    Genetic counseling for individuals with hemoglobin disorders and for their relatives: a systematic literature review

    Objective: To identify genetic counseling programs that do not encourage therapeutic abortion for individuals with hemoglobin disorders and/or for their relatives. Method: Systematic literature review of articles published from 2001 to 2012 that are located in the PubMed, LILACS, SciELO and SCOPUS databases using keywords in Portuguese, English and Spanish and that met the inclusion and exclusion criteria described on a standardized form. Results: A total of 409 articles were located, but only eight (1.9%) were selected for analysis. Conclusion: Although seldom mentioned in the literature, educational/preventive programs targeting hemoglobinopathies are feasible and allow the affected individuals to acquire knowledge on the consequences of this condition and their odds of transmitting it.

Objetivo Identificar programas de aconselhamento genético sem incentivo ao aborto terapêutico para indivíduos com alterações das hemoglobinas e/ou seus familiares. Método Revisão sistemática de literatura realizada nas bases de dados PubMed, LILACS, SciELO e SCOPUS por artigos publicados de 2001 a 2012, utilizando descritores em português, inglês e espanhol, de acordo com formulários padronizados contendo critérios de inclusão e exclusão.     Prevalência de hemoglobinas variantes em estudo realizado no estado de Goiás

    Hemoglobinas variantes são as alterações genéticas humanas mais comuns no Brasil, com influência na economia das famílias e dos governos. Entendendo que a formação do povo goiano teve a contribuição de diferentes etnias, este estudo teve como objetivo estabelecer a prevalência das hemoglobinas variantes em uma amostragem populacional atendida durante um evento científico na Universidade Federal de Goiás. A prevalência da heterozigose para a hemoglobina “S” foi de 4,4% e a da heterozigose para a hemoglobina “C” foi de 1%. Após a obtenção dos resultados, os participantes da pesquisa foram alertados sobre a importância do conhecimento acerca do diagnóstico de hemoglobinopatias. Os resultados obtidos demonstram a necessidade de implantação de serviços públicos preventivos e de acompanhamento específico em hemoglobinopatias no estado de Goiás, bem como para a relevância das atividades de extensão universitária para a comunidade

    Cytotoxicity and genotoxicity of intravitreal adalimumab administration in rabbit retinal cells

    Purpose: To assess the cytotoxicity and genotoxicity of intravitreal adalimumab treatment in an animal experimental model using cytological and molecular techniques. Methods: Eighteen rabbits were randomly assigned to three groups: control, adalimumab treatment, and placebo. Cytotoxicity on retinal cells was evaluated using flow cytometry assays to determine the level of apoptosis and necrosis. Genotoxicity was evaluated by comet assays to assess DNA damage, and quantitative real-time polymerase chain reaction (qPCR) was used to evaluate expression of apoptosis-inducing caspases (8 and 3). Results: No cytotoxicity or genotoxicity was observed in any of the two treatment groups (adalimumab and placebo) following intravitreal administration compared with the control group. Flow cytometry analysis revealed that more than 90% of the cells were viable, and only a low proportion of retinal cells presented apoptotic (~10%) or necrotic (<1%) activity across all groups. Molecular damage was also low with a maximum of 6.4% DNA degradation observed in the comet assays. In addition, no increase in gene expression of apoptosis-inducing caspases was observed on retinal cells by qPCR in both the adalimumab and placebo groups compared with the control group. Conclusion: The use of adalimumab resulted in no detectable cytotoxicity or genotoxicity on retinal cells for up to 60 days upon administration. These results therefore indicate that adalimumab may be a safe option for intravitreal application to treat ocular inflammatory diseases in which TNF-α is involved

    Occurrence and seasonal monitoring of helminthes in the main rivers and lakes of Goiânia, Goiás, Brazil, 2007

    Water constitutes one of the fundamental elements for human existence, which is demonstrated by its multiple uses, for public supply, industry, agriculture, livestock, recreation, and transportation. Nevertheless, contamination from various sources has been occurring for years impairing this resource and considerably increasing the risk of waterborne diseases caused mainly by bacteria, viruses, protozoa, and helminthes. Seasonal monitoring of helminthes is relevant to understand environmental contamination processes and provide information for decision-making measures to block the access of these parasites and other pathogens to man and animals. This study aimed at using parasitologic techniques to analyze the presence of helminthes and verify the water quality of the main bodies of water in Goiânia, the capital of the state of Goiás, Brazil. Samples were collected monthly from February 2006 to January 2007 in six pre-set points, in a total of 72 samples. Of these, 17 (23.61%) were positive for free-living larvae and 2 (2.77%) for Ascaris sp. and Hymenolepis diminuta. This study showed that the rivers and lakes of Goiânia are contaminated by eggs and/or larvae of helminthes

