Algunos aspectos del deporte en el ritual y en la religiĂłn en el antiguo Egipto
Ritual and religion are connected to the sport of Pharaoh. During the jubilee festival, the ageing king had to demonstrate his fitness in a running ritual which also gave him new powers. In the New Kingdom, Pharaoh is compared with Month, the god of war, when practicing sport, a qualification for assuming the mantle of ruler. Enscribed on the Sphinx-stela of Amenophis II, the longest Egyptian text depicting sport, the gods Month (six times), Amun (five times) and Atum (three times) are mentioned, whereas Astarte, Geb, Horus, Maat, Ra, Reshef and Seth are only mentioned once each. Further, it may be surprising to discover that the right to rule among the gods was disputed twice by means of sporting competition, as is the case in the mythological story entitled “The Conflict of Horus and Seth”.El ritual y la religiĂłn están conectados al deporte del FaraĂłn. Murine interleukin 1 stimulates α2-macroglobulin synthesis in rat hepatocyte primary cultures
AbstractIn rat hepatocyte primary cultures recombinant interleukin 1 was found to stimulate α2-macroglobulin synthesis, whereas albumin synthesis was decreased. Quantitative promoter analysis in Physcomitrella patens: a set of plant vectors activating gene expression within three orders of magnitude
BACKGROUND: In addition to studies of plant gene function and developmental analyses, plant biotechnological use is largely dependent upon transgenic technologies. The moss Physcomitrella patens has become an exciting model system for studying plant molecular processes due to an exceptionally high rate of nuclear gene targeting by homologous recombination compared with other plants. However, its use in transgenic approaches requires expression vectors that incorporate sufficiently strong promoters. To satisfy this requirement, a set of plant expression vectors was constructed and equipped with either heterologous or endogenous promoters. RESULTS: Promoter activity was quantified using the dual-luciferase reporter assay system. The eight different heterologous promoter constructs tested exhibited expression levels spanning three orders of magnitude. Of these, the complete rice actin1 gene promoter showed the highest activity in Physcomitrella, followed by a truncated version of this promoter and three different versions of the cauliflower mosaic virus 35S promoter. In contrast, the Agrobacterium tumefaciens nopaline synthase promoter induced transcription rather weakly. Constructs including promoters commonly used in mammalian expression systems also proved to be functional in Physcomitrella. In addition, the 5' -regions of two Physcomitrella glycosyltransferases (i.e. α1,3-fucosyltransferase and β1,2-xylosyltransferase) were identified and functionally characterised in comparison to the heterologous promoters. Furthermore, motifs responsible for enhancement of translation efficiency – such as the TMV omega element and a modified sequence directly prior the start codon – were tested in this model. CONCLUSION: We developed a vector set that enables gene expression studies, both in lower and higher land plants, thus providing valuable tools applicable in both basic and applied molecular research

Background: 5-Fluorouracil (FU) is one of the most commonly used cytostatic drugs in the systemic treatment of
cancer. Treatment with FU may cause severe or life-threatening side effects and the treatment-related mortality rate is 0.2–1.0%. Summary: Among other risk factors associated
with increased toxicity, a genetic deficiency in dihydropyrimidine dehydrogenase (DPD), an enzyme responsible for
the metabolism of FU, is well known. This is due to variants
in the DPD gene (DPYD). Up to 9% of European patients carry a DPD gene variant that decreases enzyme activity, and
DPD is completely lacking in approximately 0.5% of patients.
