    Astroglial-axonal interactions during early stages of myelination in mixed cultures using in vitro and ex vivo imaging techniques

    <b>Background</b><p></p> Myelination is a very complex process that requires the cross talk between various neural cell types. Previously, using cytosolic or membrane associated GFP tagged neurospheres, we followed the interaction of oligodendrocytes with axons using time-lapse imaging in vitro and ex vivo and demonstrated dynamic changes in cell morphology. In this study we focus on GFP tagged astrocytes differentiated from neurospheres and their interactions with axons.<p></p> <b>Results</b><p></p> We show the close interaction of astrocyte processes with axons and with oligodendrocytes in mixed mouse spinal cord cultures with formation of membrane blebs as previously seen for oligodendrocytes in the same cultures. When GFP-tagged neurospheres were transplanted into the spinal cord of the dysmyelinated shiverer mouse, confirmation of dynamic changes in cell morphology was provided and a prevalence for astrocyte differentiation compared with oligodendroglial differentiation around the injection site. Furthermore, we were able to image GFP tagged neural cells in vivo after transplantation and the cells exhibited similar membrane changes as cells visualised in vitro and ex vivo.<p></p> <b>Conclusion</b><p></p> These data show that astrocytes exhibit dynamic cell process movement and changes in their membrane topography as they interact with axons and oligodendrocytes during the process of myelination, with the first demonstration of bleb formation in astrocytes

    In all dimensions and arbitrary signature, we demonstrate the existence of a new local potential -- a double (2,3)-form -- for the Weyl curvature tensor, and more generally for all tensors with the symmetry properties of the Weyl curvature tensor.     Upon notification, in 1995, that a portion of a Spanish Colonial acequia had been exposed in Brackenridge Park, the Parks and Recreation Department of the City of San Antonio entered into a contract with the Center for Archaeological Research (CAR) of the University of Texas at San Antonio to investigate and document the structure. In September of 1996, a team from CAR, utilizing a backhoe and operator provided by Parks and Recreation, began a limited investigation of the structure. The structure proved to be the dam for the Upper Labor and displayed two distinct construction periods, Spanish Colonial and post -1860. It was further documented that the structure acted as a diversion device rather than a full empoundment structure

    Socially-mediated arousal and contagion within domestic chick broods

    Emotional contagion – an underpinning valenced feature of empathy – is made up of simpler, potentially dissociable social processes which can include socially-mediated arousal and behavioural/physiological contagion. Previous studies of emotional contagion have often conflated these processes rather than examining their independent contribution to empathic response. We measured socially-mediated arousal and contagion in 9-week old domestic chicks (n = 19 broods), who were unrelated but raised together from hatching. Pairs of observer chicks were exposed to two conditions in a counterbalanced order: air puff to conspecifics (AP) (during which an air puff was applied to three conspecifics at 30 s intervals) and control with noise of air puff (C) (during which the air puff was directed away from the apparatus at 30 s intervals). Behaviour and surface eye temperature of subjects and observers were measured throughout a 10-min pre-treatment and 10-min treatment period. Subjects and observers responded to AP with increased freezing, and reduced preening and ground pecking. Subjects and observers also showed reduced surface eye temperature - indicative of stress-induced hyperthermia. Subject-Observer behaviour was highly correlated within broods during both C and AP conditions, but with higher overall synchrony during AP.     Solid catalysts obtained from wastes for FAME production using mixtures of refined palm oil and waste cooking oils

    More than 95% of biodiesel production feedstocks come from edible oils, however it may cause some problems such as the competition of land use between food production and biodiesel production. The waste cooking oils (WCO) are an alternative feedstock for biodiesel production; its usage reduces significantly the cost of biodiesel production and has environmental benefits, e.g., a waste recovery instead of its elimination. This work aims to produce a low-cost efficient solid catalyst for fatty acid methyl esters (FAME) production using mixtures of refined palm oil (RPO) and WCO. Four low cost catalysts were prepared (biomass fly ashes, natural dolomite rock, chicken eggshells and polyethylene terephthalate - PET), characterized (by SEM, EDX, XRD, BET, FT-IR and Hammett indicators) and tested regarding their performance in FAME production. The maximum yield of FAME achieved was around 96%wt. for biomass fly ashes catalyst at 60 °C, 9:1 (mol/mol) of methanol to oil mixture, 10%wt. catalyst to oil mixture, over 180 min in batch reactor. The results point out for promising bifunctional catalysts able to achieve also conversion of free fatty acids up to 100% using mixtures of RPO and WCO.publishe

    Demyelination and axonal preservation in a transgenic mouse model of Pelizaeus-Merzbacher disease

    It is widely thought that demyelination contributes to the degeneration of axons and, in combination with acute inflammatory injury, is responsible for progressive axonal loss and persistent clinical disability in inflammatory demyelinating disease. In this study we sought to characterize the relationship between demyelination, inflammation and axonal transport changes using a Plp1-transgenic mouse model of Pelizaeus-Merzbacher disease. In the optic pathway of this non-immune mediated model of demyelination, myelin loss progresses from the optic nerve head towards the brain, over a period of months. Axonal transport is functionally perturbed at sites associated with local inflammation and 'damaged' myelin. Surprisingly, where demyelination is complete, naked axons appear well preserved despite a significant reduction of axonal transport.     Optimization of FAME production from blends of waste cooking oil and refined palm oil using biomass fly ash as a catalyst

