    "December 2013.""A Thesis presented to the Faculty of the Graduate School University of Missouri In Partial Fulfillment Of the Requirements for the Degree Master of Science."Thesis advisor: Dr. Anthony Lupo.After the devastating tornadoes in Joplin, MO and in the Deep South in 2011, it seemed appropriate to look at the impact that broadcast meteorologists (and their TV coverage) have on their viewers during severe weather events. Broadcast meteorologists play a vital role in the severe weather warning process and in persuading the public to take the appropriate actions during severe weather. This research was done by developing a survey that addressed the following questions: 1) Is the media doing everything they can persuade viewers to take shelter and protect themselves and their property?; 2) What do you do when a tornado warning is issued?; 3) Is there anything broadcast meteorologists can do or say that will make you take immediate action during severe weather? The survey was disseminated through television markets in Missouri. The goal of this research was to find new, improved and different ways of "connecting" with viewing during severe weather coverage. After looking at the results, we want to see if there are specific words, images or anything else a broadcaster can do that will trigger a response by viewers to take cover. It is my hope the results and analyses from this survey will provide broadcast meteorologists with new and improved techniques to connect with the public and to assist them in making an informed decision during severe weather events.Includes bibliographical references (page 77)

    Analysis of Students’ Behavior Watching iMooX Courses with Interactive Elements

    Digital learning technologies are becoming increasingly important for our modern educational system. In addition to teaching methods that incorporate interactivity, these approaches benefit students’ overall learning experience and success by enhancing their attention and fostering a positive attitude towards the learning content being presented. Interactivity comes in various forms, and while a combination of distinct activities is beneficial, some are more effective at engaging students. Using digital technologies in an educational environment opens up new possibilities for students, teachers, and researchers. It provides new insights into learning behavior and enables the collection of interaction information. This data could, for example, show how often a video was paused or at what point students lost interest and left, but gaining such knowledge requires further processing. The use of visualizations that depict behavior, such as the change of attention over time, can be an effective way to present extracted information. Therefore, our research focuses on developing an application that enables us to generate various visualizations from the collected data. A single command-line input will be sufficient to create them. Furthermore, a video course was created from which we collected behavioral data. Our results aim to showcase the benefits of interactivity, and that the created figures can be used for data evaluation verifies the versatility of the generated visualizations

    Molecular and immunological characterization of profilin from mugwort pollen

    In late summer in Europe, pollen of mugwort is one of the major sources of atopic allergens. No information about the complete molecular structure of any mugwort allergen has been published so far. Here we report the isolation and characterization of mugwort pollen cDNA clones coding for two isoforms of the panallergen profilin. Thirtysix percent of the mugwort allergic patients tested displayed IgE antibodies against natural and recombinant profilin, and no significant differences were observed in the IgEbinding properties of the isoforms. One profilin isoform was purified to homogeneity and detailed structural analysis indicated that the protein exists in solution as dimers and tetramers stabilized by sulfydryl and/or ionic interactions. Profilin monomers were detectable only after exposure of multimers to harsh denaturing conditions. Dimers and tetramers did not significantly differ in their ability to bind serum IgE from mugwort pollenallergic patients. However, oligomeric forms might have a higher allergenic potential than monomers because larger molecules would have additional epitopes for IgEmediated histamine release. Profilin isolated from mugwort pollen also formed multimers. Thus, oligomerization is not an artifact resulting from the recombinant production of the allergen. Inhibition experiments showed extensive IgE crossreactivity of recombinant mugwort profilin and profilin from various pollen and food extracts

    The function of the left angular gyrus in mental arithmetic: Evidence from the associative confusion effect

