    BACKGROUND: Pax7 is a transcription factor involved in the specification and maintenance of muscle stem cells (MuSCs). Upon injury, MuSCs leave their quiescent state, downregulate Pax7 and differentiate, contributing to skeletal muscle regeneration. In the majority of regeneration studies, MuSCs are isolated by fluorescence-activated sorting (FACS), based on cell surface markers. It is known that MuSCs are a heterogeneous population and only a small percentage of isolated cells are true stem cells that are able to self-renew. A strong Pax7 reporter line would be valuable to study the in vivo behavior of Pax7-expressing stem cells. METHODS: We generated and characterized the muscle properties of a new transgenic Pax7EGFP mouse. Utilizing traditional immunofluorescence assays, we analyzed whole embryos and muscle sections by fluorescence microscopy, in addition to whole skeletal muscles by 2-photon microscopy, to detect the specificity of EGFP expression. Skeletal muscles from Pax7EGFP mice were also evaluated in steady state and under injury conditions. Finally, MuSCs-derived from Pax7EGFP and control mice were sorted and analyzed by FACS and their myogenic activity was comparatively examined. RESULTS: Our studies provide a new Pax7 reporter line with robust EGFP expression, detectable by both flow cytometry and fluorescence microscopy. Pax7EGFP-derived MuSCs have identical properties to that of wild-type MuSCs, both in vitro and in vivo, excluding any positional effect due to the transgene insertion. Furthermore, we demonstrated high specificity of EGFP to label MuSCs in a temporal manner that recapitulates the reported Pax7 expression pattern. Interestingly, immunofluorescence analysis showed that the robust expression of EGFP marks cells in the satellite cell position of adult muscles in fixed and live tissues. CONCLUSIONS: This mouse could be an invaluable tool for the study of a variety of questions related to MuSC biology, including but not limited to population heterogeneity, polarity, aging, regeneration, and motility, either by itself or in combination with mice harboring additional genetic alterations

    Factors affecting the recovery of Mexican wolves in the Southwest United States

    Recovering and maintaining large carnivore populations is a global conservation challenge that requires better knowledge of the factors affecting their populations, particularly in shared landscapes (i.e. non-protected areas where people occupy and or utilize the land). The Mexican wolf (Canis lupus baileyi) is an endangered wolf subspecies being recovered on shared landscapes in the Southwest United States and Mexico. We used data from the U.S. program to model population growth, evaluate the impact of management removal and illegal killing relative to other demographic factors, and test hypotheses about factors influencing rates of management removal and illegal killing. From 1998 to 2019, the population growth averaged 12% per year. Rates of natural reproduction, illegal killing and other mortality remained consistent over the 22 years; while releases, translocations and management removals varied markedly between two time periods, phase 1:1998–2007 and phase 2:2008–2019. The number of wolves removed for conflict management was higher during phase 1 (average ~ 13 per year, rate = 24.8%) than phase 2 (average of ~5 per year, rate = 5.2%). This decrease in management removal resulted in the wolf population resuming growth after a period of population stagnation. Two factors influenced this decrease, a change in policy regarding removal of wolves (stronger modelling support) and a decrease in the number of captive-reared adult wolves released into the wild (weaker modelling support). Illegal mortality was relatively constant across both phases, but after the decrease in management removal, illegal mortality became the most important factor (relative importance shifted from 28.2% to 50.1%). Illegal mortality was positively correlated with rates of reintroduction and translocation of wolves and negatively correlated with the rate of management removal. 6. Synthesis and applications. Using management removal to reduce human–carnivore conflict can have negative population impacts if not used judiciously. Recovering and maintaining carnivore populations in shared landscapes may require greater tolerance of conflict and more emphasis on effective conflict prevention strategies and compensation programs for affected stakeholders

    The Influence of Climate and Livestock Reservoirs on Human Cases of Giardiasis.

    Giardia duodenalis is an intestinal parasite which causes diarrhoeal illness in people. Zoonotic subtypes found in livestock may contribute to human disease occurrence through runoff of manure into multi-use surface water. This study investigated temporal associations among selected environmental variables and G. duodenalis occurrence in livestock reservoirs on human giardiasis incidence using data collected in the Waterloo Health Region, Ontario, Canada. The study objectives were to: (1) evaluate associations between human cases and environmental variables between 1 June 2006 and 31 December 2013, and (2) evaluate associations between human cases, environmental variables and livestock reservoirs using a subset of this time series, with both analyses controlling for seasonal and long-term trends. Human disease incidence exhibited a seasonal trend but no annual trend. A Poisson multivariable regression model identified an inverse association with water level lagged by 1 month (IRR = 0.10, 95% CI 0.01, 0.85, P < 0.05). Case crossover analysis found varying associations between lagged variables including livestock reservoirs (1 week), mean air temperature (3 weeks), river water level (1 week) and flow rate (1 week), and precipitation (4 weeks). This study contributes to our understanding of epidemiologic relationships influencing human giardiasis cases in Ontario, Canada

    Superhydrophobic surfaces as a source of airborne singlet oxygen through free space for photodynamic therapy

