    The Reduction of the Permeability of a Lateritic Soil through the Application of Microbially Induced Calcite Precipitation

    Lateritic soils are frequently utilised in tropical areas of the developing world as an engineering material in the construction of rural earth roads, usually in the form of engineered natural surface (ENS) roads. The heavy, seasonal rainfalls common to the tropics results in ENS roads becoming quickly saturated with rainwater, and no longer accessible to motorised transportation. Microbially induced calcite precipitation (MICP) has been successfully used as a treatment process to decrease the permeability of clean, cohesionless sands by studies trying to impede the movement of groundwater, and any pollutants they may contain. In order to see if MICP treatment can also reduce the susceptibility of ENS road lateritic soils to rainwater saturation, this study has treated a Brazilian sample extracted from an ENS road in Espirito do Santo, Brazil, using the MICP bacterium Sporosarcina pasteurii contained within a urea-calcium chloride solution inoculum. Investigation, by means of a Rowe cell, of the post-treatment permeability, to untreated control samples, has shown an average decrease in the vertical coefficient of permeability of 83%, from 1.15 × 10-7 m/s for the untreated control samples, to 1.92 × 10-8 m/s in treated samples

    Stakeholder influence on teaming and absorptive capacity in innovation networks

    Through technological developments, innovation increasingly occurs within a network of organizations such as Industry 4.0 fieldlabs. As a result, collaboration between different companies and institutions with different interests needs to take place. Three Dutch smart industry fieldlabs were analysed to study how these collaborative relationships are being established and what their impact is on the absorptive capacity of the network in question. Contrary to what was expected, we found that stakeholders hardly exercised power. Also, a high level of psychological safety was found in the network, which positively affects collaboration. Furthermore, collaborative elements—such as open conversation, collaborating, experimenting and reflecting—are important factors affecting the absorptive capacity in the fieldlabs examined.     Toenail Manganese: A Sensitive and Specific Biomarker of Exposure to Manganese in Career Welders

    Manganese (Mn) is an essential trace metal. It is also a component of welding fume. Chronic inhalation of manganese from welding fume has been associated with decreased neurological function. Currently, there is not a universally recognized biomarker for Mn exposure; however, hair and toenails have shown promise. In a cohort of 45 male welders and 35 age-matched factory control subjects, we assessed the sensitivity and specificity of toenail Mn to distinguish occupationally exposed subjects from unexposed controls. Further we examined the exposure time window that best correlates with the proposed biomarker, and investigated if non-occupational exposure factors impacted toenail Mn concentrations. Toenail clippings were analyzed for Mn using Inductively Coupled Plasma Mass Spectrometry (ICP-MS). Exposure to respirable Mn-containing particles (<4 µm) was estimated using an exposure model that combines personal air monitoring, work history information, and dietary intake to estimate an individual's exposure to Mn from inhalation of welding fume. We assessed the group differences in toenail concentrations using a Student's t-test between welders and control subjects and performed a receiver operating characteristic (ROC) curve analysis to identify a threshold in toenail concentration that has the highest sensitivity and specificity in distinguishing welders from control subjects. Additionally, we performed mixed-model regressions to investigate the association between different exposure windows and toenail Mn concentrations. We observed that toenail Mn concentrations were significantly elevated among welders compared to control subjects (6.87 ± 2.56 versus 2.70 ± 1.70 µg g-1; P < 0.001). Our results show that using a toenail Mn concentration of 4.14 µg g-1 as cutoff allows for discriminating between controls and welders with 91% specificity and 94% sensitivity [area under curve (AUC) = 0.98]. Additionally, we found that a threshold of 4.66 µg g-1 toenail Mn concentration enables a 90% sensitive and 90% specific discrimination (AUC = 0.96) between subjects with average exposure above or below the American Conference of Governmental Industrial Hygienist (ACGIH) Threshold Limit Value (TLV) of 0.02 mg m-3 during the exposure window of 7-12 months prior to the nail being clipped. Investigating which exposure window was best reflected by toenail Mn reproduced the result from another study of toenail Mn being significantly (P < 0.001) associated with exposure 7-12 months prior to the nail being clipped. Lastly, we found that dietary intake, body mass index, age, smoking status, and ethnicity had no significant effect on toenail Mn concentrations. Our results suggest that toenail Mn is a sensitive, specific, and easy-to-acquire biomarker of Mn exposure, which is feasible to be used in an industrial welder population

    Pharmaceutical care for elderly patients shared between community pharmacists and general practitioners: A randomised evaluation. RESPECT (Randomised Evaluation of Shared Prescribing for Elderly people in the Community over Time) [ISRCTN16932128]