    While the left angular gyrus (lAG) has been repeatedly implicated in mental arithmetic, its precise functional role has not been established. On the one hand, it has been speculated that the lAG is involved in task-specific processes. On the other hand, the observation of relative deactivation during arithmetic has led to the contention that differential lAG activation reflects task-unrelated difficulty effects associated with the default mode network (DMN). Using functional magnetic resonance imaging, we investigated the neural correlates of the associative confusion effect that allowed us to dissociate effects of task difficulty and task-related arithmetic processes on lAG activation. The associative confusion effect is characterized by poorer performance while verifying addition and multiplication equations whose solutions are associated with the other operation (confusion equations: e.g., 9 × 6 = 15 ) compared with solutions unrelated to both operations (non-confusion equations: e.g., 9 × 6 = 52 ). Comparing these two conditions revealed higher activation of the anterior lAG (areas PGa, PFm, and PF) and the left dorsolateral prefrontal cortex for the confusion problems. This effect displayed only slight anatomical overlap with the well-established reverse problem-size effect (small minus large problems) and task-related deactivation in the parietal cortex. The finding of greater lAG activity (less deactivation) in the more difficult task condition is inconsistent with the hypothesis that lAG activation during mental arithmetic reflects task difficulty related modulations of the DMN. Instead, the present findings provide further support for the symbol-referent mapping hypothesis, suggesting that the lAG mediates the automatic mapping of arithmetic problems onto solutions stored in memory. © 2011 Wiley Periodicals, Inc

    Modeling the growth of stylolites in sedimentary rocks

    [1] Stylolites are ubiquitous pressure solution seams found in sedimentary rocks. Their morphology is shown to follow two self-affine regimes. Analyzing the scaling properties of their height over their average direction shows that (1) at small scale, they are self-affine surfaces with a Hurst exponent around 1, and (2) at large scale, they follow another self-affine scaling with Hurst exponent around 0.5. In the present paper, we show theoretically the influence of the main principal stress and the local geometry of the stylolitic interface on the dissolution reaction rate. We compute how it is affected by the deviation between the principal stress axis and the local interface between the rock and the soft material in the stylolite. The free energy entering in the dissolution reaction kinetics is expressed from the surface energy term and via integration from the stress perturbations due to these local misalignments. The resulting model shows the interface evolution at different stress conditions. In the stylolitic case, i.e., when the main principal stress is normal to the interface, two different stabilizing terms dominate at small and large scales which are linked respectively to the surface energy and to the elastic interactions. Integrating the presence of small-scale heterogeneities related to the rock properties of the grains in the model leads to the formulation of a Langevin equation predicting the dynamic evolution of the surface. This equation leads to saturated surfaces obeying the two observed scaling laws. Analytical and numerical analysis of this surface evolution model shows that the crossover length separating both scaling regimes depends directly on the applied far-field stress magnitude. This method gives the basis for the development of a paleostress magnitude marker. We apply the computation of this marker, i.e., the morphological analysis, on a stylolite found in the Dogger limestone layer located in the neighborhood of the ANDRA Underground Research Laboratory at Bure (eastern France). The results are consistent with the two scaling regimes expected, and the practical determination of the major principal paleostress, from the estimation of a crossover length, is illustrated on this example

    Association between polymorphisms in NOS3 and KCNH2 and social memory

    Social memory, including the ability to recognize faces and voices, is essential for social relationships. It has a large heritable component, but the knowledge about the contributing genes is sparse. The genetic variation underlying inter-individual differences in social memory was investigated in an exploratory sample (n=55), genotyped with a chip comprising approximately 200.000 single nucleotide polymorphisms (SNPs), and in a validation sample (n=582), where 30 SNPs were targeted. In the exploratory study face identity recognition was measured. The validation study also measured vocal sound recognition, as well as recognition of faces and vocal sounds combined (multimodal condition). In the exploratory study, the 30 SNPs that were associated with face recognition at puncorrected<0.001 and located in genes, were chosen for further study. In the validation study two of the SNPs showed significant associations with recognition of faces, vocal sounds, and multimodal stimuli: rs1800779 in the gene encoding nitric oxide synthase 3 (NOS3) and rs3807370 in the gene encoding the voltage-gated channel, subfamily H, member 2 (KCNH2), in strong linkage disequilibrium with each other. The uncommon alleles were associated with superior performance, and the effects were present for men only (p<0.0002). The exploratory study also showed a weaker but significant association with (non-emotional) word recognition, an effect that was independent of the effect on face recognition. This study demonstrates evidence for an association between NOS3 and KCNH2 SNPs and social memory

