Relaksyna – hormon ciążowy zaangażowany w proces nowotworzenia
Relaxin 2 is a polypeptide hormone structurally related to insulin and insulin-like growth factors (IGFs). However, it does not interact with insulin receptors and has a decidedly different biological properties. Relaxin 2 activates LGR 7 and LGR 8 relaxin receptors, that belong to the leucine-rich repeat-containing (LRR), G protein-coupled receptors. The characteristic functions of relaxin are associated with female reproductive system, especially during pregnancy. However, recent studies have shown that polypeptides with relaxin-like factor family (RLF) are involved in cell proliferation and differentiation, as well as invasion and angiogenesis of female and male reproductive cancers. This review provides information on the structure and function of relaxin and its receptors. Furthermore, we present evidence of the involvement of these elements in the process of cancerogenesis.Relaksyna 2 jest polipeptydem o budowie podobnej do insuliny i insulino-podobnych czynników wzrostu (IGFs). Hormon ten nie wchodzi jednak w interakcje z receptorami insulinowymi, przez co wykazuje zdecydowanie odmienne właściwości biologiczne. Relaksyna 2 powoduje aktywację receptorów: LGR7 i LGR8. Receptory te należą do siedmiodomenowych receptorów transbłonowych, sprzężonych z białkiem G, zawierających powtórzenia bogate w leucynę (LRR). Funkcja relaksyny 2 kojarzona jest przede wszystkim z żeńskim układem rozrodczym, zwłaszcza w czasie ciąży. Jednak ostatnie badania wykazały, że ten polipeptyd jest także zaangażowany w proliferację i różnicowanie komórek. Ponadto odnotowano, że peptydy z rodziny czynników relaksynopodobnych, wpływają na inwazyjność oraz angiogenezę nowotworów żeńskiego i męskiego układu rozrodczego. Niniejszy artykuł poglądowy przedstawia wiele interesujących informacji na temat struktury oraz funkcji relaksyny 2 i jej receptorów. Dostarcza również wielu dowodów na udział tego peptydu w procesie nowotworzenia

The influence of peptides from the angiotensin family on tyrosine kinase activity and cell viability in a human hormone-dependent prostate cancer line
Wstęp: Wyniki wielu badań wskazują, że peptydy z rodziny angiotensyn są włączone w regulację cyklu komórkowego, apoptozę, różnicowanie
się komórek, jak również odgrywają istotną rolę w procesach zapalnych i migracji komórek. Ich zaangażowanie w tak istotne
procesy sugeruje, że mogą one odgrywać istotną rolę w procesie kancerogenezy. Stosunkowo mało wiadomo na temat roli systemu
renina-angiotensyna w zapoczątkowaniu i późniejszym rozwoju raka stercza.
Materiał i metody: Poniższe wyniki przedstawiają wpływ Ang II, Ang III, Ang IV na linię komórkową ludzkiego hormonozależnego raka
prostaty. Używając metody izotopowej, badano wpływ wybranych angiotensyn na aktywność kinaz tyrozynowych. Podczas gdy przeżywalność
komórkowa była oznaczana za pomocą testu MTT.
Wyniki: Wyniki wskazują, że tylko Ang IV w sposób istotny statystycznie redukuje aktywność kinaz tyrozynowych i obniża przeżywalność
komórek linii LNCaP. Proces ten był pośredniczony przez receptor AT2 i prawdopodobnie przez inny receptor, o wysokim powinowactwie
dla Ang IV i niskim dla PD123319 i losartanu.
Wnioski: Otrzymane wyniki sugerują, że komponenty układu renina–angiotensyna, w tym szczególnie peptydy angiotensynowe oraz
klasyczne receptory dla angiotensyn (AT1, AT2), mogą modyfikować przeżywalność komórek raka gruczołu krokowego.Introduction: The results of many studies have reported that peptides from the angiotensin family are involved in the regulation of cell
growth, proliferation, cell migration, apoptosis, inflammation, differentiation, and angiogenesis, which suggests that they might play an
important role in carcinogenesis. The role of the renin–angiotensin system in supporting prostate cancer induction and progression has so
far received little study.
Material and methods: The present study was to examine the influence of Ang II, Ang III, and Ang IV on a human hormone-dependent
prostate cancer line (LNCaP). Using an isotopic method, we tested the effects of angiotensins on tyrosine kinase activity, and measured
cell viability using an MTT Assay.