    A superhydrophobic (SH) sandwich system has been developed to enable "contact-free" airborne singlet oxygen (1O2) delivery to a water droplet. The contact-free feature means that the sensitizer is physically separated from the droplet, which presents opportunities for photodynamic therapy (PDT). Trapping of airborne 1O2 in a H2O droplet residing on a lower SH surface was monitored with 9,10-anthracene dipropionate dianion by varying distances to an upper 1O2-generating surface. Short distances of 20 ÎĽm efficiently delivered airborne 1O2 to the droplet in single-digit picomolar steady-state concentrations. Delivery decreases linearly with distance, but 50% of the 1O2 steady-state concentration is trapped at a distance of 300 ÎĽm from the generating surface. The 1270 nm luminescence intensity was measured within the SH sandwich system, confirming the presence of airborne 1O2. Physical quenching of 1O2 to ground-state 3O2 by the water droplet itself and both physical and chemical quenching of 1O2 by the water droplet containing the trap 9,10-anthracene dipropionate dianion are observed. Unlike a majority of work in the field of PDT with dissolved sensitizers, where 1O2 diffuses short (hundreds of nanometers) distances, we show the delivery of airborne 1O2 via a superhydrophobic surface is effective through air in tenths of millimeters distances to oxidize an organic compound in water.     Gene expression profiling of NaĂŻve sheep genetically resistant and susceptible to gastrointestinal nematodes

    BACKGROUND: Gastrointestinal nematodes constitute a major cause of morbidity and mortality in grazing ruminants. Individual animals or breeds, however, are known to differ in their resistance to infection. Gene expression profiling allows us to examine large numbers of transcripts simultaneously in order to identify those transcripts that contribute to an animal's susceptibility or resistance. RESULTS: With the goal of identifying genes with a differential pattern of expression between sheep genetically resistant and susceptible to gastrointestinal nematodes, a 20,000 spot ovine cDNA microarray was constructed. This array was used to interrogate the expression of 9,238 known genes in duodenum tissue of four resistant and four susceptible female lambs. NaĂŻve animals were used in order to look at genes that were differentially expressed in the absence of infection with gastrointestinal nematodes. Forty one unique known genes were identified that were differentially expressed between the resistant and susceptible animals. Northern blotting of a selection of the genes confirmed differential expression. The differentially expressed genes had a variety of functions, although many genes relating to the stress response and response to stimulus were more highly expressed in the susceptible animals. CONCLUSION: We have constructed the first reported ovine microarray and used this array to examine gene expression in lambs genetically resistant and susceptible to gastrointestinal nematode infection. This study indicates that susceptible animals appear to be generating a hyper-sensitive immune response to non-nematode challenges. The gastrointestinal tract of susceptible animals is therefore under stress and compromised even in the absence of gastrointestinal nematodes. These factors may contribute to the genetic susceptibility of these animals

    Dose-Response of Aerobic Exercise on Cognition: A Community-Based, Pilot Randomized Controlled Trial

    Epidemiological studies suggest a dose-response relationship exists between physical activity and cognitive outcomes. However, no direct data from randomized trials exists to support these indirect observations. The purpose of this study was to explore the possible relationship of aerobic exercise dose on cognition. Underactive or sedentary participants without cognitive impairment were randomized to one of four groups: no-change control, 75, 150, and 225 minutes per week of moderate-intensity semi-supervised aerobic exercise for 26-weeks in a community setting. Cognitive outcomes were latent residual scores derived from a battery of 16 cognitive tests: Verbal Memory, Visuospatial Processing, Simple Attention, Set Maintenance and Shifting, and Reasoning. Other outcome measures were cardiorespiratory fitness (peak oxygen consumption) and measures of function functional health. In intent-to-treat (ITT) analyses (n = 101), cardiorespiratory fitness increased and perceived disability decreased in a dose-dependent manner across the 4 groups. No other exercise-related effects were observed in ITT analyses. Analyses restricted to individuals who exercised per-protocol (n = 77) demonstrated that Simple Attention improved equivalently across all exercise groups compared to controls and a dose-response relationship was present for Visuospatial Processing. A clear dose-response relationship exists between exercise and cardiorespiratory fitness. Cognitive benefits were apparent at low doses with possible increased benefits in visuospatial function at higher doses but only in those who adhered to the exercise protocol. An individual’s cardiorespiratory fitness response was a better predictor of cognitive gains than exercise dose (i.e., duration) and thus maximizing an individual’s cardiorespiratory fitness may be an important therapeutic target for achieving cognitive benefits

    CARMA Measurements of the Sunyaev-Zel'dovich Effect in RXJ1347.5-1145

    We demonstrate the Sunyaev-Zel'dovich (SZ) effect imaging capabilities of the Combined Array for Research in Millimeter-wave Astronomy (CARMA) by presenting an SZ map of the galaxy cluster RXJ1347.5-1145. By combining data from multiple CARMA bands and configurations, we are able to capture the structure of this cluster over a wide range of angular scales, from its bulk properties to its core morphology. We find that roughly 9% of this cluster's thermal energy is associated with sub-arcminute-scale structure imparted by a merger, illustrating the value of high-resolution SZ measurements for pursuing cluster astrophysics and for understanding the scatter in SZ scaling relations. We also find that the cluster's SZ signal is lower in amplitude than suggested by a spherically-symmetric model derived from X-ray data, consistent with compression along the line of sight relative to the plane of the sky. Finally, we discuss the impact of upgrades currently in progress that will further enhance CARMA's power as an SZ imaging instrument.Comment: 8 pages, 6 figure
