    Integrated process production and extraction of the fibrinolytic protease from Bacillus sp. UFPEDA 485

    Fibrinolytic proteases are enzymes that degrade fibrin; these enzymes are a promising alternative for thrombolytic therapy, and microorganisms produce them. The aim of this study was to evaluate the optimum conditions for the integrated production and purification of fibrinolytic protease from Bacillus sp. UFPEDA 485. Extractive fermentation was carried out in a culture medium containing soybean flour and by adding polyethylene glycol (PEG) and Na2SO4 according to a 23 experimental design. In all assays, the enzyme preferentially partitioned to the bottom phase (K < 1), with an optimum activity of 835 U ml−1 in the bottom phase (salt-rich phase). The best conditions for extractive fermentation were obtained with 18 % PEG 8000 and 13 % Na2SO4. Characterization showed that it is a metalloprotease, as a strong inhibition—residual activity of 3.13 %—occurred in the presence of ethylenediaminetetraacetic acid. It was also observed that enzymatic activity was stimulated in the presence of ions: CaCl2 (440 %), MgCl2 (440 %), FeSO4 (268 %), and KCl (268 %). The obtained results indicate that the use of a low-cost substrate and the integration of fermentation with an aqueous two-phase system extraction may be an interesting alternative for the production of fibrinolytic protease.The authors thank CAPES (National Council for the Improvement of Higher Education) for the scholarship and CNPq (National Counsel of Technological and Scientific Development) and RENORBIO for the financial support

    Epidemiologic study of odontogenic and non-odontogenic cysts in children and adolescents of a Brazilian population

    The objective of this study was to describe the frequency of cystic lesions in a Brazilian population of patients histopathologically diagnosed in the first and second decade of life. Retrospective descriptive cross-sectional study was performed. Biopsy records were obtained from the archives of a Brazilian referral center between 1980 and 2016. A total of 2.114 biopsy records of pediatric patients were analyzed with oral and maxillofacial lesions. Data such as gender, age, anatomical location, and histopathological diagnosis were collected and categorized. Among all oral and maxillofacial lesions (n=2.114), were diagnosed 294 cases of odontogenic cysts (13.9%) and 16 cases of non-odontogenic cysts (0.8%). The most frequent lesions in each group were, respectively: radicular cyst (n=145) and epidermoid cyst (n=4). These lesions were most common in female (n=158), with a mean age of 14 years. For intraosseous lesions, the mandible (n=148) was the most affected anatomic site; moreover, the floor of the mouth (n=6) was most affected by cysts in soft tissues. Odontogenic cysts were relatively common in population studied, but non-odontogenic cysts were rare in these patients

    The purpose of this article is to explain the activities carried out in the Extension Project: Efficient use of energy at Universidade Federal Rural do Semiárido in its ninth year.  The research also reflects on how the academic experience in this project contributes to the training of an engineer, not only through knowledge about the areas of electrical engineering which are addressed during the activities, but also in the development of the participating members, in the acquired experience and resourcefulness in managing activities and team relationships, as well as the social concern in disseminating knowledge about energy efficiency.El objetivo de este artículo es explicar las actividades que se han llevado a cabo hasta el momento en el Proyecto de Extensión: Uso Eficiente de la Energía en Universidade Federal Rural do Semiárido en su IX año y a partir de ahí, reflexionar sobre cómo la experiencia académica en este proyecto contribuye a la formación de un ingeniero, no solo a través del conocimiento sobre las áreas de la ingeniería eléctrica, las cuales se abordan durante las actividades, sino también en el desarrollo de los miembros participantes, en la experiencia adquirida y la habilidad en la gestión de actividades y relaciones de equipo, así como en la preocupación social en la difusión de conocimientos sobre eficiencia energética.O objetivo do presente artigo é explanar as atividades que foram realizadas até o momento no Projeto de Extensão: Uso Eficiente de Energia na Universidade Federal Rural do Semiárido no seu IX ano e a partir disso, refletir sobre como a vivência acadêmica neste projeto contribui para a formação de um engenheiro, não somente através do conhecimento à respeito das áreas de engenharia elétrica que são abordadas no decorrer das atividades, mas também no desenvolvimento dos membros participantes, na experiência adquirida e desenvoltura em gerir atividades e relacionamento em equipe, bem como a preocupação social em disseminar os conhecimentos sobre eficiência energética