Results: The results showed that only Ang IV significantly reduced tyrosine kinase activity and cell viability in LNCaP cells. The process
seemed to be mediated partly by AT2 and probably by another type of receptor with high affinity for Ang IV and low affinity for PD123319
and Losartan.
Conclusions: These findings suggest that components of the renin-angiotensin system, specifically angiotensin peptides and receptors
(AT1, AT2) can modify prostate cancer cell viability
Angiotensin II as a factor modulating protein tyrosine kinase activity in two breast cancer lines — MCF-7 and MDA-MB-231
Wstęp: Angiotensyna II (AngII), peptyd systemu renina-angiotensyna biorący udział w regulacji równowagi wodno-elektrolitowej oraz
ciśnienia krwi, jest także związkiem wpływającym na proliferację i wzrost komórek. Efekt działania tego peptydu uzależniony jest od
typu receptora. Angiotensyna II po związaniu z receptorem AT1 stymuluje proliferację i wzrost komórek natomiast poprzez aktywację
receptora AT2 — hamuje wzrost komórek oraz pobudza proces apoptozy. Proproliferacyjne działanie angiotensyny II zaobserwowano zarówno
w komórkach tkanek zdrowych, jak i w komórkach nowotworowych między innymi komórkach przysadki i nadnerczy, ale także raka
piersi czy hiperplazji mięśni naczyń krwionośnych.
Materiał i metody: Celem pracy było zbadanie wpływu AngII na aktywność białkowych kinaz tyrozynowych dwóch linii nowotworowych
gruczołu piersiowego: hormono-zależnej linii MCF-7 i hormono-niezależnej MDA-MB-231. Aktywność kinaz tyrozynowych jest
przyjętym markerem proliferacji. Oszacowano także wpływ 17b-estradiolu na zmiany w aktywności kinaz tyrozynowych wywołane
angiotensyną II.
Wyniki: AngII radykalnie hamowała aktywności kinaz tyrozynowych w linii MDA-MB-231, natomiast 17b-estradiol tylko w stężeniu 10–6 M
pogłębiał efekt działania angiotensyny II w stężeniu 10–9 M. W linii hormono-zależnej MCF-7 Ang II miała silny hamujący wpływ na
aktywność badanych enzymów w obecności estradiolu 10–6 M. Sam estradiol obniżał aktywność badanych enzymów w obu liniach komórkowych.
Obecność receptora angiotensynowego typu 1 została potwierdzona w obydwu badanych liniach raka piersi, natomiast
ekspresję receptora typu 2 odnotowano jedynie w komórkach MDA-MB-231.
Wnioski: Otrzymane wyniki wskazują, że AngII może modulować aktywność kinaz tyrozynowych w badanych komórkach raka gruczołu
sutkowego. (Endokrynol Pol 2011; 62 (2): 151–157)Introduction: Angiotensin II (AngII), a peptide that regulates the water-electrolytic balance and blood pressure, is also known to influence cell
proliferation. It can either induce cell growth, when binding to angiotensin type-I receptor, or trigger growth inhibition via angiotensin
type-II receptor. AngII stimulates proliferation of some normal and tumour cell lines, e.g. pituitary, adrenal glands and breast cancer.
Material and methods: The aim of this study was to evaluate possible AngII effect on the growth of two breast cancer cell lines —
hormone-dependent MCF-7 and hormone-independent MDA-MB-231. We measured tyrosine kinase activity as a potential proliferation
marker. We also estimated the influence of 17b-oestradiolon AngII-induced changes.
Results: In the MDA-MB-231 line, AngII radically slowed the activity of tyrosine kinases and 17b-oestradiol only at a concentration of 10–6 M,
while it enhanced the effect of angiotensin II at a concentration of 10–9 M. In MCF-7, Ang II had a strong inhibitory effect in the presence
of oestradiol (10–6 M). Oestradiol alone decreased the activity of examined enzymes in both cell lines. AngII receptor type 1 was found in
both studied lines, but type 2 only in MDA-MB-231.
Conclusions: Our results show that AngII can modulate tyrosine kinase activity in breast tumour cell lines.(Pol J Endocrinol 2011; 62 (2): 151–157
Influence of myocardial infarction on changes in the expression of angiotensin type 1 receptor in the rat prostate
Angiotensin II (AngII) is the biologically active peptide of the renin-angiotensin system (RAS). Tissue-
based, local RAS has been identified in the prostate, testis, epididymis and coagulating glands. Experimental
and clinical studies have consistently shown that myocardial infarction (MI) is associated with activation of
the systemic RAS with increased concentration of angiotensin peptides in the blood and changes in expression
of angiotensin receptors (AT). Changes in angiotensin receptors in the renal and cardiovascular system after MI
are well recognized, but the effects of MI influence on changes in other tissue like the prostate gland are unknown.