    One of the problems associated with biomass combustion is the amount of fly ashes generated and its subsequent management. The search for ways of valorizing these ashes has been a challenge for the academic and industrial community. On the other hand, used cooking oils are wastes which management is quite difficult, by they have a very important energetic potential. The goal of this work was to optimize the Fatty Acid Methyl Esters (FAME) process, recovering two residual materials (waste cooking oils (WCO), and biomass fly flash (BFA)). The optimization of the process was achieved using the response surface methodology and a Box-Benhken experimental design applied to mixtures of WCO and refined palm oil (RPO), using BFA as catalyst. The influence on FAME yield of four variables (catalyst loading, methanol/oil molar ratio, RPO/WCO ratio and reaction temperature) was studied. The higher FAME yield achieved was 73.8% for the following operating conditions: 13.57 wt% of catalyst loading, 6.7 of methanol/oil molar ratio, 28.04 wt% of RPO in the oil mixture with WCO and 55 °C for the reaction temperature. The reusability of the BFA catalyst in the process was also studied through three successive usage cycles finding no loss of catalytic activity.publishe

    Participating in a fruit and vegetable intervention trial improves longer term fruit and vegetable consumption and barriers to fruit and vegetable consumption: A follow-up of the ADIT study

    Background: Fruit and vegetable (FV) based intervention studies can be effective in increasing short term FV consumption. However, the longer term efficacy of such interventions is still unclear. The aim of the current study was to examine the maintenance of change in FV consumption 18-months after cessation of a FV intervention and to examine the effect of participating in a FV intervention on barriers to FV consumption. Methods: A follow-up of a randomised controlled FV trial in 83 older adults (habitually consuming ≤2 portions/day) was conducted. At baseline, participants were assigned to continue consuming ≤2 portions FV/day or consume ≥5 portions FV/day for 16-weeks. We assessed FV intake and barriers to FV consumption at baseline, end of intervention and 18-months post-intervention. Results: At 18-months, mean FV intakes in both groups were greater than baseline. The 5 portions/day group continued to show greater increases in FV consumption at 18-months than the 2 portions/day group (p < 0.01). At 18-months, both groups reported greater liking (p < 0.01) and ease in consuming FV (p = 0.001) while difficulties with consuming FV decreased (p < 0.001). The 2 portions/day group reported greater awareness of FV recommendations at 18-months (p < 0.001). Conclusions: Participating in a FV intervention can lead to longer-term positive changes in FV consumption regardless of original group allocation.     Invariant construction of solutions to Einstein's field equations - LRS perfect fluids II

    The properties of LRS class II perfect fluid space-times are analyzed using the description of geometries in terms of the Riemann tensor and a finite number of its covariant derivatives. In this manner it is straightforward to obtain the plane and hyperbolic analogues to the spherical symmetric case. For spherically symmetric static models the set of equations is reduced to the Tolman-Oppenheimer-Volkoff equation only. Some new non-stationary and inhomogeneous solutions with shear, expansion, and acceleration of the fluid are presented. Among these are a class of temporally self-similar solutions with equation of state given by p=(γ1)μ,1<γ<2p=(\gamma-1)\mu, 1<\gamma<2, and a class of solutions characterized by σ=Θ/6\sigma=-\Theta/6. We give an example of geometry where the Riemann tensor and the Ricci rotation coefficients are not sufficient to give a complete description of the geometry. Using an extension of the method, we find the full metric in terms of curvature quantities.Comment: 24 pages, 1 figur

    On the structure of the new electromagnetic conservation laws

    New electromagnetic conservation laws have recently been proposed: in the absence of electromagnetic currents, the trace of the Chevreton superenergy tensor, HabH_{ab} is divergence-free in four-dimensional (a) Einstein spacetimes for test fields, (b) Einstein-Maxwell spacetimes. Subsequently it has been pointed out, in analogy with flat spaces, that for Einstein spacetimes the trace of the Chevreton superenergy tensor HabH_{ab} can be rearranged in the form of a generalised wave operator L\square_L acting on the energy momentum tensor TabT_{ab} of the test fields, i.e., Hab=LTab/2H_{ab}=\square_LT_{ab}/2. In this letter we show, for Einstein-Maxwell spacetimes in the full non-linear theory, that, although, the trace of the Chevreton superenergy tensor HabH_{ab} can again be rearranged in the form of a generalised wave operator G\square_G acting on the electromagnetic energy momentum tensor, in this case the result is also crucially dependent on Einstein's equations; hence we argue that the divergence-free property of the tensor Hab=GTab/2H_{ab}=\square_GT_{ab}/2 has significant independent content beyond that of the divergence-free property of TabT_{ab}