    Background: This trial aims to investigate the effectiveness and cost implications of 'pharmaceutical care' provided by community pharmacists to elderly patients in the community. As the UK government has proposed that by 2004 pharmaceutical care services should extend nationwide, this provides an opportunity to evaluate the effect of pharmaceutical care for the elderly. Design: The trial design is a randomised multiple interrupted time series. We aim to recruit 700 patients from about 20 general practices, each associated with about three community pharmacies, from each of the five Primary Care Trusts in North and East Yorkshire. We shall randomise the five resulting groups of practices, pharmacies and patients to begin pharmaceutical care in five successive phases. All five will act as controls until they receive the intervention in a random sequence. Until they receive training community pharmacists will provide their usual dispensing services and so act as controls. The community pharmacists and general practitioners will receive training in pharmaceutical care for the elderly. Once trained, community pharmacists will meet recruited patients, either in their pharmacies (in a consultation room or dispensary to preserve confidentiality) or at home. They will identify drug-related issues/problems, and design a pharmaceutical care plan in conjunction with both the GP and the patient. They will implement, monitor, and update this plan monthly. The primary outcome measure is the 'Medication Appropriateness Index'. Secondary measures include adverse events, quality of life, and patient knowledge and compliance. We shall also investigate the cost of pharmaceutical care to the NHS, to patients and to society as a whole.published_or_final_versio

    Galaxy Zoo: Disentangling the Environmental Dependence of Morphology and Colour

    We analyze the environmental dependence of galaxy morphology and colour with two-point clustering statistics, using data from the Galaxy Zoo, the largest sample of visually classified morphologies yet compiled, extracted from the Sloan Digital Sky Survey. We present two-point correlation functions of spiral and early-type galaxies, and we quantify the correlation between morphology and environment with marked correlation functions. These yield clear and precise environmental trends across a wide range of scales, analogous to similar measurements with galaxy colours, indicating that the Galaxy Zoo classifications themselves are very precise. We measure morphology marked correlation functions at fixed colour and find that they are relatively weak, with the only residual correlation being that of red galaxies at small scales, indicating a morphology gradient within haloes for red galaxies. At fixed morphology, we find that the environmental dependence of colour remains strong, and these correlations remain for fixed morphology \textit{and} luminosity. An implication of this is that much of the morphology--density relation is due to the relation between colour and density. Our results also have implications for galaxy evolution: the morphological transformation of galaxies is usually accompanied by a colour transformation, but not necessarily vice versa. A spiral galaxy may move onto the red sequence of the colour-magnitude diagram without quickly becoming an early-type. We analyze the significant population of red spiral galaxies, and present evidence that they tend to be located in moderately dense environments and are often satellite galaxies in the outskirts of haloes. Finally, we combine our results to argue that central and satellite galaxies tend to follow different evolutionary paths.Comment: 19 pages, 18 figures. Accepted for publication in MNRA

    A Neural Circuit Arbitrates between Persistence and Withdrawal in Hungry Drosophila

    In pursuit of food, hungry animals mobilize significant energy resources and overcome exhaustion and fear. How need and motivation control the decision to continue or change behavior is not understood. Using a single fly treadmill, we show that hungry flies persistently track a food odor and increase their effort over repeated trials in the absence of reward suggesting that need dominates negative experience. We further show that odor tracking is regulated by two mushroom body output neurons (MBONs) connecting the MB to the lateral horn. These MBONs, together with dopaminergic neurons and Dop1R2 signaling, control behavioral persistence. Conversely, an octopaminergic neuron, VPM4, which directly innervates one of the MBONs, acts as a brake on odor tracking by connecting feeding and olfaction. Together, our data suggest a function for the MB in internal state-dependent expression of behavior that can be suppressed by external inputs conveying a competing behavioral drive

    Agave negatively regulates YAP and TAZ transcriptionally and post-translationally in osteosarcoma cell lines

    Osteosarcoma (OS) is the most aggressive type of primary solid tumor that develops in bone. Whilst conventional chemotherapy can improve survival rates, the outcome for patients with metastatic or recurrent OS remains poor, so novel treatment agents and strategies are required. Research into new anticancer therapies has paved the way for the utilisation of natural compounds as they are typically less expensive and less toxic compared to conventional chemotherapeutics. Previously published works indicate that Agave exhibits anticancer properties, however potential molecular mechanisms remain poorly understood. In the present study, we investigate the anticancer effects of Agave leaf extract in OS cells suggesting that Agave inhibits cell viability, colony formation, and cell migration, and can induce apoptosis in OS cell lines. Moreover, Agave sensitizes OS cells to cisplatin (CDDP) and radiation, to overcome chemo- and radio-resistance. We demonstrate that Agave extract induces a marked decrease of Yes Associated Protein (YAP) and Tafazzin (TAZ) mRNA and protein expression upon treatment. We propose an initial mechanism of action in which Agave induces YAP/TAZ protein degradation, followed by a secondary event whereby Agave inhibits YAP/TAZ transcription, effectively deregulating the Nuclear Factor kappa B (NF-\u3baB) p65:p50 heterodimers responsible for transcriptional induction of YAP and TAZ
