NGC1052 - A study of the pc-scale twin jet
We present results of a VLBA multi-frequency study of the pc-scale twin jet
in NGC1052. We observed this object at epoch 1998.99 with the VLBA at 5, 8.4,
22 and 43 GHz both in total and linearly polarized intensity. The spectral
analysis confirms the necessity of a free-free absorbing medium, obscuring the
innermost part of both jets. At 5 GHz we found a compact linearly polarized
emission region at the base of the eastern jet with a degree of polarization of
1.5%. At higher frequencies there is no evidence for polarization in our data.
A core shift analysis constrains the position of the central engine to ~0.03
pc. The shift rates of the apparent core position with frequency confirm the
strong influence of free-free absorption in conjunction with steep pressure

A genomic analysis and transcriptomic atlas of gene expression in Psoroptes ovis reveals feeding- and stage-specific patterns of allergen expression
Background: Psoroptic mange, caused by infestation with the ectoparasitic mite, Psoroptes ovis, is highly contagious, resulting in intense pruritus and represents a major welfare and economic concern for the livestock industry Worldwide. Control relies on injectable endectocides and organophosphate dips, but concerns over residues, environmental contamination, and the development of resistance threaten the sustainability of this approach, highlighting interest in alternative control methods. However, development of vaccines and identification of chemotherapeutic targets is hampered by the lack of P. ovis transcriptomic and genomic resources.
Results: Building on the recent publication of the P. ovis draft genome, here we present a genomic analysis and transcriptomic atlas of gene expression in P. ovis revealing feeding- and stage-specific patterns of gene expression, including novel multigene families and allergens. Network-based clustering revealed 14 gene clusters demonstrating either single- or multi-stage specific gene expression patterns, with 3075 female-specific, 890 male-specific and 112, 217 and 526 transcripts showing larval, protonymph and tritonymph specific-expression, respectively. Detailed analysis of P. ovis allergens revealed stage-specific patterns of allergen gene expression, many of which were also enriched in "fed" mites and tritonymphs, highlighting an important feeding-related allergenicity in this developmental stage. Pair-wise analysis of differential expression between life-cycle stages identified patterns of sex-biased gene expression and also identified novel P. ovis multigene families including known allergens and novel genes with high levels of stage-specific expression.
Conclusions: The genomic and transcriptomic atlas described here represents a unique resource for the acarid-research community, whilst the OrcAE platform makes this freely available, facilitating further community-led curation of the draft P. ovis genome
Additively Manufactured Graphitic Electrochemical Sensing Platforms
Additive manufacturing (AM)/3D printing technology provides a novel platform for the rapid prototyping of low cost 3D platforms. Herein, we report for the first time, the fabrication, characterisation (physicochemical and electrochemical) and application (electrochemical sensing) of bespoke nanographite (NG)-loaded (25 wt. %) AM printable (via fused deposition modelling) NG/PLA filaments. We have optimised and tailored a variety of NG-loaded filaments and their AM counterparts in order to achieve optimal printability and electrochemical behaviour. Two AM platforms, namely AM macroelectrodes (AMEs) and AM 3D honeycomb (macroporous) structures are benchmarked against a range of redox probes and the simultaneous detection of lead (II) and cadmium (II). A nano-indentation study of the contact resistance and resistivity of a bi-layered Au/multi-walled carbon nanotube composite
A bi-layer metal-carbon nanotube composite has been developed as a potential low-force electrical contact surface, for application in micro-electromechanical systems switching devices. The samples consist of a vertically aligned forest of multi-walled carbon nanotube (MWCNT), sputter coated with a layer of Au. The effect of varying the components and composition are investigated by means of a modified nano-indenter. By measuring the contact resistance of the composites under various loading conditions, the electrical properties and performance can be evaluated. The composites are shown to have homogenous properties, with each of the layers influencing the total electrical characteristics of the samples. The internal structure of the sample, the MWCNT height and penetration of gold into the forest is shown to directly influence the performance and characteristics of the samples. By analyzing the samples as bulk, the effective resistivities of the composites are also determined to have values from 303 n? m down to 54 n? m, depending on the composition of the sample

Metabolic correlates of prevalent mild cognitive impairment and Alzheimer's disease in adults with Down syndrome.
