    Near-Field Pressure Signature Splicing for Low-Fidelity Design Space Exploration of Supersonic Aircraft

    As interest in supersonic overland flight intensifies, new ways to meet government restrictions on sonic boom loudness must be implemented. Low-fidelity aerodynamic tools, such as PANAIR, can estimate the near-field pressure signature that ultimately determines the loudness of the sonic boom at the ground. These tools can greatly benefit the exploration of large design spaces due to their computational efficiency. One of the limitations of low-fidelity tools is the accuracy of the solution produced, which is dependent on the fundamental physical assumptions made in the development of the governing equations. If flow patterns are produced that severely violate these fundamental assumptions, the validity of the near-field pressure signature is compromised. A method is proposed that splices together near-field pressure signatures from a low-fidelity and a higher-fidelity tool by cutting each pressure signature at a critical point and then blending the low-fidelity signature into the higher-fidelity signature. By splicing the signatures together, sections of the low-fidelity signature that represent fundamental violations of the governing equation are removed. This method allows for the exploration of the design space corresponding to areas on the geometry that produce accurate results in a low-fidelity signature. The method is tested on the JAXA Wing Body geometry from the Second AIAA Sonic Boom Prediction Workshop and shows that perturbations to this geometry can produce loudness results that match the high-fidelity results to within 0.4 PLdB

    Damage function for historic paper. Part I: Fitness for use

    Background In heritage science literature and in preventive conservation practice, damage functions are used to model material behaviour and specifically damage (unacceptable change), as a result of the presence of a stressor over time. For such functions to be of use in the context of collection management, it is important to define a range of parameters, such as who the stakeholders are (e.g. the public, curators, researchers), the mode of use (e.g. display, storage, manual handling), the long-term planning horizon (i.e. when in the future it is deemed acceptable for an item to become damaged or unfit for use), and what the threshold of damage is, i.e. extent of physical change assessed as damage. Results In this paper, we explore the threshold of fitness for use for archival and library paper documents used for display or reading in the context of access in reading rooms by the general public. Change is considered in the context of discolouration and mechanical deterioration such as tears and missing pieces: forms of physical deterioration that accumulate with time in libraries and archives. We also explore whether the threshold fitness for use is defined differently for objects perceived to be of different value, and for different modes of use. The data were collected in a series of fitness-for-use workshops carried out with readers/visitors in heritage institutions using principles of Design of Experiments. Conclusions The results show that when no particular value is pre-assigned to an archival or library document, missing pieces influenced readers/visitors’ subjective judgements of fitness-for-use to a greater extent than did discolouration and tears (which had little or no influence). This finding was most apparent in the display context in comparison to the reading room context. The finding also best applied when readers/visitors were not given a value scenario (in comparison to when they were asked to think about the document having personal or historic value). It can be estimated that, in general, items become unfit when text is evidently missing. However, if the visitor/reader is prompted to think of a document in terms of its historic value, then change in a document has little impact on fitness for use

    Emerging dietary patterns: impact on child health

    What we eat today and how we decide to produce sufficient food to meet both demand and nutritional requirements will determine the availability, diversity and quality of diets for future generations. Acceptability and normalisation of new dietary patterns must be prioritised to the same extent as education on environmental sustainability and health, because these are important social levers. They can help to reduce the psychosocial barriers and anxieties often connected to the behaviour changes that are needed to transition away from the traditional Western eating habits that are now increasingly subject to critical scrutiny. Without understanding the motivations and facilitators to change eating habits (and then designing and amplifying solutions which make those changes attractive and compelling in the modern world) positive changes to young people’s health and their environment through better food will not come quickly enough to halt potentially catastrophic consequences for people and planet. Changing consumption is seen by experts as the most powerful lever compared to food waste reduction or more efficient food production methods and technological solutions. There is simply not enough on Earth to sustain the increase of intensification required to continue feeding a global population on its current trajectory with a diet so rich in animal produce. However, radical behaviour modification is challenging and therefore cross-sector, bold and perhaps unorthodox leadership is necessary to inspire alteration in the food system, which will have a knock-on effect in terms of changing food environments and therefore food perceptions, narratives and behaviours. There are no ‘quick fix’ solutions, but what is undeniable is that populations can only make effective shifts in dietary patterns if government and the food industry help them to make those changes. This requires budgetary investment in public health information and the implementation of policies that promote ways to eat healthily that are also affordable and environmentally sustainable. The aim is to offer food that tastes as good as it can be made to look so that uptake is maximised and the benefits are cumulative. The inevitable upheaval incurred by making any alteration to entrenched patterns of consumption that have been centuries in the making should not be underestimated, but carrying on as before is no longer credible nor desirable.     Caracterização de acessos de pimenta-do-reino com base em sistemas enzimáticos.

