    Production of triploid Sandersonia aurantiaca plants

    Triploid plants of Sandersonia aurantiaca were produced by crossing diploid and tetraploid forms of S. aurantiaca. Enlarged ovules were transferred to in vitro culture 14–30 days after pollination. The triploid nature of the embryo derived plants was determined by flow cytometry and chromosome counts both of which showed that the triploid plants had features that were midway between those of the two parents. The mean nuclear DNA contents of 2C nuclei from diploid, triploid and tetraploid forms of S. aurantiaca were 6.86pg, 10.04pg and 13.55pg, respectively. The nuclear DNA content of 1C nuclei of sperm cells from pollen grains was 2.94pg. Mitotic chromosome counts from the three plants gave 2n = 24, 36 and 48 chromosomes for the diploid, triploid and tetraploid forms, respectively. Meiotic chromosome counts for the diploid and tetraploid plants were n = 12 and n = 24, respectively. The triploid showed mainly bivalents, but lagging chromosomes led to micronuclei and infertility in gametes. The morphological features of the various plants corroborated other evidence indicating that the triploid plants were the result of a cross between diploid and tetraploid plants

    Human papillomavirus 16 L2 inhibits the transcriptional activation function, but not the DNA replication function, of HPV-16 E2

    In this study we analysed the outcome of the interaction between HPV-16 L2 and E2 on the transactivation and DNA replication functions of E2. When E2 was expressed on its own, it transactivated a number of E2-responsive promoters but co-expression of L2 led to the down-regulation of the transcription transactivation activity of the E2 protein. This repression is not mediated by an increased degradation of the E2 protein. In contrast, the expression of L2 had no effect on the ability of E2 to activate DNA replication in association with the viral replication factor E1. Deletion mutagenesis identified L2 domains responsible for binding to E2 (first 50 N-terminus amino acid residues) and down-regulating its transactivation function (residues 301–400). The results demonstrate that L2 selectively inhibits the transcriptional activation property of E2 and that there is a direct interaction between the two proteins, although this is not sufficient to mediate the transcriptional repression. The consequences of the L2–E2 interaction for the viral life cycle are discussed

    On subducting slab entrainment of buoyant asthenosphere

    Laboratory and numerical experiments and boundary layer analysis of the entrainment of buoyant asthenosphere by subducting oceanic lithosphere implies that slab entrainment is likely to be relatively inefficient at removing a buoyant and lower viscosity asthenosphere layer. Asthenosphere would instead be mostly removed by accretion into and eventual subduction of the overlying oceanic lithosphere. The lower (hot) side of a subducting slab entrains by the formation of a ∼10–30 km-thick downdragged layer, whose thickness depends upon the subduction rate and the density contrast and viscosity of the asthenosphere, while the upper (cold) side of the slab may entrain as much by thermal 'freezing' onto the slab as by mechanical downdragging. This analysis also implies that proper treatment of slab entrainment in future numerical mantle flow experiments will require the resolution of ∼10–30 km-thick entrainment boundary layers

    Cognitive rehabilitation for attention and memory in people with multiple sclerosis: study protocol for a randomised controlled trial (CRAMMS)

    Background People with multiple sclerosis have problems with memory and attention. Cognitive rehabilitation is a structured set of therapeutic activities designed to retrain an individual’s memory and other cognitive functions. Cognitive rehabilitation may be provided to teach people strategies to cope with these problems, in order to reduce the impact on everyday life. The effectiveness of cognitive rehabilitation for people with multiple sclerosis has not been established. Methods This is a multi-centre, randomised controlled trial investigating the clinical and cost-effectiveness of a group-based cognitive rehabilitation programme for attention and memory problems for people with multiple sclerosis. Four hundred people with multiple sclerosis will be randomised from at least four centres. Participants will be eligible if they have memory problems, are 18 to 69 years of age, are able to travel to attend group sessions and give informed consent. Participants will be randomised in a ratio of 6:5 to the group rehabilitation intervention plus usual care or usual care alone. Intervention groups will receive 10 weekly sessions of a manualised cognitive rehabilitation programme. The intervention will include both restitution strategies to retrain impaired attention and memory functions and compensation strategies to enable participants to cope with their cognitive problems. All participants will receive a follow-up questionnaire and an assessment by a research assistant at 6 and 12 months after randomisation. The primary outcome is the Multiple Sclerosis Impact Scale (MSIS) Psychological subscale at 12 months. Secondary outcomes include the Everyday Memory Questionnaire, General Health Questionnaire-30, EQ-5D and a service use questionnaire from participants, and the Everyday Memory Questionnaire-relative version and Carer Strain Index from a relative or friend. The primary analysis will be based on intention to treat. A mixed-model regression analysis of the MSIS Psychological subscale at 12 months will be used to estimate the effect of the group cognitive rehabilitation programme. Discussion The study will provide evidence regarding the clinical and cost-effectiveness of a group-based cognitive rehabilitation programme for attention and memory problems in people with multiple sclerosis.     Lombardi Drawings of Graphs

    We introduce the notion of Lombardi graph drawings, named after the American abstract artist Mark Lombardi. In these drawings, edges are represented as circular arcs rather than as line segments or polylines, and the vertices have perfect angular resolution: the edges are equally spaced around each vertex. We describe algorithms for finding Lombardi drawings of regular graphs, graphs of bounded degeneracy, and certain families of planar graphs.Comment: Expanded version of paper appearing in the 18th International Symposium on Graph Drawing (GD 2010). 13 pages, 7 figure

    Structure and Magnetization of Two-Dimensional Vortex Arrays in the Presence of Periodic Pinning

    Ground-state properties of a two-dimensional system of superconducting vortices in the presence of a periodic array of strong pinning centers are studied analytically and numerically. The ground states of the vortex system at different filling ratios are found using a simple geometric argument under the assumption that the penetration depth is much smaller than the spacing of the pin lattice. The results of this calculation are confirmed by numerical studies in which simulated annealing is used to locate the ground states of the vortex system. The zero-temperature equilibrium magnetization as a function of the applied field is obtained by numerically calculating the energy of the ground state for a large number of closely spaced filling ratios. The results show interesting commensurability effects such as plateaus in the B-H diagram at simple fractional filling ratios.Comment: 12 pages, 19 figures, submitted for publicatio