In the present study, we investigated the effect of myocardial infarction on angiotensin receptor protein
and mRNA expression in the rat prostate gland. MI model was established in Wistar rats by ligating the left
coronary artery (modified Selye method). The levels of AT1a-b and AT2 receptor mRNAs and proteins were
measured in the rat prostate. Our study demonstrates tissue-specific changes in AT1a-b and AT2 receptor expression
after myocardial infarction. The results show that MI has a strong influence on the expression of angiotensin
receptor type AT1 in the prostate at the protein and mRNA level. (Folia Histochemica et Cytobiologica

Relaxin-like peptides in male reproduction: a human perspective
The relaxin family of peptide hormones and their cognate GPCRs are becoming physiologically well-characterized in the cardiovascular system and particularly in female reproductive processes. Much less is known about the physiology and pharmacology of these peptides in male reproduction, particularly as regards humans. H2-relaxin is involved in prostate function and growth, while insulin-like peptide 3 (INSL3) is a major product of the testicular Leydig cells and, in the adult, appears to modulate steroidogenesis and germ cell survival. In the fetus, INSL3 is a key hormone expressed shortly after sex determination and is responsible for the first transabdominal phase of testicular descent. Importantly, INSL3 is becoming a very useful constitutive biomarker reflecting both fetal and post-natal development. Nothing is known about roles for INSL4 in male reproduction and only very little about relaxin-3, which is mostly considered as a brain peptide, or INSL5. The former is expressed at very low levels in the testes, but has no known physiology there, whereas the INSL5 knockout mouse does exhibit a testicular phenotype with mild effects on spermatogenesis,probably due to a disruption of glucose homeostasis. INSL6 is a major product of male germ cells, although it is relatively unexplored with regard to its physiology or pharmacology, except that in mice disruption of the INSL6 gene leads to a disruption of spermatogenesis. Clinically, relaxin analogues may be useful in the control of prostate cancer, and both relaxin and INSL3 have been considered as sperm adjuvants for in vitro fertilization
The importance of the local renin-angiotensin system in the pathology of prostate
Results of in vivo and in vitro experiments have demonstrated the presence of a local renin-angiotensin system (RAS) in the prostate. However, the role of this system in the physiology and pathology of the prostate is still unclear. Recent reports have indicated that RAS is also involved in the regulation of cell proliferation, differentiation, migration as well as angiogenesis, inflammation and apoptosis, which suggests its important role in carcinogenesis. Reorganization of individual elements of the renin-angiotensin system has been reported in both benign prostatic hyperplasia and prostate cancer. These changes concerned the expression of AT1 receptor, angiotensin converting enzyme (ACE) and local concentration of angiotensin II or angiotensinogen. This review focuses on the role of RAS in the induction and progression of prostate cancer as well as on the analysis of potential benefits of RAS modulation in anticancer therapy.Wyniki badań in vivo oraz in vitro ujawniły obecność lokalnego układu renina-angiotensyna (RAS) w gruczole krokowym, niemniej jednak rola tego układu zarówno w fizjologii jak i patologii stercza nadal nie jest dobrze znana. Najnowsze doniesienia wskazują, że RAS obok swojej klasycznej roli jest zaangażowany w proliferację, różnicowanie i migrację komórek, angiogenezę, procesy zapalne oraz apoptozę, co sugeruje jego istotny udział w procesie kancerogenezy. Reorganizacja poszczególnych elementów RAS została odnotowana zarówno w łagodnym rozroście jak i nowotworach złośliwych gruczołu krokowego. Powyższe zmiany dotyczyły między innymi, poziomu receptora AT1, ekspresji enzymu konwertującego angiotensynę I jak również lokalnego stężenia angiotensyny II i angiotensynogenu. Niniejsza praca ma na celu podsumowanie dotychczasowej wiedzy odnośnie udziału RAS w zapoczątkowaniu i późniejszym rozwoju raka stercza, jak również przeanalizowanie potencjalnych korzyści wynikających z modulacji RAS w kontekście terapii antynowotworowej
Synthesis and Preliminary Evaluation of the Cytotoxicity of Potential Metabolites of Quinoline Glycoconjugates
