    Detection of Leishmania infantum by PCR, serology and cellular immune response in a cohort study of Brazilian dogs

    The sensitivity and specificity of PCR, serology (ELISA) and lymphoproliferative response to Leishmania antigen for the detection of Leishmania infantum infection were evaluated in a cohort of 126 dogs exposed to natural infection in Brazil. For PCR, Leishmania DNA from bone-marrow was amplified with both minicircle and ribosomal primers. The infection status and time of infection of each dog were estimated from longitudinal data. The sensitivity of PCR in parasite-positive samples was 98%. However, the overall sensitivity of PCR in post-infection samples, from dogs with confirmed infection, was only 68%. The sensitivity of PCR varied during the course of infection, being highest (78–88%) 0–135 days post-infection and declining to around 50% after 300 days. The sensitivity of PCR also varied between dogs, and was highest in sick dogs. The sensitivity of serology was similar in parasite-positive (84%), PCR-positive (86%) and post-infection (88%) samples. The sensitivity of serology varied during the course of infection, being lowest at the time of infection and high (93–100%) thereafter. Problems in determining the specificity of serology are discussed. The sensitivity and specificity of cellular responsiveness were low. These data suggest that PCR is most useful in detecting active or symptomatic infection, and that serology can be a more sensitive technique for the detection of all infected dogs

    Hypercholesterolaemia and Atherosclerosis Induced in Vervet l\'lonkeys by Cholesterol-free, Semisynthetic Diets

    Vervet monkeys (Cercopithecus aethiops pygerethrus) were fed diets containing 40% carbohydrate, 25% casein and 14% hydrogenated coconut oil for 6 months. Three carbohydrates were fed: glucose, fructose and sucrose. All three diets were cholesterolaemic, the most severe cholesterolaemias being observed in the fructose and sucrose groups. All diets led to aortic sudanophilia. The fructose diet resulted in the most severe atherosclerosis and sudanophilia. This study' demonstrates the feasibility of using semisynthetic, cholesterol-free diets for the induction of hyperlipaemia and atherosclerosis in monkeys

    Data access and integration in the ISPIDER proteomics grid

    Grid computing has great potential for supporting the integration of complex, fast changing biological data repositories to enable distributed data analysis. One scenario where Grid computing has such potential is provided by proteomics resources which are rapidly being developed with the emergence of affordable, reliable methods to study the proteome. The protein identifications arising from these methods derive from multiple repositories which need to be integrated to enable uniform access to them. A number of technologies exist which enable these resources to be accessed in a Grid environment, but the independent development of these resources means that significant data integration challenges, such as heterogeneity and schema evolution, have to be met. This paper presents an architecture which supports the combined use of Grid data access (OGSA-DAI), Grid distributed querying (OGSA-DQP) and data integration (AutoMed) software tools to support distributed data analysis. We discuss the application of this architecture for the integration of several autonomous proteomics data resources

    WASP restricts active Rac to maintain cells' front-rear polarization

    YesEfficient motility requires polarized cells, with pseudopods at the front and a retracting rear. Polarization is maintained by restricting the pseudopod catalyst, active Rac, to the front. Here, we show that the actin nucleation-promoting factor Wiskott-Aldrich syndrome protein (WASP) contributes to maintenance of front-rear polarity by controlling localization and cellular levels of active Rac. Dictyostelium cells lacking WASP inappropriately activate Rac at the rear, which affects their polarity and speed. WASP’s Cdc42 and Rac interacting binding (“CRIB”) motif has been thought to be essential for its activation. However, we show that the CRIB motif’s biological role is unexpectedly complex. WASP CRIB mutants are no longer able to restrict Rac activity to the front, and cannot generate new pseudopods when SCAR/WAVE is absent. Overall levels of Rac activity also increase when WASP is unable to bind to Rac. However, WASP without a functional CRIB domain localizes normally at clathrin pits during endocytosis, and activates Arp2/3 complex. Similarly, chemical inhibition of Rac does not affect WASP localization or activation at sites of endocytosis. Thus, the interaction between small GTPases and WASP is more complex than previously thought—Rac regulates a subset of WASP functions, but WASP reciprocally restricts active Rac through its CRIB motif.Cancer Research UK grants A15672, A24450, and multidisciplinary grant A20017