    EXPERIENCE OF THE MATERNAL WEANING PROCESS AMONG PRIMIPAROUS WOMEN: QUALITATIVE STUDY

    Objective: To understand the experience of the weaning process for primiparous mothers in a municipality in the interior of Ceará, Brazil.Method: Using semi-structured interviews, a descriptive, qualitative study was conducted with eight women. Data collection from August 2022 to March 2023 in Primary Health Care, using Bardin's content analysis.Results: three thematic categories emerged: reasons for the mother's decision to wean, strategies employed by the mother to wean, and the consequences of weaning for the mother and child. Maternal perceptions of the motivation for weaning were related to behavioral aspects of the child and maternal needs. Various strategies were classified as gradual or abrupt weaning, which generated changes in the children's behavior and contradictory feelings in the mothers.Final considerations: weaning is experienced unpleasantly, generating suffering. It reveals the lack of professional assistance and the need to expand studies

    Analysis of the acute neuroinflammatory response and endogenous markers in the amygdala of animals submitted to status epilepticus by intrahippocampal pilocarpine application

    Introduction: Experimental evidence and clinical evidence indicate that the inflammatory process is a crucial mechanism in the pathophysiology of epileptic seizures and temporal lobe epilepsy. The amygdala when involved in an atypical processing is associated with multiple moods such as depression and anxiety disorder and psychiatric disorders such as schizophrenia. Objective: This study investigated the acute inflammatory process and modulation of the endogenous proteins’ galectins and AnxA1 in the amygdala of animals submitted to an experimental model of temporal lobe epilepsy. Methods: The experimental procedures were approved by the Ethics Committee on the Use of Animals at UNIFESP (CEUA nº2958050814). The experiments performed in this study used data and materials that were obtained from the project “Neuroprotective and anti-inflammatory role of the mimetic peptide ac2-26 of the annexin a1 protein in intrahippocampal pilocarpine-induced status epilepticus” conducted by the advisor. The experimental model used male Wistar rats that were divided into 3 experimental groups (NAIVE; SHAM, Status Epilepticus or SE - n = 5 animals/group). Once acclimated, the animals in the SHAM and SE groups underwent stereotaxic surgery for implantation of the intracerebral cannula in the right hippocampus. The SHAM animals received sterile saline in all procedures and the NAIVE group only manipulated. The animals were monitored throughout the period and after 24 hours of experiment all animals were euthanized by overdose of thiopental to remove the brain and performed histological and immunohistochemical analysis. Results and Conclusion: Initial results demonstrate that SE and the acute inflammatory process cause damage to the amygdala, and there is also modulation of inflammatory markers in this structure. However, further studies are needed to better understand the mechanism of action in neuroinflammation in status epilepticus

    HOSPITALIDADE SUSTENTÁVEL NAS PRÁTICAS DE BUGUEIROS NO POLO COSTA DAS DUNAS, RN

    O presente estudo pretende apreender a percepção de hospitalidade sustentável no oficio de bugueiros no Polo Costa das Dunas/RN. Então, realizou-se estudo de caso cujas percepções foram obtidas com 30 bugueiros devidamente certificados. Os atributos da hospitalidade foram analisados em três categorias, abrangendo o domínio do idioma ou comunicação; personalização e qualidade da visitação turística do passeio. Como resultado constatou-se que os bugueiros percebem o ecossistema dunar como local de trabalho, priorizando a educação ambiental durante o passeio. A hospitalidade é ineficiente em relação ao domínio de idiomas, mas garante a segurança de público idoso e infantil. Concluiu-se que o bugueiro exerce importante papel na sensibilização/conscientização ambiental e a hospitalidade demonstrada proporciona uma experiência única para o turismo sustentável
