Direct observation of charge inversion by multivalent ions as a universal electrostatic phenomenon
We have directly observed reversal of the polarity of charged surfaces in
water upon the addition of tri- and quadrivalent ions using atomic force
microscopy. The bulk concentration of multivalent ions at which charge
inversion reversibly occurs depends only very weakly on the chemical
composition, surface structure, size and lipophilicity of the ions, but is
dominated by their valence. These results support the theoretical proposal that
spatial correlations between ions are the driving mechanism behind charge

Stress, Physical Activity, and Resilience Resources: Tests of Direct and Moderation Effects in Young Adults
Stress is an important consideration for understanding why individuals take part in limited or no physical activity (PA). The effects of stress on PA do not hold for everyone, so examinations of possible moderators that protect individuals from the harmful effects of stress are required. Aligned with a resilience framework, individual resources (e.g., hope and self-efficacy) may buffer the maladaptive effects of stress, such that people who have access to these resources in greater quantity may be more "resilient" to the deleterious effects of stress on PA. This study was designed to test this expectation. In total, 140 Australian undergraduate students (70.7% female, Mage = 21.68 ± 4.88) completed a multisection survey and provided a sample for hair cortisol concentration analysis using immunoassays. Main effects demonstrated primarily small and nonsignificant associations between perceived stress and hair cortisol concentration with different intensities of PA. Similar findings were observed between individual-level resilience resources and PA intensities, with the exception of hope (i.e., positive association with vigorous PA and negative association with sitting), self-efficacy (i.e., positive association with vigorous PA), and resilience (i.e., positive association with walking). Although certain individual-level resilience resources were perceived as beneficial for PA and sedentary time, the moderating role of resilience resources was not supported by the findings. The direct and moderating effects between stress, PA, and resilience resources require further testing using longitudinal designs in which stressful periods occur naturally (e.g., exams for students) or are experimentally manipulated
One-carbon metabolism in cancer
Cells require one-carbon units for nucleotide synthesis, methylation and reductive metabolism, and these pathways support the high proliferative rate of cancer cells. As such, anti-folates, drugs that target one-carbon metabolism, have long been used in the treatment of cancer. Amino acids, such as serine are a major one-carbon source, and cancer cells are particularly susceptible to deprivation of one-carbon units by serine restriction or inhibition of de novo serine synthesis. Recent work has also begun to decipher the specific pathways and sub-cellular compartments that are important for one-carbon metabolism in cancer cells. In this review we summarise the historical understanding of one-carbon metabolism in cancer, describe the recent findings regarding the generation and usage of one-carbon units and explore possible future therapeutics that could exploit the dependency of cancer cells on one-carbon metabolism
The adhesive properties of pyridine-terminated self-assembled monolayers
The atomic force microscopy (AFM) adhesion force behaviour and contact angle titration behaviour of self-assembled monolayers (SAMs) presenting surface pyridine and substituted pyridine moieties has been investigated as a function of pH and electrolyte concentration. The pKas of the pyridine moieties were modified through the incorporation of fluorine, chlorine and bromine substituents in the pyridyl ring. Contact angle titration and AFM adhesion force measurements were performed using aqueous phosphate buffered saline solutions over the pH range 3–9, and at concentrations of 150 mM and 0.1 mM. Synthetic ozone deposition and stomatal uptake at flux tower sites
We develop and evaluate a method to estimate O-3 deposition and stomatal O-3 uptake across networks of eddy covariance flux tower sites where O-3 concentrations and O-3 fluxes have not been measured. The method combines standard micrometeorological flux measurements, which constrain O-3 deposition velocity and stomatal conductance, with a gridded dataset of observed surface O-3 concentrations. Measurement errors are propagated through all calculations to quantify O-3 flux uncertainties. We evaluate the method at three sites with O(3 )flux measurements: Harvard Forest, Blodgett Forest, and Hyytiala Forest. The method reproduces 83 % or more of the variability in daily stomatal uptake at these sites with modest mean bias (21 % or less). At least 95 % of daily average values agree with measurements within a factor of 2 and, according to the error analysis, the residual differences from measured O-3 fluxes are consistent with the uncertainty in the underlying measurements. The product, called synthetic O-3 flux or SynFlux, includes 43 FLUXNET sites in the United States and 60 sites in Europe, totaling 926 site years of data. This dataset, which is now public, dramatically expands the number and types of sites where O-3 fluxes can be used for ecosystem impact studies and evaluation of air quality and climate models. Across these sites, the mean stomatal conductance and O-3 deposition velocity is 0.03-1.0 cm s(-1). The stomatal O-3 flux during the growing season (typically April-September) is 0.5-11.0 nmol O-3 m(-2) s(-1) with a mean of 4.5 nmol O(3 )m(-2) s(-1) and the largest fluxes generally occur where stomatal conductance is high, rather than where O-3 concentrations are high. The conductance differences across sites can be explained by atmospheric humidity, soil moisture, vegetation type, irrigation, and land management. These stomatal fluxes suggest that ambient O-3 degrades biomass production and CO2 sequestration by 20 %-24 % at crop sites, 6 %-29 % at deciduous broadleaf forests, and 4 %-20 % at evergreen needleleaf forests in the United States and Europe.Peer reviewe

