    The Innate Immune Receptor PGRP-LC Controls Presynaptic Homeostatic Plasticity

    SummaryIt is now appreciated that the brain is immunologically active. Highly conserved innate immune signaling responds to pathogen invasion and injury and promotes structural refinement of neural circuitry. However, it remains generally unknown whether innate immune signaling has a function during the day-to-day regulation of neural function in the absence of pathogens and irrespective of cellular damage or developmental change. Here we show that an innate immune receptor, a member of the peptidoglycan pattern recognition receptor family (PGRP-LC), is required for the induction and sustained expression of homeostatic synaptic plasticity. This receptor functions presynaptically, controlling the homeostatic modulation of the readily releasable pool of synaptic vesicles following inhibition of postsynaptic glutamate receptor function. Thus, PGRP-LC is a candidate receptor for retrograde, trans-synaptic signaling, a novel activity for innate immune signaling and the first known function of a PGRP-type receptor in the nervous system of any organism

    Spatial variation in leopard (Panthera pardus) site use across a gradient of anthropogenic pressure in Tanzania's Ruaha landscape

    Understanding large carnivore occurrence patterns in anthropogenic landscapes adjacent to protected areas is central to developing actions for species conservation in an increasingly human-dominated world. Among large carnivores, leopards (Panthera pardus) are the most widely distributed felid. Leopards occupying anthropogenic landscapes frequently come into conflict with humans, which often results in leopard mortality. Leopards’ use of anthropogenic landscapes, and their frequent involvement with conflict, make them an insightful species for understanding the determinants of carnivore occurrence across human-dominated habitats. We evaluated the spatial variation in leopard site use across a multiple-use landscape in Tanzania’s Ruaha landscape. Our study region encompassed i) Ruaha National Park, where human activities were restricted and sport hunting was prohibited; ii) the Pawaga-Idodi Wildlife Management Area, where wildlife sport hunting, wildlife poaching, and illegal pastoralism all occurred at relatively low levels; and iii) surrounding village lands where carnivores and other wildlife were frequently exposed to human-carnivore conflict related-killings and agricultural habitat conversion and development. We investigated leopard occurrence across the study region via an extensive camera trapping network. We estimated site use as a function of environmental (i.e. habitat and anthropogenic) variables using occupancy models within a Bayesian framework. We observed a steady decline in leopard site use with downgrading protected area status from the national park to the Wildlife Management Area and village lands. Our findings suggest that human-related activities such as increased livestock presence and proximity to human households exerted stronger influence than prey availability on leopard site use, and were the major limiting factors of leopard distribution across the gradient of human pressure, especially in the village lands outside Ruaha National Park. Overall, our study provides valuable information about the determinants of spatial distribution of leopards in human-dominated landscapes that can help inform conservation strategies in the borderlands adjacent to protected areas

    Public perceptions of trophy hunting are pragmatic, not dogmatic

    Funding: This work is an output from the Morally Contested Conservation research project, supported by Jamma International, WWF Deutschland, and the Luc Hoffmann Institute (now Unearthodox) to the University of Oxford [grant number ATR04380].Fierce international debates rage over whether trophy hunting is socially acceptable, especially when people from the Global North hunt well-known animals in sub-Saharan Africa. We used an online vignette experiment to investigate public perceptions of the acceptability of trophy hunting in sub-Saharan Africa among people who live in urban areas of the USA, UK and South Africa. Acceptability depended on specific attributes of different hunts as well as participants' characteristics. Zebra hunts were more acceptable than elephant hunts, hunts that would provide meat to local people were more acceptable than hunts in which meat would be left for wildlife, and hunts in which revenues would support wildlife conservation were more acceptable than hunts in which revenues would support either economic development or hunting enterprises. Acceptability was generally lower among participants from the UK and those who more strongly identified as an animal protectionist, but higher among participants with more formal education, who more strongly identified as a hunter, or who would more strongly prioritize people over wild animals. Overall, acceptability was higher when hunts would produce tangible benefits for local people, suggesting that members of three urban publics adopt more pragmatic positions than are typically evident in polarized international debates.Peer reviewe

    Effect of COVID-19 lockdown on hospital admissions and mortality in rural KwaZulu-Natal, South Africa: interrupted time series analysis.