    Glioblastoma and the search for non-hypothesis driven combination therapeutics in academia

    Glioblastoma (GBM) remains a cancer of high unmet clinical need. Current standard of care for GBM, consisting of maximal surgical resection, followed by ionisation radiation (IR) plus concomitant and adjuvant temozolomide (TMZ), provides less than 15-month survival benefit. Efforts by conventional drug discovery to improve overall survival have failed to overcome challenges presented by inherent tumor heterogeneity, therapeutic resistance attributed to GBM stem cells, and tumor niches supporting self-renewal. In this review we describe the steps academic researchers are taking to address these limitations in high throughput screening programs to identify novel GBM combinatorial targets. We detail how they are implementing more physiologically relevant phenotypic assays which better recapitulate key areas of disease biology coupled with more focussed libraries of small compounds, such as drug repurposing, target discovery, pharmacologically active and novel, more comprehensive anti-cancer target-annotated compound libraries. Herein, we discuss the rationale for current GBM combination trials and the need for more systematic and transparent strategies for identification, validation and prioritisation of combinations that lead to clinical trials. Finally, we make specific recommendations to the preclinical, small compound screening paradigm that could increase the likelihood of identifying tractable, combinatorial, small molecule inhibitors and better drug targets specific to GBM.Peer reviewe

    Predictors for long-term survival free from whole brain radiation therapy in patients treated with radiosurgery for limited brain metastases

    PURPOSE: To identify predictors for prolonged survival free from salvage whole brain radiation therapy (WBRT) in patients with brain metastases treated with stereotactic radiosurgery (SRS) as their initial radiotherapy approach. MATERIALS AND METHODS: Patients with brain metastases treated with SRS from 2001 to 2013 at our institution were identified. SRS without WBRT was typically offered to patients with 1-4 brain metastases, Karnofsky performance status \u3e /=70, and life expectancy \u3e /=3 months. Three hundred and eight patients met inclusion criteria for analysis. Medical records were reviewed for patient, disease, and treatment information. Two comparison groups were identified: those with \u3e /=1-year WBRT-free survival (N = 104), and those who died or required salvage WBRT within 3 months of SRS (N = 56). Differences between these groups were assessed by univariate and multivariate analyses. RESULTS: Median survival for all patients was 11 months. Among patients with \u3e /=1-year WBRT-free survival, median survival was 33 months (12-107 months) with only 21% requiring salvage WBRT. Factors significantly associated with prolonged WBRT-free survival on univariate analysis (p \u3c 0.05) included younger age, asymptomatic presentation, RTOG RPA class I, fewer brain metastases, surgical resection, breast primary, new or controlled primary, absence of extracranial metastatic disease, and oligometastatic disease burden ( \u3c /=5 metastatic lesions). After controlling for covariates, asymptomatic presentation, breast primary, single brain metastasis, absence of extracranial metastases, and oligometastatic disease burden remained independent predictors for favorable WBRT-free survival. CONCLUSION: A subset of patients with brain metastases can achieve long-term survival after upfront SRS without the need for salvage WBRT. Predictors identified in this study can help select patients that might benefit most from a treatment strategy of SRS alone

    The Palladium-Catalyzed Aerobic Kinetic Resolution of Secondary Alcohols: Reaction Development, Scope, and Applications

    The first palladium-catalyzed enantioselective oxidation of secondary alcohols has been developed, utilizing the readily available diamine (-)-sparteine as a chiral ligand and molecular oxygen as the stoichiometric oxidant. Mechanistic insights regarding the role of the base and hydrogen-bond donors have resulted in several improvements to the original system. Namely, addition of cesium carbonate and tert-butyl alcohol greatly enhances reaction rates, promoting rapid resolutions. The use of chloroform as solvent allows the use of ambient air as the terminal oxidant at 23 °C, resulting in enhanced catalyst selectivity. These improved reaction conditions have permitted the successful kinetic resolution of benzylic, allylic, and cyclopropyl secondary alcohols to high enantiomeric excess with good-to-excellent selectivity factors. This catalyst system has also been applied to the desymmetrization of meso-diols, providing high yields of enantioenriched hydroxyketones
