenuated p-AKT/total AKT responses to exogenous insulin and increased glycerol content in naive murine adipocytes, without altering pre-adipocyte differentiation. Conclusions: Using in vitro adipocyte-based functional reporter assays, alterations in plasma exosomal cargo occur in SDB, and appear to contribute to adipocyte metabolic dysfunction. Further exploration of exosomal miRNA signatures in either human subjects or animal models and their putative organ and cell targets appears warranted

    In vitro and in silico ADME-Tox profiling and safety significance of multifunctional monoamine oxidase inhibitors targeting neurodegenerative deseases

    Herein we report in vitro metabolic stability in human liver microsomes (HLMs), interactions with cytochrome P450 isoenzymes (CYP3A4, CYP2D6, and CYP2C9), and cytotoxicity analyses on HEK-293, HepG2, Huh7, and WTIIB cell lines of our most recent multitarget directed ligands PF9601N, ASS234, and contilisant. Based on these results, we conclude that (1) PF9601N and contilisant are metabolically stable in the HLM assay, in contrast to the very unstable ASS234; (2) CYP3A4 activity was decreased by PF9601N at all the tested concentrations and by ASS234 and contilisant only at the highest concentration; CYP2D6 activity was reduced by ASS234 at 1, 10, and 25 μM and by PF9601N at 10 and 25 μM, whereas contilisant increased its activity at the same concentrations; CYP2C9 was inhibited by the three compounds; (3) contilisant did not affect cell viability in the widest range of concentrations: up to 10 μM on HEK-293 cells, up to 30 μM on Huh7 cells, up to 50 μM on HepG2 cells, and up to 30 or 100 μM on WTIIB cells. Based on these results, we selected contilisant as a metabolically stable and nontoxic lead compound for further studies in Alzheimer's disease therapy.This study received financial support from the National Science Centre Poland (Grant No. 2016/23/D/NZ7/01328).