Casimir Forces between Spherical Particles in a Critical Fluid and Conformal Invariance
Mesoscopic particles immersed in a critical fluid experience long-range
Casimir forces due to critical fluctuations. Using field theoretical methods,
we investigate the Casimir interaction between two spherical particles and
between a single particle and a planar boundary of the fluid. We exploit the
conformal symmetry at the critical point to map both cases onto a highly
symmetric geometry where the fluid is bounded by two concentric spheres with
radii R_- and R_+. In this geometry the singular part of the free energy F only
depends upon the ratio R_-/R_+, and the stress tensor, which we use to
calculate F, has a particularly simple form. Different boundary conditions
(surface universality classes) are considered, which either break or preserve
the order-parameter symmetry. We also consider profiles of thermodynamic
densities in the presence of two spheres. Explicit results are presented for an
ordinary critical point to leading order in epsilon=4-d and, in the case of
preserved symmetry, for the Gaussian model in arbitrary spatial dimension d.
Fundamental short-distance properties, such as profile behavior near a surface
or the behavior if a sphere has a `small' radius, are discussed and verified.

The inter-relationship of symptom severity and quality of life in 2055 patients with primary biliary cholangitis
BackgroundAge at presentation with primary biliary cholangitis (PBC) is associated with differ-ential response to ursodeoxycholic acid (UDCA) therapy. Younger-presentingpatients are less likely to respond to treatment and more likely to need transplantor die from the disease. PBC has a complex impact on quality of life (QoL), withsystemic symptoms often having significant impact.AimTo explain the impact of age at presentation on perceived QoL and the inter-related symptoms which impact upon it.MethodsUsing the UK-PBC cohort, symptoms were assessed using the PBC-40 and othervalidated tools. Data were available on 2055 patients.ResultsOf the 1990 patients reporting a global PBC-QoL score, 66% reported good/neutralscores and 34% reported poor scores. Each 10-year increase in age at presentationwas associated with a 14% decrease in risk of poor perceived QoL (OR = 0.86,95% CI: 0.75–0.98, P < 0.05). All sympto m domains were similarly age-associated(P < 0.01). Social dysfunction was the symptom factor with the greatest impa ct onQoL. Median (interquartile range) PBC-40 social scores for patient s with good per-ceived QoL were 18 (14–23) compared with 34 (29–39) for those with poor QoL.ConclusionThe majo rity of patients with primary biliary cholangitis do not feel their QoL isimpaired, although impairment is reported by a sizeable minority. Age at presenta-tion is associated with impact on per ceived QoL and the symptoms impairing it,with younger patients being more affected. Social dysfunction makes the greatestcontribution to QoL impairment, and it should be targeted in trials aimed atimproving life quality

Metabolic Profiling Reveals a Dependency of Human Metastatic Breast Cancer on Mitochondrial Serine and One-Carbon Unit Metabolism.
Breast cancer is the most common cancer among American women and a major cause of mortality. To identify metabolic pathways as potential targets to treat metastatic breast cancer, we performed metabolomics profiling on the breast cancer cell line MDA-MB-231 and its tissue-tropic metastatic subclones. Here, we report that these subclones with increased metastatic potential display an altered metabolic profile compared with the parental population. In particular, the mitochondrial serine and one-carbon (1C) unit pathway is upregulated in metastatic subclones. Mechanistically, the mitochondrial serine and 1C unit pathway drives the faster proliferation of subclones through enhanced de novo purine biosynthesis. Inhibition of the first rate-limiting enzyme of the mitochondrial serine and 1C unit pathway, serine hydroxymethyltransferase (SHMT2), potently suppresses proliferation of metastatic subclones in culture and impairs growth of lung metastatic subclones at both primary and metastatic sites in mice. Some human breast cancers exhibit a significant association between the expression of genes in the mitochondrial serine and 1C unit pathway with disease outcome and higher expression of SHMT2 in metastatic tumor tissue compared with primary tumors. In addition to breast cancer, a few other cancer types, such as adrenocortical carcinoma and kidney chromophobe cell carcinoma, also display increased SHMT2 expression during disease progression. Together, these results suggest that mitochondrial serine and 1C unit metabolism plays an important role in promoting cancer progression, particularly in late-stage cancer. IMPLICATIONS: This study identifies mitochondrial serine and 1C unit metabolism as an important pathway during the progression of a subset of human breast cancers
No Rise in Incidence but Geographical Heterogeneity in the Occurrence of Primary Biliary Cirrhosis in North East England
In this study, we examined temporal changes in the incidence of primary biliary cirrhosis (PBC) and investigated associations between PBC incidence and sociodemographic factors and spatial clustering. We included 982 patients aged ≥40 years from North East England with incident PBC diagnosed during 1987–2003. Age-standardized incidence rates with 95% confidence intervals were calculated. Negative binomial regression was used to analyze incidence and socioeconomic deprivation. Clustering analysis was performed using point process methods, testing the null hypothesis that disease risk does not vary spatially and that PBC cases occur independently. The age-standardized incidence rate was 53.50 per million persons per year (95% confidence interval: 48.65, 58.35) in 1987–1994 and 45.09 per million persons per year (95% confidence interval: 41.10, 49.07) in 1995–2003. Risk of PBC increased in areas with higher levels of socioeconomic deprivation (P = 0.035). More specifically, risk increased in areas with higher levels of overcrowded homes (P = 0.040), higher levels of households without cars (P < 0.001), and higher levels of non-owner-occupied homes (P < 0.001). Overall, there was evidence of spatial clustering (P = 0.001). The findings confirm that overall incidence of PBC did not rise over time, but sociodemographic variations suggest that certain aspects of deprivation are involved in its etiology