The design of prodrugs is one of the important strategies for selective anti-cancer therapies. When designing prodrugs, attention is paid to the possibility of their targeting tumor-specific markers such as proteins responsible for glucose uptake. That is why glycoconjugation of biologically active compounds is a frequently used strategy. Glycoconjugates consisting of three basic building blocks: a sugar unit, a linker containing a 1,2,3-triazole ring, and an 8-hydroxyquinoline fragment was described earlier. It is not known whether their cytotoxicity is due to whole glycoconjugates action or their metabolites. To check the biological activity of products that can be released from glycoconjugates under the action of hydrolytic enzymes, the synthetically obtained potential metabolites were tested in vitro for the inhibition of proliferation of HCT-116, MCF-7, and NHDF-Neo cell lines using the MTT assay. Research shows that for the full activity of glycoconjugates, the presence of all three building blocks in the structure of a potential drug is necessary. For selected derivatives, additional tests of targeted drug delivery to tumor cells were carried out using polymer nanocarriers in which they are encapsulated. This approach significantly lowered the determined IC50 values of the tested compounds and improved their selectivity and effectiveness
Primary cognitive appraisal as a determinant for strategy of coping with difficulty social situation among junior high school (gimnazjum) students
Celem artykułu jest określenie zależności pomiędzy oceną poznawczą a wyborem
strategii obronnych (agresja, unik) i strategii zadaniowej stosowanych przez młodzież
w trudnych sytuacjach społecznych. W badaniach posłużono się własnym (autorskim)
kwestionariuszem do badania radzenia sobie młodzieży w trudnych sytuacjach
społecznych ( KNO) oraz Kwestionariuszem Oceny Stresu ( KOS) autorstwa K. Wrześniewskiego,
A. Jakubowskiej-Winieckiej i D. Włodarczyk. Badania empiryczne
prowadzone były w szkołach gimnazjalnych województwa dolnośląskiego. Objęły
300 gimnazjalistów (150 chłopców, 150 dziewcząt) w wieku 13 –15 lat. W świetle
przeprowadzonych badań stwierdzono, że ocena poznawcza (w kategoriach wyzwania,
krzywdy/straty i zagrożenia) determinuje stosowanie obronnych (agresja/unik) i zadaniowych
sposobów reagowania młodzieży w sytuacjach społecznie trudnych.The aim of the following article is to establish the relation between cognitive appraisal
and the selection of defence strategy (aggression, evasion) and task-based strategy applied
by the youth in difficult social situations. In the research process two questionnaires
were in use, one questionnaire of my own which was designed to learn how the youth
copes with difficult social situations ( kNO) and the other named The Stress Evaluation
Questionnaire ( kOS) by Wrześniewski, Jakubowska-Winnicka and Włodarczyk. Empirical
research was conducted in junior high schools of the Lower Silesia region. It covered 300
junior high school students (150 boys and 150 girls) at the ages between 13-15. In the
light of the carried out research it was concluded that cognitive appraisal in the categories
of challenge, injury/loss and threat determine the selection of defensive (aggression/evasion)
and task-based ways of students reacting to difficult social situations
DEVELOPING WRITING STRATEGIES OF POOR LANGUAGE LEARNERS AT THE LEVEL OF JUNIOR-HIGH SCHOOL
Although language learning strategy research has been going on for almost forty years and has provided both theorists and practitioners with an abundant body of knowledge on the subject, there are still areas that need further investigation. One such area is the use of strategies in language skills development, with the skill of writing being singled out as deserving special attention. It is suggested that in order to better understand the processes involved, we need more information concerning the development of the skill by learners, as well as more data revealing the effects of strategy based instruction, especially with reference to children and adolescents in different foreign language learning contexts. Hence, this article makes an attempt to contribute to the ongoing discussion by focusing on a special group of learners – poor language learners, at a risk of educational failure – who are learning a foreign language (English) in the context of junior high school. The article consists of two parts: its theoretical sections focus on some issues related to poor language learners, with emphasis on factors impacting their school problems in general and learning of the writing skill in particular. Then, based on the literature review, some research findings concerning writing strategies and the effects of strategic training are discussed. The second part presents and comments on the data obtained during the course of an informal study carried out in a junior high school within the framework of an educational project which was remedial in character
