    Change in interplanetary shock acceleration preceding STIP Interval 17

    The intensity and frequency of shock acceleration events in the interplanetary medium decreased dramatically in early 1985. Low energy ions were observed by IMP 8 at 1 AU and Voyagers 1 and 2 at 22 and 16 AU, respectively. Voyager 1 was at 25 deg heliographic latitude while IMP 8 and Voyager 2 were near the solar equatorial plane. The decrease in low energy shock events led to a drop in the average ion flux by a factor of 20 to 50. It started about day 10 of 1985 in the approximately .5 MeV channel on IMP8 and took approximately 75 days to reach the new, lower, background level. The decrease at the Voyagers started approximately 50 days later. The time delay between the start of the decrease at IMP and at Voyager 2 implies that decrease was convected outward with a velocity of approximately 535 km/sec. The intensity and frequency of interplanetary shock events remained at the lower level for at least 1.5 years

    Radial Velocity along the Voyager 1 Trajectory: The Effect of Solar Cycle

    As Voyager 1 and Voyager 2 are approaching the heliopause (HP)—the boundary between the solar wind (SW) and the local interstellar medium (LISM)—we expect new, unknown features of the heliospheric interface to be revealed. A seeming puzzle reported recently by Krimigis et al. concerns the unusually low, even negative, radial velocity components derived from the energetic ion distribution. Steady-state plasma models of the inner heliosheath (IHS) show that the radial velocity should not be equal to zero even at the surface of the HP. Here we demonstrate that the velocity distributions observed by Voyager 1 are consistent with time-dependent simulations of the SW-LISM interaction. In this Letter, we analyze the results from a numerical model of the large-scale heliosphere that includes solar cycle effects. Our simulations show that prolonged periods of low to negative radial velocity can exist in the IHS at substantial distances from the HP. It is also shown that Voyager 1 was more likely to observe such regions than Voyager 2

    A computer-operated binocular vision testing device was developed as one part of a system designed for NASA to evaluate the visual function of astronauts during spaceflight. This particular device, called the Mark 3 Haploscope, employs semi-automated psychophysical test procedures to measure visual acuity, stereopsis, phoria, fixation disparity, refractive state and accommodation/convergence relationships. Test procedures are self-administered and can be used repeatedly without subject memorization. The Haploscope was designed as one module of the complete NASA Vision Testing System. However, it is capable of stand-alone operation. Moreover, the compactness and portability of the Haploscope make possible its use in a broad variety of testing environments

Echokardiographisch fand sich eine dilatative Kardiomyopathie. Ein erhöhtes Serumferritin und eine Eisenüberladung in der Leberbiopsie ließen eine hereditäre Hämochromatose vermuten. Die kardiale Beteiligung konnte mittels Magnetresonanztomographie nachgewiesen werden. Genetische Abklärungen ergaben eine homozygote Mutation für G320V und bewiesen eine juvenile Hämochromatose.     Real-Time Microsensor Measurement of Local Metabolic Activities in Ex Vivo Dental Biofilms Exposed to Sucrose and Treated with Chlorhexidine

    Dental biofilms are characterized by structural and functional heterogeneity. Due to bacterial metabolism, gradients develop and diverse ecological microniches exist. The aims of this study were (i) to determine the metabolic activity of microorganisms in naturally grown dental biofilms ex vivo by measuring dissolved oxygen (DO) and pH profiles with microelectrodes with high spatial resolution and (ii) to analyze the impact of an antimicrobial chlorhexidine (CHX) treatment on microbial physiology during stimulation by sucrose in real time. Biofilms were cultivated on standardized human enamel surfaces in vivo. DO and pH profiles were measured in a flow cell system in sterile human saliva, after sucrose addition (10%), again after alternative treatment of the sucrose exposed biofilms with CHX (0.2%) for 1 or 10 min or after being killed with paraformaldehyde (4%). Biofilm structure was visualized by vitality staining with confocal microscopy. With saliva as the sole nutrient source oxygen consumption was high within the superficial biofilm layers rendering deeper layers (>220 ÎĽm) anoxic. Sucrose addition induced the thickness of the anaerobic zone to increase with a concurrent decrease in pH (7.1 to 4.4). CHX exposure reduced metabolic activity and microbial viability at the biofilm surface and drove metabolic activity deeper into the biofilm. CHX treatment led to a reduced viability at the biofilm surface with minor influence on overall biofilm physiology after 1 min; even after 10 min there was measurable respiration and fermentation inside the biofilm. However, the local microenvironment was more aerated, less acidogenic, and presumably less pathogenic

    Process Engineering of Biopharmaceutical Production in Moss Bioreactors via Model-Based Description and Evaluation of Phytohormone Impact

    The moss Physcomitrella is an interesting production host for recombinant biopharmaceuticals. Here we produced MFHR1, a synthetic complement regulator which has been proposed for the treatment of diseases associated to the complement system as part of human innate immunity. We studied the impact of different operation modes for the production process in 5 L stirred-tank photobioreactors. The total amount of recombinant protein was doubled by using fed-batch or batch compared to semi-continuous operation, although the maximum specific productivity (mg MFHR1/g FW) increased just by 35%. We proposed an unstructured kinetic model which fits accurately with the experimental data in batch and semi-continuous operation under autotrophic conditions with 2% CO2_{2} enrichment. The model is able to predict recombinant protein production, nitrate uptake and biomass growth, which is useful for process control and optimization. We investigated strategies to further increase MFHR1 production. While mixotrophic and heterotrophic conditions decreased the MFHR1-specific productivity compared to autotrophic conditions, addition of the phytohormone auxin (NAA, 10 µM) to the medium enhanced it by 470% in shaken flasks and up to 230% and 260%, in batch and fed-batch bioreactors, respectively. Supporting this finding, the auxin-synthesis inhibitor L-kynurenine (100 µM) decreased MFHR1 production significantly by 110% and 580% at day 7 and 18, respectively. Expression analysis revealed that the MFHR1 transgene, driven by the Physcomitrella actin5 (PpAct5) promoter, was upregulated 16 h after NAA addition and remained enhanced over the whole process, whereas the auxin-responsive gene PpIAA1A was upregulated within the first 2 hours, indicating that the effect of auxin on PpAct5 promoter-driven expression is indirect. Furthermore, the day of NAA supplementation was crucial, leading to an up to 8-fold increase of MFHR1-specific productivity (0.82 mg MFHR1/g fresh weight, 150 mg accumulated over 7 days) compared to the productivity reported previously. Our findings are likely to be applicable to other plant-based expression systems to increase biopharmaceutical production and yields