IntroductionDisruption of metabolic function is a recognized feature of late onset Alzheimer's disease (LOAD). We sought to determine whether similar metabolic pathways are implicated in adults with Down syndrome (DS) who have increased risk for Alzheimer's disease (AD).MethodsWe examined peripheral blood from 292 participants with DS who completed baseline assessments in the Alzheimer's Biomarkers Consortium-Down Syndrome (ABC-DS) using untargeted mass spectrometry (MS). Our sample included 38 individuals who met consensus criteria for AD (DS-AD), 43 who met criteria for mild cognitive impairment (DS-MCI), and 211 who were cognitively unaffected and stable (CS).ResultsWe measured relative abundance of 8,805 features using MS and 180 putative metabolites were differentially expressed (DE) among the groups at false discovery rate-corrected q< 0.05. From the DE features, a nine-feature classifier model classified the CS and DS-AD groups with receiver operating characteristic area under the curve (ROC AUC) of 0.86 and a two-feature model classified the DS-MCI and DS-AD groups with ROC AUC of 0.88. Metabolite set enrichment analysis across the three groups suggested alterations in fatty acid and carbohydrate metabolism.DiscussionOur results reveal metabolic alterations in DS-AD that are similar to those seen in LOAD. The pattern of results in this cross-sectional DS cohort suggests a dynamic time course of metabolic dysregulation which evolves with clinical progression from non-demented, to MCI, to AD. Metabolomic markers may be useful for staging progression of DS-AD
Automated prediction of mastitis infection patterns in dairy herds using machine learning
© 2020, The Author(s). Mastitis in dairy cattle is extremely costly both in economic and welfare terms and is one of the most significant drivers of antimicrobial usage in dairy cattle. A critical step in the prevention of mastitis is the diagnosis of the predominant route of transmission of pathogens into either contagious (CONT) or environmental (ENV), with environmental being further subdivided as transmission during either the nonlactating “dry” period (EDP) or lactating period (EL). Using data from 1000 farms, random forest algorithms were able to replicate the complex herd level diagnoses made by specialist veterinary clinicians with a high degree of accuracy. An accuracy of 98%, positive predictive value (PPV) of 86% and negative predictive value (NPV) of 99% was achieved for the diagnosis of CONT vs ENV (with CONT as a “positive” diagnosis), and an accuracy of 78%, PPV of 76% and NPV of 81% for the diagnosis of EDP vs EL (with EDP as a “positive” diagnosis). An accurate, automated mastitis diagnosis tool has great potential to aid non-specialist veterinary clinicians to make a rapid herd level diagnosis and promptly implement appropriate control measures for an extremely damaging disease in terms of animal health, productivity, welfare and antimicrobial use
The landscape of RNA polymerase II transcription initiation in C. elegans reveals promoter and enhancer architectures
RNA polymerase transcription initiation sites are largely unknown in Caenorhabditis elegans. The initial 5′ end of most protein-coding transcripts is removed by trans-splicing, and noncoding initiation sites have not been investigated. We characterized the landscape of RNA Pol II transcription initiation, identifying 73,500 distinct clusters of initiation. Bidirectional transcription is frequent, with a peak of transcriptional pairing at 120 bp. We assign transcription initiation sites to 7691 protein-coding genes and find that they display features typical of eukaryotic promoters. Strikingly, the majority of initiation events occur in regions with enhancer-like chromatin signatures. Based on the overlap of transcription initiation clusters with mapped transcription factor binding sites, we define 2361 transcribed intergenic enhancers. Remarkably, productive transcription elongation across these enhancers is predominantly in the same orientation as that of the nearest downstream gene. Directed elongation from an upstream enhancer toward a downstream gene could potentially deliver RNA polymerase II to a proximal promoter, or alternatively might function directly as a distal promoter. Our results provide a new resource to investigate transcription regulation in metazoans
Black Mantle Tissue of Endolithic Mussels (Leiosolenus spp.) Is Cloaking Borehole Orifices in Caribbean Reef Corals
Bioerosion caused by boring mussels (Mytilidae: Lithophaginae) can negatively impact coral reef health. During biodiversity surveys of coral-associated fauna in Curaçao (southern Caribbean), morphological variation in mussel boreholes was studied. Borings were found in 22 coral species, 12 of which represented new host records. Dead corals usually showed twin siphon openings, for each mussel shaped like a figure of eight, which were lined with a calcareous sheath and protruded as tubes from the substrate surface. Most openings surrounded by live coral tissue were deeper and funnel-shaped, with outlines resembling dumbbells, keyholes, ovals or irregular ink blotches. The boreholes appeared to contain black siphon and mantle tissue of the mussel. Because of the black color and the hidden borehole opening in live host corals, the mantle tissue appeared to mimic dark, empty holes, while they were actually cloaking live coral tissue around the hole, which is a new discovery. By illustrating the morphological range of borehole orifices, we aim to facilitate the easy detection of boring mussels for future research
