    Altered intrinsic functional connectivity in language-related brain regions in association with verbal memory performance in euthymic bipolar patients

    Potential abnormalities in the structure and function of the temporal lobes have been studied much less in bipolar disorder than in schizophrenia. This may not be justified because language-related symptoms, such as pressured speech and flight of ideas, and cognitive deficits in the domain of verbal memory are amongst the hallmark of bipolar disorder (BD), and contribution of temporal lobe dysfunction is therefore likely. In the current study, we examined resting-state functional connectivity (FC) between the auditory cortex (Heschl’s gyrus [HG], planum temporale [PT]) and whole brain using seed correlation analysis in n = 21 BD euthymic patients and n = 20 matched healthy controls and associated it with verbal memory performance. In comparison to controls BD patients showed decreased functional connectivity between Heschl’s gyrus and planum temporale and the left superior and middle temporal gyrus. Additionally, fronto-temporal functional connectivity with the right inferior frontal/precentral gyrus and the insula was increased in patients. Verbal episodic memory deficits in the investigated sample of BD patients and language-related symptoms might therefore be associated with a diminished FC within the auditory/temporal gyrus and a compensatory fronto-temporal pathway

    A journey around my classroom: the psychogeography of learning spaces

    During a workshop held at the UAL Learning and Teaching Day we led a session where we used a number of techniques to disrupt the traditional format of a classroom or lecture space. Lectures and classrooms are often associated with static, immovable furniture organised in neat rows, all facing towards a whiteboard or screen, affording specific pedagogies, behaviours, rules and power structures. Our aim in the workshop was to challenge participants’ preconceptions of what might happen in such spaces. Using our observations and reflections on the workshop, this article explores how participants responded to and engaged with the different structures and activities imposed

    Mutual Unbiasedness in Coarse-grained Continuous Variables

    The notion of mutual unbiasedness for coarse-grained measurements of quantum continuous variable systems is considered. It is shown that while the procedure of "standard" coarse graining breaks the mutual unbiasedness between conjugate variables, this desired feature can be theoretically established and experimentally observed in periodic coarse graining. We illustrate our results in an optics experiment implementing Fraunhofer diffraction through a periodic diffraction grating, finding excellent agreement with the derived theory.     White matter differences between healthy young ApoE4 carriers and non-carriers identified with tractography and support vector machines.

    The apolipoprotein E4 (ApoE4) is an established risk factor for Alzheimer's disease (AD). Previous work has shown that this allele is associated with functional (fMRI) changes as well structural grey matter (GM) changes in healthy young, middle-aged and older subjects. Here, we assess the diffusion characteristics and the white matter (WM) tracts of healthy young (20-38 years) ApoE4 carriers and non-carriers. No significant differences in diffusion indices were found between young carriers (ApoE4+) and non-carriers (ApoE4-). There were also no significant differences between the groups in terms of normalised GM or WM volume. A feature selection algorithm (ReliefF) was used to select the most salient voxels from the diffusion data for subsequent classification with support vector machines (SVMs). SVMs were capable of classifying ApoE4 carrier and non-carrier groups with an extremely high level of accuracy. The top 500 voxels selected by ReliefF were then used as seeds for tractography which identified a WM network that included regions of the parietal lobe, the cingulum bundle and the dorsolateral frontal lobe. There was a non-significant decrease in volume of this WM network in the ApoE4 carrier group. Our results indicate that there are subtle WM differences between healthy young ApoE4 carriers and non-carriers and that the WM network identified may be particularly vulnerable to further degeneration in ApoE4 carriers as they enter middle and old age

    Reduced hippocampal volume in healthy young ApoE4 carriers: an MRI study.

    The E4 allele of the ApoE gene has consistently been shown to be related to an increased risk of Alzheimer's disease (AD). The E4 allele is also associated with functional and structural grey matter (GM) changes in healthy young, middle-aged and older subjects. Here, we assess volumes of deep grey matter structures of 22 healthy younger ApoE4 carriers and 22 non-carriers (20-38 years). Volumes of the nucleus accumbens, amygdala, caudate nucleus, hippocampus, pallidum, putamen, thalamus and brain stem were calculated by FMRIB's Integrated Registration and Segmentation Tool (FIRST) algorithm. A significant drop in volume was found in the right hippocampus of ApoE4 carriers (ApoE4+) relative to non-carriers (ApoE4-), while there was a borderline significant decrease in the volume of the left hippocampus of ApoE4 carriers. The volumes of no other structures were found to be significantly affected by genotype. Atrophy has been found to be a sensitive marker of neurodegenerative changes, and our results show that within a healthy young population, the presence of the ApoE4+ carrier gene leads to volume reduction in a structure that is vitally important for memory formation. Our results suggest that the hippocampus may be particularly vulnerable to further degeneration in ApoE4 carriers as they enter middle and old age. Although volume reductions were noted bilaterally in the hippocampus, atrophy was more pronounced in the right hippocampus. This finding relates to previous work which has noted a compensatory increase in right hemisphere activity in ApoE4 carriers in response to preclinical declines in memory function. Possession of the ApoE4 allele may lead to greater predilection for right hemisphere atrophy even in healthy young subjects in their twenties

    Children’s understanding of first and third person perspectives in complement clauses and false belief tasks

    De Villiers (2007) and others have claimed that children come to understand false beliefs as they acquire linguistic constructions for representing a proposition and the speaker’s epistemic attitude toward that proposition. In the current study, English-speaking children (N=64) of 3 and 4 years of age were asked to interpret propositional attitude constructions with a first-person or a third-person subject of the propositional attitude (e.g., I think the sticker is in the red box or The cow thinks the sticker is in the red box, respectively). They were also assessed for an understanding of their own and others’ false beliefs. We found that 4-year-olds showed a better understanding of both third-person propositional attitude constructions and false belief than their younger peers. No significant developmental differences were found for first-person propositional attitude constructions. The older children also showed a better understanding of their own than of others’ false beliefs. In addition, regression analyses suggest that the older children’s comprehension of their own false belief was mainly related to their understanding of third-person propositional attitude constructions. These results indicate that we need to take a closer look at the propositional attitude constructions that are supposed to support children’s false-belief reasoning. Children may come to understand their own and others’ beliefs in different ways, and this may affect both their use and understanding of propositional attitude constructions and their performance in various types of false-belief tasks

    Global Stochastic Optimization of Stellarator Coil Configurations

    In the construction of a stellarator, the manufacturing and assembling of the coil system is a dominant cost. These coils need to satisfy strict engineering tolerances, and if those are not met the project could be canceled as in the case of the National Compact Stellarator Experiment (NCSX) project [25]. Therefore, our goal is to find coil configurations that increase construction tolerances without compromising the performance of the magnetic field. In this paper, we develop a gradient-based stochastic optimization model which seeks robust stellarator coil configurations in high dimensions. In particular, we design a two-step method: first, we perform an approximate global search by a sample efficient trust-region Bayesian optimization; second, we refine the minima found in step one with a stochastic local optimizer. To this end, we introduce two stochastic local optimizers: BFGS applied to the Sample Average Approximation and Adam, equipped with a control variate for variance reduction. Numerical experiments performed on a W7-X-like coil configuration demonstrate that our global optimization approach finds a variety of promising local solutions at less than 0.1% of the cost of previous work, which considered solely local stochastic optimization.Comment: 19 pages, 4 figures, 3 table