    OBJECTIVE: To assess the effect of lockdown during the 2020 COVID-19 pandemic on daily all-cause admissions, and by age and diagnosis subgroups, and the odds of all-cause mortality in a hospital in rural KwaZulu-Natal (KZN). DESIGN: Observational cohort. SETTING: Referral hospital for 17 primary care clinics in uMkhanyakude District. PARTICIPANTS: Data collected by the Africa Health Research Institute on all admissions from 1 January to 20 October: 5848 patients contributed to 6173 admissions. EXPOSURE: Five levels of national lockdown in South Africa from 27 March 2020, with restrictions decreasing from levels 5 to 1, respectively. OUTCOME MEASURES: Changes and trends in daily all-cause admissions and risk of in-hospital mortality before and at each stage of lockdown, estimated by Poisson and logistic interrupted time series regression, with stratification for age, sex and diagnosis. RESULTS: Daily admissions decreased during level 5 lockdown for infants (incidence rate ratio (IRR) compared with prelockdown 0.63, 95% CI 0.44 to 0.90), children aged 1-5 years old (IRR 0.43, 95% CI 028 to 0.65) and respiratory diagnoses (IRR 0.57, 95% CI 0.36 to 0.90). From level 4 to level 3, total admissions increased (IRR 1.17, 95% CI 1.06 to 1.28), as well as for men >19 years (IRR 1.50, 95% CI 1.17 to 1.92) and respiratory diagnoses (IRR 4.26, 95% CI 2.36 to 7.70). Among patients admitted to hospital, the odds of death decreased during level 5 compared with prelockdown (adjusted OR 0.48, 95% CI 0.28 to 0.83) and then increased in later stages. CONCLUSIONS: Level 5 lockdown is likely to have prevented the most vulnerable population, children under 5 years and those more severely ill from accessing hospital care in rural KZN, as reflected by the drop in admissions and odds of mortality. Subsequent increases in admissions and in odds of death in the hospital could be due to improved and delayed access to hospital as restrictions were eased

    Relative efforts of countries to conserve world’s megafauna

    Surprisingly little attention has been paid to variation among countries in contributions to conservation. As a first step, we developed a Megafauna Conservation Index (MCI) that assesses the spatial, ecological and financial contributions of 152 nations towards conservation of the world’s terrestrial megafauna. We chose megafauna because they are particularly valuable in economic, ecological and societal terms, and are challenging and expensive to conserve. We categorised these 152 countries as being above- or below-average performers based on whether their contribution to megafauna conservation was higher or lower than the global mean; ‘major’ performers or underperformers were those whose contribution exceeded 1 SD over or under the mean, respectively. Ninety percent of countries in North/Central America and 70% of countries in Africa were classified as major or above-average performers, while approximately one-quarter of countries in Asia (25%) and Europe (21%) were identified as major underperformers. We present our index to emphasize the need for measuring conservation performance, to help nations identify how best they could improve their efforts, and to present a starting point for the development of more robust and inclusive measures (noting how the IUCN Red List evolved over time). Our analysis points to three approaches that countries could adopt to improve their contribution to global megafauna conservation, depending on their circumstances: 1) upgrading or expanding their domestic protected area networks, with a particular emphasis on conserving large carnivore and herbivore habitat, 2) increase funding for conservation at home or abroad, or 3) ‘rewilding’ their landscapes. Once revised and perfected, we recommend publishing regular conservation rankings in the popular media to recognise major-performers, foster healthy pride and competition among nations, and identify ways for governments to improve their performance

    Density responses of lesser-studied carnivores to habitat and management strategies in southern Tanzania's Ruaha-Rungwa landscape

    Compared to emblematic large carnivores, most species of the order Carnivora receive little conservation attention despite increasing anthropogenic pressure and poor understanding of their status across much of their range. We employed systematic camera trapping and spatially explicit capture-recapture modelling to estimate variation in population density of serval, striped hyaena and aardwolf across the mixed-use Ruaha-Rungwa landscape in southern Tanzania. We selected three sites representative of different habitat types, management strategies, and levels of anthropogenic pressure: Ruaha National Park’s core tourist area, dominated by Acacia-Commiphora bushlands and thickets; the Park’s miombo woodland; and the neighbouring community-run MBOMIPA Wildlife Management Area, also covered in Acacia-Commiphora. The Park’s miombo woodlands supported a higher serval density (5.56 [Standard Error = ±2.45] individuals per 100 km2) than either the core tourist area (3.45 [±1.04] individuals per 100 km2) or the Wildlife Management Area (2.08 [±0.74] individuals per 100 km2). Taken together, precipitation, the abundance of apex predators, and the level of anthropogenic pressure likely drive such variation. Striped hyaena were detected only in the Wildlife Management Area and at low density (1.36 [±0.50] individuals per 100 km2), potentially due to the location of the surveyed sites at the edge of the species’ global range, high densities of sympatric competitors, and anthropogenic edge effects. Finally, aardwolf were captured in both the Park’s core tourist area and the Wildlife Management Area, with a higher density in the Wildlife Management Area (13.25 [±2.48] versus 9.19 [±1.66] individuals per 100 km2), possibly as a result of lower intraguild predation and late fire outbreaks in the area surveyed. By shedding light on three understudied African carnivore species, this study highlights the importance of miombo woodland conservation and community-managed conservation, as well as the value of by-catch camera trap data to improve ecological knowledge of lesser-studied carnivores

