    The Receptor for Urokinase Regulates TLR2 Mediated Inflammatory Responses in Neutrophils

    The urokinase-type plasminogen activator receptor (uPAR), a glycosylphosphatidylinositol (GPI) anchored membrane protein, regulates urokinase (uPA) protease activity, chemotaxis, cell-cell interactions, and phagocytosis of apoptotic cells. uPAR expression is increased in cytokine or bacteria activated cell populations, including macrophages and monocytes. However, it is unclear if uPAR has direct involvement in the response of inflammatory cells, such as neutrophils and macrophages, to Toll like receptor (TLR) stimulation. In this study, we found that uPAR is required for optimal neutrophil activation after TLR2, but not TLR4 stimulation. We found that the expression of TNF-α and IL-6 induced by TLR2 engagement in uPAR-/- neutrophils was less than that in uPAR+/+ (WT) neutrophils. Pretreatment of neutrophils with PI-PLC, which cleaves GPI moieties, significantly decreased TLR2 induced expression of TNF-α in WT neutrophils, but demonstrated only marginal effects on TNF-α expression in PAM treated uPAR-/- neutrophils. IκB-α degradation and NF-κB activation were not different in uPAR-/- or WT neutrophils after TLR2 stimulation. However, uPAR is required for optimal p38 MAPK activation after TLR2 engagement. Consistent with the in vitro findings that uPAR modulates TLR2 engagement induced neutrophil activation, we found that pulmonary and systemic inflammation induced by TLR2, but not TLR4 stimulation is reduced in uPAR-/- mice compared to WT counterparts. Therefore, our data suggest that neutrophil associated uPAR could be a potential target for treating acute inflammation, sepsis, and organ injury related to severe bacterial and other microbial infections in which TLR2 engagement plays a major role

    Isospin dependence of projectile-like fragment production at intermediate energies

    The cross sections of fragments produced in 140 AA MeV 40,48^{40,48}Ca + 9^9Be and 58,64^{58,64}Ni + 9^9Be reactions are calculated by the statistical abration-ablation(SAA) model and compared to the experimental results measured at the National Superconducting Cyclotron Laboratory (NSCL) at Michigan State University. The fragment isotopic and isotonic cross section distributions of 40^{40}Ca and 48^{48}Ca, 58^{58}Ni and 64^{64}Ni, 40^{40}Ca and 58^{58}Ni, and 48^{48}Ca and 64^{64}Ni are compared and the isospin dependence of the projectile fragmentation is studied. It is found that the isospin dependence decreases and disappears in the central collisions. The shapes of the fragment isotopic and isotonic cross section distributions are found to be very similar for symmetric projectile nuclei. The shapes of the fragment isotopic and isotonic distributions of different asymmetric projectiles produced in peripheral reactions are found very similar. The similarity of the distributions are related to the similar proton and neutron density distributions inside the nucleus in framework of the SAA model.Comment: 7 pages, 4 figures; to be published in Phys Rev

    Global Stability of a Variation Epidemic Spreading Model on Complex Networks

    Epidemic spreading on networks becomes a hot issue of nonlinear systems, which has attracted many researchers’ attention in recent years. A novel epidemic spreading model with variant factors in complex networks is proposed and investigated in this paper. One main feature of this model is that virus variation is investigated in the process of epidemic dynamical spreading. The global dynamics of this model involving an endemic equilibrium and a disease-free equilibrium are, respectively, discussed. Some sufficient conditions are given for the existence of the endemic equilibrium. In addition, the global asymptotic stability problems of the disease-free equilibrium and the endemic equilibrium are also investigated by the Routh-Hurwitz stability criterion and Lyapunov stability criterion. And the uniform persistence condition of the new system is studied. Finally, numerical simulations are provided to illustrate obtained theoretical results

    In Vivo Study of the Effects of Peptide-Conjugated Near-Infrared Fluorescent Quantum Dots on the Tumorigenic and Lymphatic Metastatic Capacities of Squamous Cell Carcinoma Cell Line Tca8113 and U14

    Quantum dots (QDs) have great potential in non-invasive monitoring and imaging of tumor cells in vivo, but it is unknown if QDs affect their tumorigenesis and metastasis. Here, we applied peptide-conjugated near-infrared fluorescent QDs (NIRF-QDs) to label the squamous cell carcinoma cells Tca8113 and U14. We tested the proliferation and apoptotic capacities of both cells, and the capacity of cervical lymph node metastasis after tumorigenesis in U14 cells’. We find that QDs do not affect the tumor cells’ capacities to grow, proliferate, and metastasize. Our study provides critical data to support the application of NIRF-QDs in non-invasive monitoring and imaging of tumor cells in vivo

    Single top quark production via SUSY-QCD FCNC couplings at the CERN LHC in the unconstrained MSSM

    We evaluate the tcˉt\bar{c} and tuˉt\bar{u} productions at the LHC within the general unconstrained MSSM framework. We find that these single top quark productions induced by SUSY-QCD FCNC couplings have remarkable cross sections for favorable parameter values allowed by current low energy data, which can be as large as a few pb. Once large rates of the tcˉt\bar{c} and tuˉt\bar{u} productions are detected at the LHC, they may be induced by SUSY FCNC couplings. We show that the precise measurement of single top quark production cross sections at the LHC is a powerful probe for the details of the SUSY FCNC couplings.Comment: 39 pages, 10 figures.     Induced Pluripotent Stem Cell Transplantation Improves Locomotor Recovery in Rat Models of Spinal Cord Injury: a Systematic Review and Meta-Analysis of Randomized Controlled Trials

    Background/Aims: Spinal cord injury (SCI) has long been a subject of great interest in a wide range of scientific fields. Several attempts have been made to demonstrate motor function improvement in rats with SCI after transplantation of induced pluripotent stem cells (iPSC). This systematic review and meta-analysis was designed to summarize the effects of iPSC on locomotor recovery in rat models of SCI. Methods: We searched the publications in the PubMed, Medline, Science Citation Index, Cochrane Library, CNKI, and Wan-fang databases and the China Biology Medicine disc. Results were analyzed by Review Manager 5.3.0. The quality of evidence was assessed using the Grading of Recommendations Assessment, Development and Evaluation (GRADE) methodology. Results: Six randomized controlled preclinical trials covering eight comparisons and including 212 rats were selected. The subgroup analyses were based on the following items: different SCI models, cell counts, iPSC sources, iPSC differentiations and transplantation methods. The pooled results indicated that iPSC transplantation significantly improved locomotor recovery of rats after SCI by sustaining beneficial effects, especially in the subgroups of contusion, moderate cell counts (5×105), source of human fetal lung fibroblasts, iPSC-neural precursors and intraspinal injection. Conclusion: Our meta-analysis of the effects of iPSC transplantation on locomotor function in SCI models is, to our knowledge, the first meta-analysis in this field. We conclude that iPSC transplantation improves locomotor recovery in rats with SCI, implicating this strategy as an effective therapy. However, more studies are required to validate our conclusions