    Setenta e oito acessos de pimenta-do-reino, incluindo algumas espécies silvestres foram submetidos à análise eletroforética de isoenzimas em gel de poliacrilamida, visando distinguir diferenças fenotípicas que auxiliem na discriminação e seleção dos acessos. Foram utilizados os sistemas enzimáticos SKDH, GOT, ACP, ACO, PGI, FUM, 6PGDH e G6PDH. O polimorfismo de isoenzimas foi avaliado pelo número de alozimas com diferentes mobilidades por sistema enzimático, pelas freqüências de alozimas dentro de cada sistema enzimático em relação ao total de bandas do sistema e pela análise da similaridade genética, com base na ausência e presença de bandas. Todos os sistemas enzimáticos utilizados tiveram boa resolução e definição de bandas, com ênfase para SKDH, 6PGDH, PGI e ACP. Em sua totalidade, os sistemas apresentaram polimorfismo capaz de caracterizar e identificar acessos ou grupos de pequeno número de acessos, sendo que o sistema GOT foi o que apresentou maior variabilidade de alozimas e de perfis; e o que apresentou menor variabilidade foi o sistema FUM, com três alozimas e quatro perfis. Cinqüenta e sete por cento das alozimas podem ser usadas para caracterizar e identificar clones ou grupos de clones. Cerca de 64% dos acessos analisados podem ser identificados por um a seis fenótipos individuais de sistemas enzimáticos. A análise da similaridade indicou os grupos G1, G2 e G3 como os mais divergentes da coleção, os quais são indicados para cruzamentos intraespecíficos e interespecíficos visando a obtenção de clones superiores.Título em inglês: Characterization of black pepper accessions using isozymes.     Arene oxidation with malonoyl peroxides

    Malonoyl peroxide 7, prepared in a single step from the commercially available diacid, is an effective reagent for the oxidation of aromatics. Reaction of an arene with peroxide 7 at room temperature leads to the corresponding protected phenol which can be unmasked by aminolysis. An ionic mechanism consistent with the experimental findings and supported by isotopic labeling, Hammett analysis, EPR investigations and reactivity profile studies is proposed

    All grown up? The fate after 15 years of a quarter of a million UK firms born in 1998

    The theory of firm growth is in a rather unsatisfactory state. However, the analysis of large firm-level datasets which have become available in recent years allows us to begin building an evidence base which can, in turn, be used to underpin the development of more satisfactory theory. Here we study the 239 thousand UK private sector firms born in 1998 over their first 15 years of life. A first, and quite striking, finding is the extraordinary force of mortality. By age 15, 90% of the UK firms born in 1998 are dead, and, for those surviving to age 15, the hazard of death is still about 10% a year. The chance of death is related to the size and growth of firms in an interesting way. Whilst the hazard rate after 15 years is largely independent of size at birth, it is strongly affected by the current (age 14) size. In particular, firms with more than five employees are half as likely to die in the next year as firms with less than five employees. A second important finding is that most firms, even those which survive to age 15, do not grow very much. By age 15 more than half the 26,000 survivors still have less than five jobs. In other words, the growth paths – what we call the ‘growth trajectories’ – of most of the 26,000 survivors are pretty flat. However, of the firms that do grow, firms born smaller grow faster than those born larger. Another striking finding is that growth is heavily concentrated in the first five years. Whilst growth does continue, even up to age 15, each year after age five it involves only a relatively small proportion of firms. Finally, there are two groups of survivors which contribute importantly to job creation. Some are those born relatively large (with more than 20 jobs) although their growth rate is quite modest. More striking though, is a very small group of firms born very small with less than five jobs (about 5% of all survivors) which contribute a substantial proportion (more than one third) of the jobs added to the cohort total by age 15