    Optical Properties And Antiangiogenic Activity Of A Chalcone Derivate

    Chalcones and their derivatives exhibit numerous pharmacological activities such as antibacterial, antifungal, cytotoxic, antinociceptive and anti-inflammatory. Recently, they have been assessed aiming for novel application in nonlinear optics and in the treatment of immune diseases and cancers. In this study, we investigate the optical properties of synthetic chalcona 1E,4E-1-(4-chlorophenyl)-5-(2,6,6-trimethylcyclohexen-1-yl)penta-1,4-dien-3-one (CAB7β) and its antiangiogenic potential using the chorioallantoic membrane (CAM) with the S180 sarcoma cell line. Experimental and theoretical results show intense absorption in the UVA-UVC region, which is associated with a π → π* transition with intramolecular charge transfer from the trimethyl-cyclohexen-1-yl ring to the chlorophenyl ring. Quantum chemical calculations of the first hyperpolarizability, accounting for both solvent and frequency dispersion effects, are in very good concordance with hyper-Rayleigh scattering measurements. In addition, two-photon absorption allowed band centered at 650 nm was observed. Concerning antiangiogenic activity, CAB7β causes a significant reduction in the total number, junctions, length and caliber of blood vessels stimulated by S180 cells reducing the presence of blood vessels, inflammatory cells and others elements related to angiogenic process. It is found that CAB7β is a versatile compound and a promising candidate for linear and nonlinear optical applications, in therapy against sarcoma and phototherapy

    Molecular epidemiological investigation of Mayaro virus in febrile patients from Goiania City, 2017-2018.

    Mayaro virus (MAYV) has historically been associated with sylvatic transmission; however, urban outbreaks have been reported in Brazil, including cases of co-detection with dengue virus (DENV). Therefore, we performed a molecular survey to investigate MAYV circulation and cocirculation with DENV within Goiania, a major city in Central-West Brazil. Among 375 subjects with arbovirus-like symptoms, 259 were positive for DENV and 26 for MAYV. Of these, 17 were coinfected with DENV-2, suggesting co-transmission of the viruses. The most common complaints at the time of inclusion were myalgia, headache, fever, arthralgia, retro-orbital pain, and skin rash. No specific symptoms were associated with MAYV when either detected alone or co-detected with DENV, compared to that when DENV was detected alone. Most MAYV-infected subjects were women with no recent travel history to rural/sylvatic areas. Phylogenetic reconstruction indicated that the MAYV identified in this study is closely related with a lineage observed in Peru, belonging to genotype D. Our results corroborate the growing circulation of MAYV in urban environments in Brazil and reinforce the need to implement laboratory diagnosis in the Unified Health System, considering that the clinical manifestations of Mayaro fever are similar to those of other arboviruses, particularly dengue. Furthermore, most cases occurred in association with DENV-2. Further phylogenetic studies are needed to evaluate MAYV, which has not been widely examined

    et al.Four new Schiff-base ligands have been designed and prepared by condensation reaction between hydrazine derivatives (i.e. 2-hydrazinopyridine or 2-hydrazinoquinoline) and mono- or dialdehyde (3-tert-butyl-2-hydroxybenzaldehyde and 5-tert-butyl-2-hydroxyisophthalaldehyde, respectively). Six copper(ii) coordination compounds of various nuclearities have been obtained from these ligands, which are formulated as [Cu(L1)Cl](CH3OH) (1), [Cu(L2)NO3] (2), [Cu2(L3)(ClO4)2(CH3O)(CH3OH)](CH3OH) (3), [Cu2(L4)(ClO4)(OH)(CH3OH)](ClO4) (4), [Cu8(L3)4(NO3)4(OH)5](NO3)3(CH3OH)5(H2O)8 (5) and [Cu3(HL2′)4Cl6](CH3OH)6 (6), as revealed by single-crystal X-ray studies. Their DNA-interacting abilities have been investigated using different characterization techniques, which suggest that the metal complexes act as efficient DNA binders.