Here we describe the clinical and genetic background and
summarize recommendations for the genetic testing and
tailoring of treatment with 5-FU derivatives. The statement
was developed as a consensus statement organized by the
German Society for Hematology and Medical Oncology in
cooperation with 13 medical associations from Austria, Germany, and Switzerland. Key Messages: (i) Patients should be
tested for the 4 most common genetic DPYD variants before
treatment with drugs containing FU. (ii) Testing forms the
basis for a differentiated, risk-adapted algorithm with recommendations for treatment with FU-containing drugs. (iii)

Surface-Confined Self-Assembly of Di-carbonitrile Polyphenyls
This Feature Article reports on the controlled formation and structure-functionality aspects of vacuum-deposited self-assembled organic and metal-organic networks at metal surfaces using ditopic linear and nonlinear molecular bricks, namely di-carbonitrile polyphenyls. Surface confined supramolecular organization of linear aromatic molecules leads to a fascinating variety of open networks. Moreover, cobalt-directed assembly of the same linear linkers reveals highly regular, open honeycomb networks with tunable pore sizes representing versatile templates for the organization of molecular guests or metal clusters and the control of supramolecular dynamers. In addition, the 2D nanopore organic networks act as arrays of quantum corrals exhibiting confinement of the surface-electronic states of the metallic substrate. A reduction of the linker symmetry leads to the formation of disordered, glassy coordination networks, which represent a structural model for amorphous materials
14th St. Gallen International Breast Cancer Conference 2015: Evidence, Controversies, Consensus - Primary Therapy of Early Breast Cancer: Opinions Expressed by German Experts
The key topics of this year's 14th St. Gallen Consensus Conference on the diagnosis and therapy of primary breast cancer were again questions about breast surgery and axillary surgery, radio-oncology and systemic therapy options in consideration of tumor biology, and the clinical application of multigene assays. This year, the consensus conference took place in Vienna. From a German perspective, it makes sense to substantiate the results of the vote of the international panel representing 19 countries in light of the updated national therapy recommendations of the AGO (Arbeitsgemeinschaft Gynäkologische Onkologie). Therefore, 14 German breast cancer experts, 3 of whom are members of the International St. Gallen Panel, have commented on the voting results of the St. Gallen Consensus Conference 2015 in relation to clinical routine in Germany
Flavopiridol Pharmacogenetics: Clinical and Functional Evidence for the Role of SLCO1B1/OATP1B1 in Flavopiridol Disposition
Flavopiridol is a cyclin-dependent kinase inhibitor in phase II clinical development for treatment of various forms of cancer. When administered with a pharmacokinetically (PK)-directed dosing schedule, flavopiridol exhibited striking activity in patients with refractory chronic lymphocytic leukemia. This study aimed to evaluate pharmacogenetic factors associated with inter-individual variability in pharmacokinetics and outcomes associated with flavopiridol therapy.Thirty-five patients who received single-agent flavopiridol via the PK-directed schedule were genotyped for 189 polymorphisms in genes encoding 56 drug metabolizing enzymes and transporters. Genotypes were evaluated in univariate and multivariate analyses as covariates in a population PK model. Transport of flavopiridol and its glucuronide metabolite was evaluated in uptake assays in HEK-293 and MDCK-II cells transiently transfected with SLCO1B1. Polymorphisms in ABCC2, ABCG2, UGT1A1, UGT1A9, and SLCO1B1 were found to significantly correlate with flavopiridol PK in univariate analysis. Transport assay results indicated both flavopiridol and flavopiridol-glucuronide are substrates of the SLCO1B1/OATP1B1 transporter. Covariates incorporated into the final population PK model included bilirubin, SLCO1B1 rs11045819 and ABCC2 rs8187710. Associations were also observed between genotype and response. To validate these findings, a second set of data with 51 patients was evaluated, and overall trends for associations between PK and PGx were found to be consistent.Polymorphisms in transport genes were found to be associated with flavopiridol disposition and outcomes. Observed clinical associations with SLCO1B1 were functionally validated indicating for the first time its relevance as a transporter of flavopiridol and its glucuronide metabolite. A second 51-patient dataset indicated similar trends between genotype in the SLCO1B1 and other candidate genes, thus providing support for these findings. Further study in larger patient populations will be necessary to fully characterize and validate the clinical impact of polymorphisms in SLCO1B1 and other transporter and metabolizing enzyme genes on outcomes from flavopiridol therapy
