    Experimental evaluation on the applicability of necrobiome analysis in forensic veterinary science

    Despite the wide usage of animals as models in forensic studies, the investigations of fundamental legal questions involving domesticated and nondomesticated animals were always given marginal attention compared to "human forensic," and only recently the interest in the discipline is increasing. Our research focuses on the effect of the fur coat on the activity and development of microbial decomposers. In order to test this variable never assessed before, rabbit carcasses were used and results show that: (i) distinct and significant temporal changes in terms of metabolic activity and taxa distribution can be tracked over the decomposition process; (ii) the richness and the diversity of the bacterial communities does not significantly vary over time, but it does not mean that the species Operational Taxonomic Units (OTUs) do not change; (iii) the presence/absence of the fur on the carcasses does not significantly affect either the bacterial communities' functional activity or the diversity intra- and intercommunity, neither at phylum nor at family resolution; (iv) the functional activity and the ecological diversity of the bacterial communities are significantly affected by the body region, while the relative abundance is not. Obtained data confirm previous observations and provide new insight in the Forensic Veterinary field in terms of equally using them in order to derive a statistical model for the PMI estimation. As a future perspective, a contribution to the Forensic Entomology approach will be given in legal investigations when domestic or wild animals are involved, regardless of the presence of a hair layer

    Unexpected relationships between four large deletions in the human β-globin gene cluster.

    Two independent gamma delta beta-thalassemias are each associated with large deletions. We show, by comparing DNA sequences, that the deletions are due to non-homologous DNA exchanges. The 5' breakpoints are located approximately the same distance apart and in the same order along the DNA as their 3' breakpoints. Two independent cases of hereditary persistence of fetal hemoglobin, also involving large deletions, show the same unexpected relationship between their 5' and 3' breakpoints. This relationship is most simply explained if, within each pair, the deletions are of approximately the same length. The results suggest that the four deletions were generated by a common mechanism. Perhaps their 5' and 3' breakpoints are physically close in the nucleus, although far apart on the linear DNA

    The DNA spanning two large deletions in the human beta-globin gene cluster (gamma beta-thalassaemia 1 and 2) has been cloned by cosmid cloning and chromosomal walking. The entire region was mapped and analyzed for the presence of repetitive sequences. The results show that the affected loci have lost almost 100 kb of DNA in a deletion event not involving homologous or repetitive sequences

    Estudio de foraminífiros bentónicos en escala mensual en una bahía subtropical moderadamente afectada por efluentes urbanos

    Benthic foraminifera were sampled monthly during a one-year period in order to examine their biological response to the environmental factors in the Ubatuba Bay (northern coast of São Paulo State, Brazil). The area is a popular tourist destination with a population that varies during the year, as does the untreated sewage carried into the bay by the rivers. Four sites were analysed. Each station is near one of the rivers that discharge into the bay. Biological data were analysed with multivariate and univariate techniques. The influence of the abiotic parameters on the foraminiferal fauna was inferred through statistical methods and time correlation functions. Geochemical and populational parameter differences allowed the sites to be separated into two groups according to their stress conditions. One group was dominated by Ammonia tepida—a herbivore species—and showed higher densities indicating a more productive and less contaminated location. The other one was dominated by Buliminella elegantissima—a detritivore species. Species diversity did not seem to be a good indicator of environmental health in this area due to the low densities and the high dominance of few species. Nevertheless, density and richness were used as evidence of local productivity and environmental conditions. Quasi-azoic moments related to the high degree of contamination were observed. Anthropogenic effects were stronger in the austral summer period, when sewage input through the rivers increases due to mass tourism.Fueron analizados foraminíferos bentónicos en cuatro estaciones muestreadas mensualmente con la intención de investigar la respuesta biológica a los factores ambientales en la Ensenada de Ubatuba (costa noreste de São Paulo, Brasil). El área es un importante local turístico cuya población fluctúa a través del año y recibe efluentes no tratados a través de los ríos que en ella desembocan. Fueron analizados cuatro locales. Cada estación se encuentra localizada próxima a uno de los cuatro ríos que desembocan en la bahía. Los datos biológicos fueron analizados con técnicas uni y multivariadas. La influencia de los parámetros abióticos sobre la fauna de foraminíferos fue inferida a través de métodos estadísticos y de funciones de correlación temporal. Parámetros geoquímicas y poblacionales permitieron distinguir dos grupos de estaciones que reflejan las condiciones de estrés local. Uno de los grupos estuvo dominado por Ammonia tepida –especie herbívora– y se mostró más productivo y menos contaminado. El otro local estuvo dominado por Buliminella elegantísima –especie detritívora. La diversidad especifica no se mostró un buen indicador de la salud ambiental en esta región debido a las bajas densidades y a la alta dominancia de unas pocas especies. Sin embargo, la densidad y riqueza pudieron ser utilizadas como evidencias de la productividad local y condiciones ambientales. Fueron observados momentos casi azoicos relacionados con el alto grado de contaminación. Los efectos antropogénicos fueron mayores durante el período de verano austral, cuando la entrada de deshechos domésticos a través de los ríos aumenta como consecuencia del incremento del turismo

    Creation of Structural Polymer Composite Materials for Functional Application Using Physicochemical Modification

    The various aspects of physico-chemical modification of the components of structural materials of functional application based on classical composites and nanocomposites are analyzed. Potential applications of such materials are briefly described. Ultrasonic cavitation treatment is considered as a basic method of physical modification when obtaining the indicated classes of composites. The influence of ultrasonic treatment modes on the technological and operational properties of reactoplastic polymers, as well as on the hardening of reinforced composites based on them, is investigated. Technical means of ultrasonic cavitation processing of liquid binders and polymer composites based on them are briefly described. An effective spectrum of interrelated structural and technological parameters of ultrasonic treatment has been characterized, which is established by calculation and experimentally-statistically. The design issues of the technological processes of obtaining polymer composites of functional application are analyzed. The efficiency of creating carbon fiber composite materials, as well as the prospects for creating of these materials based on reinforcing fabric with nanomodified fillers is described. The methods of obtaining functional nanomodified carbon-composites with improved physicomechanical and operational properties, in particular with increased strength and electrical conductivity, are characterized. The effectiveness of the ultrasonic treatment and production of nanomodified thermoplastic composite materials by extrusion method is considered. Some issues of forming products from intelligent polymer composites are analyzed. The results of the survey can be used in the design of advanced technologies for the creation of functional polymer composites of functional application
