    The common truncation variant in pancreatic lipase related protein 2 (PNLIPRP2) is expressed poorly and does not alter risk for chronic pancreatitis

    A nonsense variant (p.W358X) of human pancreatic lipase related protein 2 (PNLIPRP2) is present in different ethnic populations with a high allele frequency. In cell culture experiments, the truncated protein mainly accumulates inside the cells and causes endoplasmic reticulum stress. Here, we tested the hypothesis that variant p.W358X might increase risk for chronic pancreatitis through acinar cell stress. We sequenced exon 11 of PNLIPRP2 in a cohort of 256 subjects with chronic pancreatitis (152 alcoholic and 104 non-alcoholic) and 200 controls of Hungarian origin. We observed no significant difference in the distribution of the truncation variant between patients and controls. We analyzed mRNA expression in human pancreatic cDNA samples and found the variant allele markedly reduced. We conclude that the p.W358X truncation variant of PNLIPRP2 is expressed poorly and has no significant effect on the risk of chronic pancreatitis

    Should We Be Confident in Published Research? A Case Study of Confidence Interval Reporting in Health Education and Behavior Research

    Confidence intervals (CIs) have been highlighted as “the best” reporting device when reporting statistical findings. However, researchers often fail to maximize the utility of CIs in research. We seek to (a) present a primer on CIs; (b) outline reporting practices of health researchers; and (c) discuss implications for statistical best practice reporting in social science research. Approximately 1,950 peer-reviewed articles were examined from six health education, promotion, and behavior journals. We recorded: (a) whether the author(s) reported a CI; (b) whether the author(s) reported a CI estimate width, either numerical or visual; and (c) whether an associated effect size was reported alongside the CI. Of the 1,245 quantitative articles in the final sample, 46.5% (n = 580) reported confidence interval use; , and 518 provided numerical/visual interval estimates. Of the articles reporting CIs, 383 (64.2%) articles reported a CI with an associated effect size, meeting the American Psychological Association’s (APA) recommendation for statistical reporting best-practice. Health education literature demonstrates inconsistent statistical reporting practices, and falls short in employing best practices and consistently outlining the minimum expectations recommended by APA. In an effort to maximize utility and implications of health education, promotion, and behavior research, future investigations should provide comprehensive information regarding research findings

    The KINDRA project – towards Open Science in Hydrogeology for higher impact

    Groundwater knowledge and research in the European Union is often scattered and non-standardised. Therefore, KINDRA is conducting an EU-wide assessment of existing groundwater-related practical and scientific knowledge based on a new Hydrogeological Research Classification System (HRC-SYS). The classification is supported by a web service, the European Inventory of Groundwater Research (EIGR), which acts not only as a knowledge repository but also as a tool to help identify relevant research topics, existing research trends and critical research challenges. These results will be useful for producing synergies, implementing policies and optimising water management in Europe. This article presents the work of the project during the first two years in relation to a common classification system and an activity for data collection and training delivered by the EFG’s National Associations in 20 European countries

    An aircraft gas turbine engine assembly includes an inter-turbine frame axially located between high and low pressure turbines. Low pressure turbine has counter rotating low pressure inner and outer rotors with low pressure inner and outer shafts which are at least in part rotatably disposed co-axially within a high pressure rotor. Inter-turbine frame includes radially spaced apart radially outer first and inner second structural rings disposed co-axially about a centerline and connected by a plurality of circumferentially spaced apart struts. Forward and aft sump members having forward and aft central bores are fixedly joined to axially spaced apart forward and aft portions of the inter-turbine frame. Low pressure inner and outer rotors are rotatably supported by a second turbine frame bearing mounted in aft central bore of aft sump member.     An attempt to explore the production routes of Astatine radionuclides: Theoretical approach

    In order to fulfil the recent thrust of Astatine radionuclides in the field of nuclear medicine various production routes have been explored in the present work. The possible production routes of 209211^{209-211}At comprise both light and heavy ion induced reactions at the bombarding energy range starting from threshold to maximum 100 MeV energy. For this purpose, we have used the nuclear reaction model codes TALYS, ALICE91 and PACE-II. Excitation functions of those radionuclides, produced through various production routes, have been calculated using nuclear reaction model codes and compared with the available measured data. Contribution of various reaction mechanisms, like, direct, preequilibrium and equilibrium reactions, to the total reaction cross section has been studied using the codes. Result shows that equilibrium reaction mechanism dominates in all cases over other reaction mechanisms

    Quality science from quality measurement: The role of measurement type with respect to replication and effect size magnitude in psychological research

The Many Labs Project (MLP) and the Open-Science-Collaboration (OSC) have collected key data on replicability and statistical effect sizes. We build on this work by investigating the role played by three measurement types: ratings, proportions and unbounded (measures without conceptual upper limits, e.g. time). Both replicability and effect sizes are dependent on the amount of variability due to extraneous factors. We predicted that the role of such extraneous factors might depend on measurement type, and would be greatest for ratings, intermediate for proportions and least for unbounded. Our results support this conjecture. OSC replication rates for unbounded, 43% and proportion 40% combined are reliably higher than those for ratings at 20% (effect size, w = .20). MLP replication rates for the original studies are: pro- portion = .74, ratings = .40 (effect size w = .33). Original effect sizes (Cohen’s d) are highest for: unbounded OSC cognitive = 1.45, OSC social = .90); next for proportions (OSC cogni- tive = 1.01, OSC social = .84, MLP = .82); and lowest for ratings (OSC social = .64, MLP = .31). These findings are of key importance to scientific methodology and design, even if the reasons for their occurrence are still at the level of conjecture.Peer reviewe

    Feasibility Study of Dual Energy Radiographic Imaging for Target Localization in Radiotherapy for Lung Tumors

    Purpose Dual-energy (DE) radiographic imaging improves tissue discrimination by separating soft from hard tissues in the acquired images. This study was to establish a mathematic model of DE imaging based on intrinsic properties of tissues and quantitatively evaluate the feasibility of applying the DE imaging technique to tumor localization in radiotherapy. Methods We investigated the dependence of DE image quality on the radiological equivalent path length (EPL) of tissues with two phantoms using a stereoscopic x-ray imaging unit. 10 lung cancer patients who underwent radiotherapy each with gold markers implanted in the tumor were enrolled in the study approved by the hospital's Ethics Committee. The displacements of the centroids of the delineated gross tumor volumes (GTVs) in the digitally reconstructed radiograph (DRR) and in the bone-canceled DE image were compared with the averaged displacements of the centroids of gold markers to evaluate the feasibility of using DE imaging for tumor localization. Results The results of the phantom study indicated that the contrast-to-noise ratio (CNR) was linearly dependent on the difference of EPL and a mathematical model was established. The objects and backgrounds corresponding to ΔEPL less than 0.08 are visually indistinguishable in the bone-canceled DE image. The analysis of patient data showed that the tumor contrast in the bone-canceled images was improved significantly as compared with that in the original radiographic images and the accuracy of tumor localization using the DE imaging technique was comparable with that of using fiducial makers. Conclusion It is feasible to apply the technique for tumor localization in radiotherapy