    On the reheating stage after inflation

    We point out that inflaton decay products acquire plasma masses during the reheating phase following inflation. The plasma masses may render inflaton decay kinematicaly forbidden, causing the temperature to remain frozen for a period at a plateau value. We show that the final reheating temperature may be uniquely determined by the inflaton mass, and may not depend on its coupling. Our findings have important implications for the thermal production of dangerous relics during reheating (e.g., gravitinos), for extracting bounds on particle physics models of inflation from Cosmic Microwave Background anisotropy data, for the production of massive dark matter candidates during reheating, and for models of baryogenesis or leptogensis where massive particles are produced during reheating.Comment: 8 pages, 2 figures.     From weak-scale observables to leptogenesis

    Thermal leptogenesis is an attractive mechanism for generating the baryon asymmetry of the Universe. However, in supersymmetric models, the parameter space is severely restricted by the gravitino bound on the reheat temperature TRHT_{RH}. For hierarchical light neutrino masses, it is shown that thermal leptogenesis {\it can} work when TRH109T_{RH} \sim 10^{9} GeV. The low-energy observable consequences of this scenario are BR(τγ)108109 BR(\tau \to \ell \gamma) \sim 10^{-8} - 10^{-9} . For higher TRHT_{RH}, thermal leptogenesis works in a larger area of parameter space, whose observable consequences are more ambiguous. A parametrisation of the seesaw in terms of weak-scale inputs is used, so the results are independent of the texture chosen for the GUT-scale Yukawa matrices.Comment: a few references adde

    Present and Future Bounds on Non-Standard Neutrino Interactions

    We consider Non-Standard neutrino Interactions (NSI), described by four-fermion operators of the form (νˉαγνβ)(fˉγf)(\bar{\nu}_{\alpha} \gamma {\nu}_{\beta}) (\bar{f} \gamma f), where ff is an electron or first generation quark. We assume these operators are generated at dimension 8\geq 8, so the related vertices involving charged leptons, obtained by an SU(2) transformation νδeδ\nu_{\delta} \to e_{\delta}, do not appear at tree level. These related vertices necessarily arise at one loop, via WW exchange. We catalogue current constraints from sin2θW\sin^2 \theta_W measurements in neutrino scattering, from atmospheric neutrino observations, from LEP, and from bounds on the related charged lepton operators. We estimate future bounds from comparing KamLAND and solar neutrino data, and from measuring sin2θW\sin^2 \theta_W at the near detector of a neutrino factory. Operators constructed with νμ\nu_\mu and νe\nu_e should not confuse the determination of oscillation parameters at a ν\nufactory, because the processes we consider are more sensitive than oscillations at the far detector.     νdμΔ++n\nu d \to \mu^- \Delta^{++} n Reaction and Axial Vector NΔN-\Delta Coupling

    The reaction νdμΔ++n\nu d \to \mu^- \Delta^{++} n is studied in the region of low q2q^2 to investigate the effect of deuteron structure and width of the Δ\Delta resonance on the differential cross section. The results are used to extract the axial vector NΔN-\Delta coupling C5AC^{A}_5 from the experimental data on this reaction. The possibility to determine this coupling from electroweak interaction experiments with high intensity electron accelerators is discussed.Comment: 14 pages, REVTEX, 5 figure

    Proton-proton bremsstrahlung below and above pion-threshold: the influence of the Δ\Delta-isobar

    The proton-proton bremsstrahlung is investigated within a coupled-channel model with the Δ\Delta degree of freedom. The model is consistent with the NNNN scattering up to 1 GeV and the γNΔ\gamma N\Delta vertex determined in the study of pion photoproduction reactions. It is found that the Δ\Delta excitation can significantly improve the agreements with the ppppγpp \rightarrow pp\gamma at Elab=280E_{lab}=280 MeV.
