In-depth comparison of the metabolic and pharmacokinetic behaviour of the structurally related synthetic cannabinoids AMB-FUBINACA and AMB-CHMICA in rats
Synthetic cannabinoids receptor agonists (SCRAs) are often almost completely metabolised, and hence their pharmacokinetics should be carefully evaluated for determining the most adequate biomarker in toxicological analysis. Two structurally related SCRAs, AMB-FUBINACA and AMB-CHMICA, were selected to evaluate their in vivo metabolism and pharmacokinetics using male Sprague-Dawley rats. Brain, liver, kidney, blood (serum) and urine samples were collected at different times to assess the differences in metabolism, metabolic reactions, tissue distribution and excretion. Both compounds experimented O-demethyl reaction, which occurred more rapidly for AMB-FUBINACA. The parent compounds and O-demethyl metabolites were highly bioaccumulated in liver, and were still detected in this tissue 48 h after injection. The different indazole/indole N-functionalisation produced diverse metabolic reactions in this moiety and thus, different urinary metabolites were formed. Out of the two compounds, AMB-FUBINACA seemed to easily cross the blood-brain barrier, presenting higher brain/serum concentrations ratio than AMB-CHMICA
Extremely metal-poor Lyman limit system at z = 2.917 toward the quasar HE 0940-1050
We report on detailed Monte Carlo inversion analysis of the Lyman limit
system observed at z = 2.917 in the VLT/UVES spectrum of the quasar HE
0940-1050. Metal absorption lines of carbon and silicon in three ionization
stages and numerous atomic hydrogen lines have been analyzed simultaneously. It
is found that in order to match the observations, the shape of the ultraviolet
background ionizing spectrum of Haardt & Madau (1996) should be modified: a
spectrum with a higher intensity of the emission feature at 3 Ryd is required.
It is also found that synthetic galactic spectra (or different mixtures of them
with power law spectra) cannot reproduce the observations, indicating that the
stellar contribution to the ionizing background is negligible at z ~= 3. For
the first time a very low carbon abundance of [C/H] = -2.93+/-0.13 and the
abundance ratio [Si/C] = 0.35+/-0.15 are directly measured in the Lyman limit
system with N(H I) = 3.2 10^{17} cm^{-2}. If the absorber at z = 2.917 provides
an example of a pristine gas enriched by the nucleosynthetic products of early
generations of stars, then the measured value of [Si/C] seems to indicate that
the initial mass functions for these stellar populations are constrained to
intermediate masses, M_up <= 25M_solar.Comment: 7 pages, 4 figures, A&A in pres
Exploring the spreading layer of GX 9+9 using RXTE and INTEGRAL
We have fitted ~200 RXTE and INTEGRAL spectra of the neutron star LMXB GX 9+9
from 2002-2007 with a model consisting of a disc blackbody and another
blackbody representing the spreading layer (SL), i.e. an extended accretion
zone on the NS surface as opposed to the more traditional disc-like boundary
layer. Contrary to theory, the SL temperature was seen to increase towards low
SL luminosities, while the approximate angular extent had a nearly linear
luminosity dependency. Comptonization was not required to adequately fit these
spectra. Together with the ~70 degree upper bound of inclination implied by the
lack of eclipses, the best-fitting normalization of the accretion disc
blackbody component implies a distance of ~10 kpc, instead of the usually
quoted 5 kpc.Comment: 14 pages, 10 figures, accepted for publication in MNRA