    Utilisation of an operative difficulty grading scale for laparoscopic cholecystectomy

    Background A reliable system for grading operative difficulty of laparoscopic cholecystectomy would standardise description of findings and reporting of outcomes. The aim of this study was to validate a difficulty grading system (Nassar scale), testing its applicability and consistency in two large prospective datasets. Methods Patient and disease-related variables and 30-day outcomes were identified in two prospective cholecystectomy databases: the multi-centre prospective cohort of 8820 patients from the recent CholeS Study and the single-surgeon series containing 4089 patients. Operative data and patient outcomes were correlated with Nassar operative difficultly scale, using Kendall’s tau for dichotomous variables, or Jonckheere–Terpstra tests for continuous variables. A ROC curve analysis was performed, to quantify the predictive accuracy of the scale for each outcome, with continuous outcomes dichotomised, prior to analysis. Results A higher operative difficulty grade was consistently associated with worse outcomes for the patients in both the reference and CholeS cohorts. The median length of stay increased from 0 to 4 days, and the 30-day complication rate from 7.6 to 24.4% as the difficulty grade increased from 1 to 4/5 (both p < 0.001). In the CholeS cohort, a higher difficulty grade was found to be most strongly associated with conversion to open and 30-day mortality (AUROC = 0.903, 0.822, respectively). On multivariable analysis, the Nassar operative difficultly scale was found to be a significant independent predictor of operative duration, conversion to open surgery, 30-day complications and 30-day reintervention (all p < 0.001). Conclusion We have shown that an operative difficulty scale can standardise the description of operative findings by multiple grades of surgeons to facilitate audit, training assessment and research. It provides a tool for reporting operative findings, disease severity and technical difficulty and can be utilised in future research to reliably compare outcomes according to case mix and intra-operative difficulty

    Unexpected Role for Helicobacter pylori DNA Polymerase I As a Source of Genetic Variability

    Helicobacter pylori, a human pathogen infecting about half of the world population, is characterised by its large intraspecies variability. Its genome plasticity has been invoked as the basis for its high adaptation capacity. Consistent with its small genome, H. pylori possesses only two bona fide DNA polymerases, Pol I and the replicative Pol III, lacking homologues of translesion synthesis DNA polymerases. Bacterial DNA polymerases I are implicated both in normal DNA replication and in DNA repair. We report that H. pylori DNA Pol I 5′- 3′ exonuclease domain is essential for viability, probably through its involvement in DNA replication. We show here that, despite the fact that it also plays crucial roles in DNA repair, Pol I contributes to genomic instability. Indeed, strains defective in the DNA polymerase activity of the protein, although sensitive to genotoxic agents, display reduced mutation frequencies. Conversely, overexpression of Pol I leads to a hypermutator phenotype. Although the purified protein displays an intrinsic fidelity during replication of undamaged DNA, it lacks a proofreading activity, allowing it to efficiently elongate mismatched primers and perform mutagenic translesion synthesis. In agreement with this finding, we show that the spontaneous mutator phenotype of a strain deficient in the removal of oxidised pyrimidines from the genome is in part dependent on the presence of an active DNA Pol I. This study provides evidence for an unexpected role of DNA polymerase I in generating genomic plasticity