    Camera trapping and spatially explicit capture-recapture for the monitoring and conservation management of lions: Insights from a globally important population in Tanzania

    Accurate and precise estimates of population status are required to inform and evaluate conservation management and policy interventions. Although the lion (Panthera leo) is a charismatic species receiving increased conservation attention, robust status estimates are lacking for most populations. While for many large carnivores population density is often estimated through spatially explicit capture–recapture (SECR) applied to camera trap data, the lack of pelage patterns in lions has limited the application of this technique to the species. Here, we present one of the first applications of this methodology to lion, in Tanzania's Ruaha-Rungwa landscape, a stronghold for the species for which no empirical estimates of status are available. We deployed four camera trap grids across habitat and land management types, and we identified individual lions through whisker spots, scars and marks, and multiple additional features. Double-blind identification revealed low inter-observer variation in photo identification (92% agreement), due to the use of xenon-flash cameras and consistent framing and angles of photographs. Lion occurred at highest densities in a prey-rich area of Ruaha National Park (6.12 ± SE 0.94 per 100 km2), and at relatively high densities (4.06 ± SE 1.03 per 100 km2) in a community-managed area of similar riparian-grassland habitat. Miombo woodland in both photographic and trophy hunting areas sustained intermediate lion densities (1.75 ± SE 0.62 and 2.25 ± SE 0.52 per 100 km2, respectively). These are the first spatially explicit density estimates for lion in Tanzania, including the first for a trophy hunting and a community-managed area, and also provide some of the first insights into lion status in understudied miombo habitats. We discuss in detail the methodology employed, the potential for scaling-up over larger areas, and its limitations. We suggest that the method can be an important tool for lion monitoring and explore the implications of our findings for lion management

    Conserving the World’s Megafauna and Biodiversity: The Fierce Urgency of Now

    First paragraph: In our recent perspective article, we noted that most (approximately 60 percent) terrestrial large carnivore and large herbivore species are now threatened with extinction, and we offered a 13-point declaration designed to promote and guide actions to save these iconic mammalian megafauna (Ripple et al. 2016). Some may worry that a focus on saving megafauna might undermine efforts to conserve biodiversity more broadly. We believe that all dimensions of biodiversity are important and that efforts to conserve megafauna are not in themselves sufficient to halt the dispiriting trends of species and population losses in recent decades. From 1970 to 2012, a recent global analysis showed a 58 percent overall decline in vertebrate population abundance (WWF 2016). Bold and varied approaches are necessary to conserve what remains of Earth’s biodiversity, and our declaration in no way disputes the value of specific conservation initiatives targeting other taxa.     Anthropogenic risk increases night-time activities and associations in African elephants (Loxodonta africana) in the Ruaha-Rungwa ecosystem, Tanzania

    Elephants face diverse threats from human activities and use temporal and social strategies to reduce human-induced mortality risk. We used data from camera trap surveys in 2018–2019 (n= 1625 independent detection events from 11,751 sampling days) to investigate elephant responses to anthropogenic risk in the Ruaha-Rungwa ecosystem, Tanzania. The study was conducted in one low- risk and three high- risk sites using 26–40 paired camera trap stations per site. Risk influenced the active pe-riods, use of roads and water sources, social associations and behaviour of elephants. Elephants demonstrated significantly more night-time and reduced daytime activ-ity in the high- risk sites relative to the low- risk site. This higher night-time activity in the high- risk sites was observed for both males and females, though it was more pronounced for cow–calf groups than lone males. Foraging events and use of water sources were more frequent at night in the high- risk sites. Elephants used roads as movement routes in the low- risk site but avoided roads in the high- risk sites. Males were significantly more likely to associate with other males and cow–calf groups in the high- risk sites. Fewer occurrences of relaxed behaviours were observed in the high- risk sites compared to the low- risk site.
