    Depression and Oral FTC/TDF Pre-exposure Prophylaxis (PrEP) Among Men and Transgender Women Who Have Sex With Men (MSM/TGW).

    We conducted a longitudinal and cross-sectional analysis of depressive symptomology in iPrEx, a randomized, placebo-controlled trial of daily, oral FTC/TDF HIV pre-exposure prophylaxis (PrEP) in men and transgender women who have sex with men. Depression-related adverse events (AEs) were the most frequently reported severe or life-threatening AEs and were not associated with being randomized to the FTC/TDF arm (152 vs. 144 respectively OR 0.66 95 % CI 0.35-1.25). Center for Epidemiologic Studies Depression scale (CES-D) and a four questions suicidal ideation scale scores did not differ by arm. Participants reporting forced sex at anal sexual debut had higher CES-D scores (coeff: 3.23; 95 % CI 1.24-5.23) and were more likely to have suicidal ideation (OR 2.2; 95 % CI 1.09-4.26). CES-D scores were higher among people reporting non-condom receptive anal intercourse (ncRAI) (OR 1.46; 95 % CI 1.09-1.94). We recommend continuing PrEP during periods of depression in conjunction with provision of mental health services

    (C^Npz^C)AuIII complexes of acyclic carbene ligands: synthesis and anticancer properties

    A series of cyclometallated gold(III) complexes supported by pyrazine-based (C^Npz^C)-type pincer ligands were synthesized via two different pathways. Nucleophilic attack on the isocyanide complex [(C^Npz^C)Au(Cā‰”NC6H3Me2-2,6)]SbF6 (2) gave [(C^Npz^C)Au(ACC)]SbF6 complexes with aniline (4Ā·SbF6), adamantylamine (5), glycine ethyl ester (6), alanine methyl ester (7), valine methyl ester (8), phenylglycine methyl ester (9) and methionine methyl ester (10) substituents (ACC = acyclic carbene). The pathway via isocyanide insertion into gold-amide bonds was also investigated; e.g. the reaction of xylyl isocyanide with (C^Npz^C)AuNHPh followed by protonation with HBF4Ā·OEt2 gave the acyclic carbene complex 4Ā·BF4. To the best of our knowledge compounds 6 - 10 represent the first examples of gold(III) acyclic carbene complexes bearing amino acid functions. The compounds provide a versatile platform for the study of anti-proliferative properties of gold(III) complexes. Tests against human adenoma-type lung cancer cells identified 5, 6, 7 and 10 as particularly promising and demonstrate the synthetic flexibility of acyclic carbene complexes and the potential of that class of compounds for anticancer applications. Compared to cisplatin, amino ester-containing ACC complexes showed significantly improved selectivity for MCF-7 breast cancer cells over healthy fibroblasts

    Evolutionary dynamics of the most populated genotype on rugged fitness landscapes

    We consider an asexual population evolving on rugged fitness landscapes which are defined on the multi-dimensional genotypic space and have many local optima. We track the most populated genotype as it changes when the population jumps from a fitness peak to a better one during the process of adaptation. This is done using the dynamics of the shell model which is a simplified version of the quasispecies model for infinite populations and standard Wright-Fisher dynamics for large finite populations. We show that the population fraction of a genotype obtained within the quasispecies model and the shell model match for fit genotypes and at short times, but the dynamics of the two models are identical for questions related to the most populated genotype. We calculate exactly several properties of the jumps in infinite populations some of which were obtained numerically in previous works. We also present our preliminary simulation results for finite populations. In particular, we measure the jump distribution in time and find that it decays as tāˆ’2t^{-2} as in the quasispecies problem.Comment: Minor changes. To appear in Phys Rev

    Nuclear Bar Catalyzed Star Formation: 13^CO, C18^O and Molecular Gas Properties in the Nucleus of Maffei 2

    (Abridged) We present resolution maps of CO, its isotopologues, and HCN from in the center of Maffei 2. The J=1-0 rotational lines of 12^CO, 13^CO, C18^O and HCN, and the J=2-1 lines of 13^CO and C18^O were observed with the OVRO and BIMA arrays. The 2-1/1-0 line ratios of the isotopologues constrain the bulk of the molecular gas to originate in low excitation, subthermal gas. From LVG modeling, we infer that the central GMCs have n(H_2) ~10^2.75 cm^-3 and T_k ~ 30 K. Continuum emission at 3.4 mm, 2.7 mm and 1.4 mm was mapped to determine the distribution and amount of HII regions and dust. Column densities derived from C18^O and 1.4 mm dust continuum fluxes indicate the CO conversion factor in the center of Maffei 2 is lower than Galactic by factors of ~2-4. Gas morphology and the clear ``parallelogram'' in the Position-Velocity diagram shows that molecular gas orbits within the potential of a nuclear (~220 pc) bar. The nuclear bar is distinct from the bar that governs the large scale morphology of Maffei 2. Giant molecular clouds in the nucleus are nonspherical and have large linewidths. Dense gas and star formation are concentrated at the sites of the x_1-x_2 orbit intersections of the nuclear bar, suggesting that the starburst is dynamically triggered.Comment: 50 pages, 14 figures, accepted for publication in Ap

    Intrafamilial variable phenotype including corticobasal syndrome in a family with p.P301L mutation in the MAPT gene: first report in South America

    Frontotemporal lobar degeneration is a neuropathological disorder that causes a variety of clinical syndromes including frontotemporal dementia (FTD), progressive supranuclear palsy, and corticobasal syndrome (CBS). FTD associated with parkinsonism occurs frequently as a result of mutations in the C9orf72 gene and also in the genes coding for the protein associated with microtubule tau (MAPT) and progranulin (GRN) on chromosome 17 (FTDP-17). Herein, we report an Argentinean family, of Basque ancestry, with an extensive family history of behavioral variant of FTD. Twenty-one members over 6 generations composed the pedigree. An extensive neurologic and neurocognitive examination was performed on 2 symptomatic individuals and 3 nonsymptomatic individuals. Two different phenotypes were identified among affected members, CBS in the proband and FTD in his brother. DNA was extracted from blood for these 5 individuals and whole-exome sequencing was performed on 3 of them followed by Sanger sequencing of candidate genes on the other 2. In both affected individuals, a missense mutation (p.P301L; rs63751273) in exon 10 of the MAPT gene (chr17q21.3) was identified. Among MAPT mutations, p.P301L is the most frequently associated to different phenotypes: (1) aggressive, symmetrical, and early-onset Parkinsonism; (2) late parkinsonism associated with FTD; and (3) progressive supranuclear palsy but only exceptionally it is reported associated to CBS. This is the first report of the occurrence of the p.P301L-MAPT mutation in South America and supports the marked phenotypic heterogeneity among members of the same family as previously reported
